1. Development of natural seasoning from ascidian
Traditional food culture was particularly specialized by each nation originated from food choices that are based from food experience of their ancestors on safety needs. A taste is thought to be one of the important factor for its choice. Monosodium glutamate, simply named as MSG, is originated from Japan and it has been widely used into Korean traditional food. However, MSG induces such phenomenon as vomiting, headache, and dizziness called as "Syndrome of Chinese Restaurant". The natural plants and animal protein hydrolysates has been studied and used to improve their tastes. The proteins from seaweeds and marine invertebrate have been tried to be a potential resources for seasoning and taste ingredients. However, extraction and application of protein from seaweeds and marine invertebrates was little reported. The objectives of this thesis were to investigate the extraction of protein from ascidian(Halocynthia roretzi ), to develop a seasoning ingredient with their hydrolysates and to evaluate food quality of the seasonings which are made with their hydrolysates.
The amounts of crude protein for muscle and tunic of ascidian were 6.9% and 5.5%, respectively. The amount of free amino acids of muscle was 331 mg/100 g and the order for its high amount was taurine, urea, glycine, and glutamic acid, which were 92.9% of total amount of free amino acids. The main amino acids and N-containing compound of cooked extracts from ascidian were hydroxyproline, taurine and urea, which were 71.4% of total amount of free amino acids. In mineral analyses of muscle and tunic, sodium was the highest amount, showing that their sodium contents were 111 and 460 mg/kg, respectively. The concentrated solution of ascidian drip and extracts of tunic showed much more sodium concentration of 1,944 mg/kg, and K, Mg and Ca in tunic were also higher than those of tunic by 2.8-4.8, 3.5 - 15.2, and 3.0-16.1 times, respectively.
As the ratio of protease to carbohydrate was increased, the extracted yields of protein and carbohydrate were increased. The yield of Protamex hydrolysates were increased with increasing pH in the range of pH 5-7. However, there were no significant differences in the amounts of ketose and hexose, and protein hydrolysates in the range of 30-50℃. As the ratio of substrate to enzyme was increased, low molecular compounds of the hydrolysates were increased. The amount of soluble protein (Y) in the experimental range was shown in the following formula: Y = 47.737 * the ratio of substrate to enzyme + 25.823 * pH -19.156 * temp. + 350.323. However, there were no significant differences of carbohydrate-degraded products in the experimental range for the related factors.
In the experimental design for making ascidian seasoning, the mixed ascidian seasonings of 90% of enzyme hydrolysates and cooked solution of ascidian and 10% of mushroom mycelium; 95% of enzyme hydrolysate and cooked solution of ascidian, 2.5% beer yeast and 2.5% mushroom mycelium had 6.2-6.8 of high scores in the sensory evaluation, showing relatively high quality. The optimum mixing ratio for making soy sauce with ascidian was the ranges of 0-1% hydrolysate, 1-5% cooked and concentrated solution, 2-5% beer yeast, 2-5% potato mycelium in the fixed amount of 84% soy sauce. In the analyses of DPPH free radical scavenging, Fe-chelating and ACE inhibition activities for bioactive functions, enzyme hydrolysates and cooked and concentrated solution of ascidian meat had no high activities.
In the results of sensory evaluation of consumers for ascidian seasoning, the first feeling on the product was a new product, one of the advantages, speciality and freshness. However, panels who had no or neutral preference for ascidian a little had a negative perception for the ascidian seasoning. Therefore, the reasons on preference and no preference characteristics are thought to be individual preference. In the results of sensory evaluation for the ascidian seasoning, total and main preferences had the average score of 5-6 points, showing a positive level for consumers, when the product had not commercially sold yet for general consumers. The ascidian product was evaluated and summarized to have dense color, salty taste and strong ascidian aroma but a little less sweetness and sourness.
In the application of the ascidian seasoning to such other foods as raw fish, fried products, "jokbal"(glazed pigs' feet), and fish cake soup, there were no significant differences for their preferences. In the case of raw fish(sasimi), there was a big difference in deviation of panels. The three kinds of the applied foods except raw fish had been evaluated to have better preferences after testing the product than before testing it. However, the ascidian seasoning had the adverse effect for raw fish. In the survey for using the ascidian seasoning after sensory evaluation, the ratio of the positive answers for "I would like to buy the ascidian seasoning." did not significantly increase after sensory evaluation than before sensory evaluation but the ratio of the negative answers for it was significantly decreased with a big difference. Therefore, the preference of the ascidian seasoning could be decided to be better after eating than expected.
In the results of the sensory evaluation of raw fish and fried products for the ascidian seasoning, the existed soy sauce with wasabi showed better preference than the ascidian seasoning, but in the sensory evaluation for jokbal and fish cake soup, the ascidian seasoning had a little better or no preference than the existed sauce. Therefore, the ascidian seasoning is thought to be valuable for its commercial application in stead of the existed sauce.
2. Development of cosmetic from ascidian
Glycosaminoglycans are a group of structurally related polysaccharides found as the carbohydrate moieties of proteoglycans and sometimes as free polysaccharides. Their functions are diverse. Glycosaminoglycans may all be components of connective tissue or cell surface carbohydrates involved in the cell’s interaction with and response to its surroundings. The production of functional cosmetic containing glycosaminoglycans has increased due to their biological functions against anti-aging, reconstruction of collagen in skin.
The wastes of ascidian tunic were crushed and extracted pigments with acetone for one night. The acetone layer were filtered and concentrated for colorful cosmetics. The tissue was extracted with 10 volumes of distilled water by pressure cooker at 121°C for 3 hrs. After concentration of the supernatant under the vacuum pressure until reach to Brix 10, the concentrated glycosaminoglycans were applied to a DEAE-Sepharose column equilibrated with 50 mM sodium phosphate buffer(pH 6.0) and washed with 50 mL of the same buffer. The fractions collected and assayed by hexoses and pentose using the phenol-sulfur acid, hexosamines and acetylhexoamines using the metachromasia and uronic acid using the carbazole reaction. This thermal extraction method was found crude sulfated polysaccharides for industrial purposes.
In the analyses of DPPH free radical scavenging, ferricyanide reduction activity and linoleic acid autoxidation activities for functionality of carotenoids containing cosmetics. DPPH radical activity showed 47.2% at 100 mg/mL of concentration, according to increasing the sample concentration with increased radical scavenging activity. Ferricyanide reduction activity of natural pigments was higher than that of ten times of ascorbic acid at concentration 10 mg/mL. Natural pigments showed low activity autoxidation in linoleic acid system compare to tocopherol which is standard of antioxidant material. Cytotoxicity by the natural pigments treatment can show the non toxicity effect depend on the different concentration which were 10 to 100 μg/mL. So the concentration was fixed to 10 μg/mL for the inflammatory activities. To clarify the anti-inflammatory activities of natural pigments, we investigated the production of pro-inflammatory mediators effect on NO and COX2 activation of the natural pigments in the stimulated RAW 264.7 cells by RT-PCR and Western blot analysis. Non-activated cells(untreated) did not express any detectable level of NO, whereas the treatment of lipopolysaccharides(LPS) induced high levels of NO transcription. However, all of the concentration of sample did not inhibit the COX-2 expression induced by LPS.
Proximate composition of the extract showed high protein and carbohydrate contents, with the mucopolysaccharides accounting for more than 50%. The GAG extracts were also high in hexose, uronic acid and hexosamine contents. Sugar components were mainly composed of galactose, glucose and mannose along with other neutral sugars in lesser quantities. Amino sugar analysis revealed higher glucosamine content compared to galactosamine. Active fractions were treated with enzymes and analyzed by agarose gel electrophoresis, exhibiting comparable migration with dermatan sulfate although the broadened bands may suggest the presence of a unique structure. The GAGs from ascidian tunics were mostly composed of repeating mono-sulfated disaccharide bearing sulfate groups at positions C-4 or C-6 of N-acetylgalactosamine.
The functional activities of extracts were investigated using cell lines. The extracted polysaccharides and its fraction from DEAE-Sepharose fast flow 6B chromatography did not show the toxicity on RAW 264.7 cell line in the range of 10-200 μg/mL, but increased the cell growth rate. The extracted polysaccharides and its fraction also increased the formation of NO, interleukin(IL)-6 and tumor necrosis factor(TNF)-α on RAW 264.7 cell. The concentration at 0.5, 5, 25 and 50 mg/mL of glycosaminoglycans were showed collagenase inhibition activity. Compare to retinyl acetate with glycosaminoglycas from ascidian, activity of the retinyl acetate at 10 mg/mL expressed 42.2% of inhibition acticity, but glycosaminoglycans from ascidian showed higher activity than that of low concentration at 0.5 mg/mL. The examination of cell growth was find significant differences between glycosaminoglycans and collagen at 50 and 100 μg/mL for fibroblast and CCD 986sk cell lines. Glycosaminoglycans from ascidian were found significant activity in collagen synthesis using these two cell lines.
In the results of the sensory evaluation of skin, lotion and essence, glycosaminoglycans cosmetic showed antioxidant, wrinkle free activities. The glycosaminoglycans showed high viscous activity, it will showing moisturizing effect. Therefore, the ascidian glycosaminoglycans cosmetics are thought to be valuable for its commercial application in stead of the existed raw materials.
