○ 연구결과
1. 동충하초의 invitro항아토피 활성 및 세포분자 표적 연구
가. invitro항아토피 (Th1/Th2)시험
- CBBF와 CBEE에 의한 IFN-γ와 IL-4분비 억제 효능을 평가한 결과, IFN-γ의 경우 서로 다른 stimuli에서도 모두 억제하였으며 IL-4의 경우 CBBF는 ConA 조건에서 그리고 CBEE는 LPS 조건에서 IL-4분비를 억제하였다.CBBF와 CBEE에 의한 염증매개 물질인 NO 분비 억제 효능을 평가한 결과, CBBF는 세포 생존률에 비해 효과적으로 NO를 억제하였다.<
○ 연구결과
1. 동충하초의 invitro항아토피 활성 및 세포분자 표적 연구
가. invitro항아토피 (Th1/Th2)시험
- CBBF와 CBEE에 의한 IFN-γ와 IL-4분비 억제 효능을 평가한 결과, IFN-γ의 경우 서로 다른 stimuli에서도 모두 억제하였으며 IL-4의 경우 CBBF는 ConA 조건에서 그리고 CBEE는 LPS 조건에서 IL-4분비를 억제하였다.CBBF와 CBEE에 의한 염증매개 물질인 NO 분비 억제 효능을 평가한 결과, CBBF는 세포 생존률에 비해 효과적으로 NO를 억제하였다.
CBBF와 CBEE에 의한 세포-세포간의 응집능을 확인한 결과, CBBF는 CD29에 의한 세포간의 응집을 억제하였으며, CBEE는 CD43에 의한 세포간의 응집을 2시간에서 억제하였다.
CBBF와 CBEE에 의한 MAPK 신호전달 단백질의 인산화 수준을 확인한 결과, CBBF는 ERK의 인산화 수준을 15분에서 억제하였으며 CBEE는 p38의 인산화 수준을 5분에서 억제하였다.CBBF와 CBEE에 의한 다양한 전사인자의 프로모터 활성을 확인한 결과, CBBF는 AP-1, NF-κB, CREB의 프로모터 활성을 모두 농도 의존적으로 억제하였으며 CBEE의 경우는 AP-1과 CREB을 효과적으로 억제하였다.
나. invitro항아토피 (APC/mastcells)시험
- 노랑다발 동충하초 추출물인 CBEE와 CBBF 모두 IL-12의 mRNA 레벨을 억제하였다. 히스타민 분비에 관여하는 TNF-α, PGE2, NO의 분비를 확인한 결과, CBBF는 TNF-α를 모든 농도에서 약하게 억제하였으며, NO는 농도 의존적으로 억제하였다.반면에 PGE2는 농도 의존적으로 증가시켰다.CBEE와 CBBF는 Fibronectin에 의한 점착능에는 크게 영향을 주지 않았다.
CBEE의 경우, p38억제제가 IL-12의 발현을 억제시키는 것으로 보아 p38을 통해 IL-12를 조절하는 것으로 생각되어진다.CBBF의 경우 JAK2의 인산화를 조절함으로써 STAT-1에 의한 IFN-γ의 생성을 조절하는 것으로 보여진다.CBEE의 경우 p65와 c-jun의 핵내 이동을 억제하였으며, CBBF는 STAT-1과 c-fos의 핵 내 이동을 억제하는 것으로 나타났다.
다. Invitro항 아토피 기전 (분자표적단백질)연구
- SNP에 의한 NO생성을 조절하지 않는 것을 확인함으로써 TLR4매개의 NO 생성을 조절한다는 것을 확인하였다.하지만, TLR4의 발현을 직접적으로 조절하지 않는 것으로 나타났다.
Adaptormolecule인 MyD88과 TRIF에 의한 NF-kappaB luciferase프로모터 활성을 확인한 결과, CBBF의 25mg/ml농도가 TRIF에 의한 NF-kappaB luciferase프로모터 활성을 약간 억제한 것을 제외하고는 조절 효과가 없었다.이러한 억제 정도도 그 효과가 미미하여 효과가 없다고 판단하였다.따라서, 동충하초 추출물은 adaptormolecule인 MyD88과 TRIF를 타겟으로 하지 않는다는 것을 확인하였다.염증반응 신호전달 기전 중, 상위 신호전달 단백질로 알려진 Src과 Syk은 인산화에 의해 kinaseactivity를 갖는 것으로 알려져 있다.따라서, 동충하초 추출물에 의한 인산화 수준의 조절 정도를 조사한 결과, CBEE의 경우 Src을, CBBF의 경우 Src과 Syk의 kinaseactivity를 억제한다는 것을 확인하였다.
2. 동충하초 유효분획 및 성분의 화학적, 생화학적 및 제약학적 특성연구
가. 원료 특성시험 및 추출공정 개선 연구
- 노랑다발동충하초를 HPLC로 분석하여 포함됨 당(sugar)의 pattern을 비교하였을 때, ethylacetate분획물 및 buthanol분획물의 peak는 retentiontime이 느린, non-polar한 peak들이 나타나는 것을 알 수 있었다.노랑다발동충하초은 일반적으로 물(water)로 추출하여 사용하므로, 4, 100도에서의 추출물 및 waterfraction과의 HPLC pattern을 비교하였다.그 결과, 커다란 차이는 보이지 않았으며, waterfraction에서는 0-10분까지의 polar성분이 보이지를 않았다.노랑다발 동충하초의 totallipid를 GasChromatography를 이용하여 지방산 분석을 하였다. 그 결과, 노랑다발 동충하초는 특이적으로 80%의 높은 불포화 지방산(unsaturatedfattyacid)을 함유하고 있다.노랑달동충하초의 aqueouslayer를 각각, hexane, Ethylacetate, butanol, 및 water 로 분획을 하여, hexane, Ethylacetate, butanol, 및 waterfraction을 얻었다.노랑다발동충하초의 수율(yield)을 계산하여 보면, 360g의 노랑다발 동충하초를 사용하여 ethanol추출을 하여 66.1g(dir19%의 수율)을 얻었다. 66.1g의 ethanol추출물에서, waterfraction (63.4%), hexanefraction (22%), butanol(7.3%), and ethylacetate fraction (7.3%)을 얻었다.노랑다발동충하초의 BuOH 분획에 대한 안전성 평가의 일환으로 식품의약품안전청 독성시험기준에 근거하여 마우스를 이용한 급성 경구독성시험을 실시하였다.
그 결과, 독성학적 이상변화 및 치사한 개체가 관찰되지 않았으므로 10000mg/kg의 용량에서도 독성 발현이 없는 안전한 물질임을 확인하였다.동충하초의 부탄올 분획의 가용성 성분은 불용성 분획보다 낮은 용해도를 보였지만 tween80에서는 가장 높은 용해도를 보임. 불용성 분획은 대체로 다양한 매질에서 보다 우수한 용해성을 보임. 이는 시험에 사용된 매질의 성질이 유용성에 가까워 불용성 성분의 용해에 적합했기 때문으로 생각됨.
나. 기능성 성분분리 및 분석법 정립 (구조분석)
- 노랑다발 동충하초 3, 000g을 100% Ethanol로 추출하여 533.7g(17.8% yield)을 얻었고 냉각시켜 얻은 흰 침전물은 39.16g(1.31% yield), 분홍색 결정물은 99.86g(3.33% yield)을 얻었다.
MajorExtract를 다시 Hexane, Butanol, EA 유기용매를 사용하여 분획을 한 결과 hexane에선 87.34g(2.91%), Butanol은 72.48g(13.9%), EA는1.18g(0.04%), 그리고 나머지 H2O에서는 336.9g(11.22%)를 얻었다.노랑다발 동충하초 시료는 ICR 마우스에 대한 급성독성시험 결과, 독성학적 이상변화 및 치사한 개체가 관찰되지 않았으므로 1000mg/kg의 용량에서도 독성발현이 없는 안전한 물질로 사료된다.아토피치료기전에 적합한 동충하초 외용제의 제제조성 수립·동충하초 함유 크림제의 제조공정 수립.동충하초의 BuOH 분획(CBBF)은 mushroom tyrosinase의 발현을 농도 의존적으로 억제하였고 MTT assay를 통하여 독성이 없음을 확인하였다.
다. 유해물질 분석 및 시험방법 정립
- 노랑다발동충하초 추출물 및 분획물의 항염 실험을 진행하였으며, Hexane분획물의 IC50은 0.88mg/ml였으며, Buthanol분획물의 IC50은 0.34mg/ml를 보였으며, Ethylacetate분획물의 IC50은 0.6mg/ml를 보였다.실험의 결과, Buthanol분획물의 IC50이 0.34mg/ml으로 가장 좋은 결과를 보였다.이를 바탕으로 Buthanol분획물(CBBF)를 silica chromatography를 이용하여 정제분리하였다.정제분리된 분획물의 항암 및 항염 실험을 진행하였으며, Cloroform의 함량이 100% 와 90%에서 가장 좋은 항암 및 항염 결과를 얻었다.1차 분리한 분획물을 2차 Silica chromatography 이용하여 재분리하였고, 이를 이용하여 항암 및 항염 특성을 확인하였다. 그 결과, 100-1, 100-2, 100-3, 100-4, 90-1, 90-2, 20, 0에서 항암 효능을 보였으며, 90-4및 90-5에서 항염 효능을 보였다.2차 활성 분획을 합친 후 HPLC 및 LC-MASS로 분석한 결과, 노랑다발 동충하초인 Cordycepsbassiana에 Beauvericin 이 함유되어있다는 것을 확인하였다. 노랑다발 동충하초의 Beauvericin의 항염, 세포독성 그리고 항암 효능을 평가한 결과, Beauvericin은 세포독성이 강하여 항염제로 사용할 수 없고 항암 효능이 있음을 알 수 있었다.
3. 동충하초의 invivo아토피성 피부염 억제 효능 및 예비임상 연구
가. 동충하초의 invivo아토피성 피부염 억제 효능 및 예비임상 연구
DNFB를 이용한 아토피 피부염에서는 대조군에 비해 CBBF-Ep를 처리한 NC/Ngamice의 아토피 피부염이 억제되었다.그러나, miteantigen으로 유발한 피부염에서는 CBBF-Ep에 의해 억제되지 피부염이 억제되지 않았으며, 대조군인 steroid물질에서도 억제하지 않는 것으로 보아 아토피 피부염을 연구에 부적합한 모델로 판단되었다.TPA를 acetone에 용해하여 쥐의 귀에 염증을 유도하고 oliveoil에 혼합한 CBBF-Ep를 처리한 결과 쥐의 귀 두께가 낮아진 것을 확인하였으며, 광학 현미경에 의한 관찰에서도 진피층의 부종이 감소한 것을 확인하였다.
나.동충하초 추출분획의 항 아토피 효능에 관한 invivo연구:제품화에 따른 효능 차이 연구
- 아토피군에서는 정상군에 비해 표피층이 두꺼워졌고 표피 돌기가 진피 속으로 깊이 침투하였으며 염증성 세포의 침윤이 증가하였으며 이러한 변화는 CBW-Es로션을 도포하면 개선되었다.0.2% DNFB를 3일에 1회씩 3회 처리하여 아토피 피부염을 유발하면서 노랑다발 동충하초 추출물인 CBW-Ep 로션을 환부에 14일간 도포였다.그 결과 CBW-Ep 로션은 DNFB로 유발한 아토피 피부염의 임상증상에 아무런 영향을 미치지 않았다.
다. 항아토피 제품의 예비임상평가
- 물 추출, 여과, 감압농축, 에탄올 분획, 원심분리, 감압농축, 동결건조 등 7개의 단계를 거쳐 CBWE를 추출하여 20mg/ml농도의 크림을 제조하였다.아토피 피부염 유발 조건에서, 첫날부터 CBWE를 20㎎/㎖의 농도로 크림에 혼합하여 매일 2회씩 14일간 환부에 도포하였다. 그 결과, 피부염 지수가 CBWE를 20㎎/㎖의 농도로 도포한 군이 아토피 대조군에 비해 유의하게 감소하였다.또한 아토피성 피부염 환부에 CBWE를 도포한 후 병리학적 변화 및 비만세포 침윤 상태를 관찰한 결과, 표피층 두께, 비만세포 침윤 상태 그리고 혈청 IgE 농도가 아토피 대조군에 비해 CBWE를 20㎎/㎖의 농도로 도포한 군에서 측정치가 유의하게 감소하였다. 이러한 결과를 바탕으로, 아토피성 피부염을 앓고 있는 환자에서 CBWE를 20㎎/㎖ 함유하는 크림을 환부에 매일 2회씩 도포하였다.그 결과, 크림 도포 7일 후부터 지원자 10명 중 4명에게서 아토피성 피부염에 대한 만족스러운 억제 효과가 나타났으며 3명에서는 약간의 억제 효과가 관찰되었다. 그리고 나머지 3명의 아토피성 피부염은 개선되지 않았다.

A. In vitro anti-atopic effect (Thl/Th2) test
We first evaluated the suppressive effect of CBBF and CBEE on the secretion of IFN-gamma and IL-4. CBBF and CBEE blocked the IFN-gamma release at all stimulus conditions, while IL-4 secretion was inhibited only at Con A and LPS treatment conditions. And also, we tested the effect of CBBF and CBEE on the production of NO, well-known inflammatory mediator. CBBF blocked effectively NO release considering the effect on cell viability. Then, we studied the cell to cell aggregation by CBBF and CBEE, CBBF diminished cell to cell aggregation mediated by CD29, and CBEE decreased cell aggregation induced by CD43 at 2 h. CBBF suppressed the phosphorylation level of ERK at 15 min, CBEE suppressed the phosphorylation of p38 at 5 min. And, we studied whether CBBF and CBEE blocked promoter activity of transcriptional factors which are involved in inflammatory responses. CBBF dose-dependently reduced the promoter activities of AP-1, NF-kB, and CREB. But, CBEE blocked only AP-1 and CREB promoter activities.
B. In vitro anti -atopic effect (APC/mast cells) test
Both CBEE and CBBF which were extracted from Cordyceps bassiana inhibited transcriptional level of IL-12. So, we explored the release of TNF-a, PGE2, and NO which are associated in histamine secretion. As a result, CBBF weakly decreased TNF-a secretion at all concentration groups and dose-dependently reduced NO production, while the release of PGE2 was up-regulated depending on the concentration of CBBF. CBEE and CBBF did not affect the attachment induced by fibronectin. Based on our results, we suggest that CBBF would regulate IL-12 through p38. We also observed that p38 inhibitor can block the expression of IL-12, which supports our suggestion. And also we expect CBBF might inhibit the STAT-I-induced IFN -gamma production by regulating JAK2 phosphorylation. Additionally, CBEE exhibited inhibitory activity against translocation of p65 and c-Jun into nucleus. CBBF also blocked the translocation of STAT-1 and c-Fos.
2. Chemical, biochemical, and pharmaceutical properties study of Cordyceps bassiana effective fractions and components
A. Component characterization and the improvement of extraction process.
- We analyzed the patterns of sugar included in each extract using HPLC. The ethyl acetate extract and the butanol extract showed peak that retention time was slow and had non-polar. Generally, Cordyceps bassiana has been extracted in water. So we compared HPLC pattern of extract at 4℃ and 100℃ with water fraction. There was no significant difference between each fraction. Water fraction did not show polar components within 0 to 10 min. We analyzed fatty acid in total lipids of Cordyceps bassiana using gas chrornathgraphy. As a result, It was revealed that Cordyceps bassiana contains 80% of unsaturated fatty acid. We have sub-fractionated the aqueous layer of Cordyceps bassiana to hexane, ethyl actate, butanol, and water extracts. We got 66.1g of ethanol extract from 360g of Cordyceps bassiana, and gained water fraction (63.7%), hexane fraction (22%), butanol (7.3%) and ethyl acetate fraction (7.3%) from ethanol fraction, respectively. We conducted acute oral toxicity test based on Korean Food & Drug administration toxicity testing protocol to verify the safety of Cordyceps bassiana butanol fraction. As a result, we confirmed Cordyceps bassiana butanol fraction with dose of 100 mg/kg was safe. The soluble components of Cordyceps bassiana butanol fraction had a low solubility than insoluble fraction, but it had a high solubility in Tween 80. Generally insoluble fraction can be easily dissolved in diverse medium. It seems that a medium used in our experiment was suitable for dissolution of insoluble fraction.
B. Separation of functional components and establishment of analysing method (structure analysis)
- We extracted 533.7 g (17.8% yield) of extract by 100% EtOH extraction from 3,000 g of Cordyceps bassiana, 39.16 g (1.31% yield) of white precipitate by cooling and 99.86 g(3.33% yield) of pink crystal. Major extract was re-extreated by organic solvents such as hexane, butanol and EA. As a result, we got 87.34 g(2.91%) of extract by hexane extraction, 72.48 g(13.9%) of extract by butanol extraction, 1.18 g(0.04%) of extract by EA extraction and 336.9 g(11.22%) of extract from H20. Cordyceps bassiana sample, therefore, is considered safe material because this sample did not observed toxicological change and lethality and it also had non-toxicity in 1000 rng/kg dosage. We set up a medicine component of suitable external preparation of Cordyceps bassiana for atopy dermatitis therapy and the process of manufacture for Cordyceps bassiana-containing cream. BuOH fraction of Cordyceps bassiana inhibited expression of mushroom tyrosinase in dose dependent manner and had non-toxic activity in MTT assay.
C. Toxic substance analysis and testing method establishment
- We estimated the anti-inflammatory activity of Cordyceps bassiana extracts. First, we investigated IC50 value of each fraction. Hexan, butanol, and ethyl acetate extracts exhibited IC50 values of 0.88 mg/ml, 0.34 mg/ml, and 0.6 mg/ml, respectively. Based on this result, the buthanol extract (CBBF) was selected to identify its active components using silica chromatography. We also conducted a series of experiments to demonstrate anti-cancer and anti-inflammatory effects of CBBF. The most effective activity was revealed in the extract which was prepared with 100% and 90% of chloroform. We re-segregated the primary extract into secondary fraction using silica chromatography, and we verified that the latter conditions were able to show higher anti-cancer and anti-inflammatory effects. As a result, 100-1, 100-2, 100-3, 100-4, 90-1, 90-2, 20, 0 had anti-cancer effect, while 90-4 and 90-5 exhibited anti-inflammatory function. When we analyzed the combined secondary fractions using HPLC and LC- MASS, we were able to identify a compound called beauvericin from the Cordyceps bassiana. So, we tested the anti-cancer and anti-inflammatory activity of this compound. Unfortunately, beauvericin had strong cytotoxicity, so we could not use it as an anti-inflammatory drug although it displayed anti-inflammatory activity.
3. The inhibitory effect of Cordyceps bassiana on atopic dermatitis and preliminary clinical experiment in vivo
A. The inhibitory effect of Cordyceps bassiana on atopic dermatitis and preliminary clinical experiment in vivo
- The atopic dermatitis was relieved by CBBF-Ep treatment comparing with control group in DNFB-induced atopic dermatitis model. In addition, we tested the anti-inflammatory properties of Cordyceps bassiana using another dermatitis model: mite antigen-induced dermatitis model. But, steroid which had been used as positive control in dermatitis model didn't show effect against dermatitis in this model. So, we concluded this model is unsuitable for atopic dermatitis model. And also we induced inflammation on the mice ear skin with treatment of TPA dissolved in acetone, then the ears were assessed for abnormalities, edema. We observed the decreased ear thickness and edama on ear dermis.
B. In vivo study of anti-atopic effects by extracts of Cordyceps bassiana a study of difference in effects by making product
- In atopy dermatitis group, there was an increase of the thickness of epidermis and infiltrated deep dendrite in epidermis. Thus, the number of inflammatory cells infiltrating in atopy dermatitis mice was increased. On the other hand, CBW - Es treated group was suppressed in this features. Next, we treated CBW - Es lotion with Cordyceps bassiana extract at affected area for 14 days after induced atopy dermatitis by treating 0.2% DNFB three times (one time per 3 days). As a result, CBW-Es lotion has no effect on atopy dermatitis symptoms induced by DNFB.
C. Preliminary clinical trial of anti-atopic product using Cordyceps bassiana
- We extracted CBWE on 7 step of extraction such as water extraction, filtration, decompressed concentration, ethanol fractionation, centrifugation, decompressed concentration, and lyophilization, and made 20 mg/ml concentration of CBWE-containing lotion. In atopy dermatitis-induced condition, we mixed cream and 20 mg/ml CBWE and treated the affected area two times a day for 14 days. As a result, atopy dermatitis score was significantly decreased at 20 mg/ml CBWE lotion treated group compared with atopy control group. We also observed pathological changes and mast cell infiltrating after treatment of CBWE on affected area of atopy dermatitis. As a result, 20 mg/ml CBWE treated group significantly decreased epidermal thickness, mast cell infiltration, and serum IgE concentration compared with atopy control. Following these results, we tried a preliminary clinical trial for atopy dermatitis patients by treating 20 mg/ml CBWE-containing cream on affected area two times a day. As a result, after 7 days, 4 patients had satisfactory effects and 3 patient had a little effect for atopy dermatitis treatment. Finally, another 3 patients had no effect for atopy dermatitis.
