피부 건강 증진 관련한 생리활성 지질의 타겟으로 식물세라마이드 및 대두 포스파티딜세린을 대상 소재로 선정하였다. 식물세라마이드 생산을 위한 원료로서 미강박, 쌀겨, 대두유 검 비지박 등의 원료들로부터 식물세라마이드 함량을 검토한 결과 대두유 검으로부터 생산하는 것이 비교적 가장 경제적인것으로 판단되었다. 국내에서 공급 가능한 대두유 유래 검질을 수배하여 식물 세라마이드를 함유한 생리활성 지질을 추출할 수 있는 최적 조건을 수립하였고 대량 생산을 위한 공장 시 생산 테스트를 실시하였다. 그리고 대두 유래의 식물 세라마이드를 순수 분리

생리 활성 지질 후보군으로서 식물 세라마이드외에 대두유 부산물인 대두 검질로부터 인지질을 농축정제하여 포스파티딜세린을 제조하는 것을 검토하였다. 기존의 두산이 개발한 대두 포스파티딜세린 공정에는 해외에서 수입된 정제 분말 레시틴을 활용하여 제조된 중순도 PC 함량의 대두레시틴 (PC함량55)을 사용하여 생산하여 왔다. 이에 국내에서 생산된 조레시틴을 활용하여 포스파티딜세린 공정을 대두검으로부터 대두 인지질을 추출하여 포스파티디세린 제조를 검토하였으며 국내에서 건강 기능식품으로 인정 받은 수준의포스파티딜세린 함량 40% 이상의 포스파티딜세린을 얻을 수 있었다.

In vitro에서의 ceramide와 PS의 미백 효능을 분석하기 위하여 B16F10 흑색종(melanoma) 세포에서의 melanin 생성 저해효과와 tyrosinase의 활성에 대한 ceramide와 PS의 저해 효과를 조사하였다.

Hairless mouse를 이용하여 ceramide와 PS의 장기간 경구섭취가 UV 조사에 의한 주름 생성에 미치는 영향을 조사하였다. 피부 수분함량의 경우 실험 10주차에 UV 대조군이 크게 감소한 반면 PS투여군은 UV 대조군에 비해 유의적으로 증가하였으며, UV 대조군의 경우 피부 주름이 깊어지고 개수도 크게 증가하였지만 PS 투여군은 UV 대조군에 비해 주름이 옅어지고 개수도 크게 감소하는 것으로 나타났다. 주름면적의 경우에도 정상대조군 대비 UV 대조군에서 크게 증가하였지만 PS 투여군에서는 UV 대조군에 비해 크게 감소하는 것으로 나타났다. 따라서 PS를 장기간 경구섭취할 경우 UV에 의한 피부 조직의 구조 손상에 따른 주름 발생과 탄력저하를 완화시킬 뿐만 아니라 피부 장벽기능 개선에도 도움을 줄 것으로 판단된다. 한편 ceramide의 in vivo 주름개선 효능시험을 한 결과, 피부 수분함량의 경우 UV 대조군이 감소한 반면 ceramide 투여군은 UV 대조군에 비해 증가하였으며 주름면적의 경우에도 ceramide 투여군에서는 UV 대조군에 비해 크게 감소하는 것으로 나타났다.
Ceramide와 PS의 경구섭취가 hairless mouse의 피부 보습에 미치는 영향을 조사하였다. Tape stripping한 TS 대조군의 경우 정상대조군에 비해 TS후 급격히 경피수분손실량(TEWL)은 증가되고 서서히 회복되는 반면 ceramide와 PS 투여군에서는 TS 처리시 피부 장벽손상이 적게 일어나 낮은 TEWL 값을 나타내었다. 농산 부산물로부터 제조한 ceramide와 phosphatidylserine(PS)의 항염증효능을 in vitro 및 in vivo 수준에서 검정하였다. 글루코실세라마이드는 LPS에 의해 유도된 대표적인 proinflammatory cytokine인 IL-1β 및 TNF-α 발현을 농도의존적으로 강하게 저해하였다.
비록 PS 또한 IL-1β 및 TNF-α 발현을 저해하였으나, 글루코실세라마이드에 비해 낮은 효능을 보였다. 류마티스 관절염 환자 유래 fibroblast-like synoviocyte에서도 글루코실세라마이드는 IL-1β에 의해 유도된 IL-6, IL-8 및 PGE2와 같은 염증성 인자의 발현을 농도의존적으로 강하게 저해하였다. 또한 연골 파괴 관련 MMPs 중 하나인 MMP-1은 IL-1β에 의해 그 발현이 증가되는데, 글루코실세라마이드가 이러한 MMP-1의 발현을 억제하였다. 반면 PS는 글루코실세라마이드에 비해 낮은 효능을 나타내었다. 14일 간 지속적인 trimellitic anhydride (TMA) 도포로 유도된 아토피성 피부염 모델을 이용하여 글루코실세라마이드 및 PS의 항아토피성 효능을 검사하였다. 생쥐의 귀에TMA의 반복적인 도포는 귀에서 사람의 아토피 피부염과 유사한 증상인 erythema, scarring/dryness, erosion이 나타내는데, 글루코실세라마이드는 농도의존적으로 임상적 증상 및 귀 두께로 측적된 부종을 완화시켰으며, 조직 내 비만세포(mast cell) 및 염증성 세포들의 침윤을 억제하였다. 더 나아가 혈청 내 아토피의 마커인 IL-4의 농도를 감소시켰으며, 아토피성 피부 조직 및 림프절에서 Th1 사이토카인인 IFN-γ와 Th2 사이토카인인 IL-4의 mRNA 발현은 물론 염증성사이토카인인 IL-1β와 TNF-α의 mRNA 발현 역시 농도의존적인 수단으로 억제하였다. 반면 PS는 ceramide에 비해 낮은 효능을 나타내었다.
또 다른 피부염 모델인 Oxalozone으로 유도된 피부습진 동물모델에서 글루코실세라마이드는 글루코실세라마이드는 농도의존적으로 임상적 증상 및 귀 두께로 측적된 부종을 완화시켰으며, 조직 내 염증성 세포들의 침윤을 억제하였다. 특히 oxazolone의 반복적인 도포는 피부 내 수분 함량손실과 수분통로 단백질인 aquaporin-3 (AQP-3)의 발현을 감소를 초래하였는데, 글루코실세라마이드는 경구 투여는 농도 의존적인 수단으로 피부 내 수분 함량을 상당한 수준으로 회복시켰으며, AQP-3의 발현 역시 정상군 수준으로 회복되었다.
결론적으로, 글루코실세라마이드 및 PS는 in vitro 및 in vivo에서 항염증 효과를 나타내었으며, 이는 관절염, 아토피, 접촉성 피부염과 같은 염증성 질환을 완화시키는데 도움이 될 것으로 사료된다.
피부 관련 건강 기능 식품 소재로서 타겟 효능에 있어서는 항염증, 아토피 개선 등의 효능 효과는 국내 건강 기능식품 법규상 취득하기 불가한 효능이므로 건강 기능 식품으로서의 효능으로서는 보습과 주름 개선이 보다 적합하다고 판단하였다.
따라서 본 연구에서는 대두유래의 식물세라마이드와 대두검질 유래의 인지질을 이용하여 제조한 대두포스파티딜세린을 활용하여 국내 전문 임상 기관에서 임상 시험을 수행하였다. 그 결과 대두 우래 식물 세라마이드 및 포스파티딜세린은 모두 섭취전에 비하여 섭취 12주후 피부보습개선효과가 대조군에 비해 시험군이 통계적으로 유의한 수준 (p<0.05)으로 개선 시키는 것으로 나타났다. 피부주름 개선 효능에 있어서는
안전성 평가 결과, 이상반응을 경험한 피험자의 보고는 없었고, (활력징후)에 대해서는 군내에서 통계적으로 유의한 수준의 차이를 보인 항목들이 있었으나 정상치 범위내에서의 변화였다. 또한, 본 시험기간동안 중대한 이상반응(SAE)뿐만 아니라 이상 약물반응(ADR) 및 이상 반응으로 인한 조기종료는 없었다.
결과적으로 대두 포스파티딜세린과 대두 당세라마이드는 눈가 주름 및 피부 보습 개선에 효과가 있는 소재임을 알 수 있었다.

Plant ceramide and soy phosphatidylserine were selected as the physiologically active lipids for the improvement of skin beauty and skin health. Rice bran, soy bean gum, and soy bean curd were evaluated as raw materials for sugar-ceramide. Among them, soy bean gum was the most suitable source as a raw material in terms of yield and availability. Optimization for the production of sugar ceramide was performed from soy bean gum and the soy ceramide 40% was produced as a pilot test. We analyzed the structure of the GlcCer using FAB-Mass spectrometry. The GlcCer used in this study was comprised primarily of ceramide with 4,8-sphingadiene (d18:2) and alpha-hydroxypalmitic. Solubilization formula for nano sized emulsion of soy ceramide was studied for the application into beverage in addition to powder type product.
In addition to plant ceramide from soy, production of lecithin from soy bean gum was tried as a raw material for the synthesis of phosphatidylserine using enzyme. As a result, phosphatidylserine 40% could be produced from the lecithin.
We evaluated the effect of ceramide and phosphatidylserine (PS), made from agricultural by-products, on skin health improvement in terms of whitening, wrinkle improvement, moisturization and so on. To test the whitening effect of ceramide and PS in vitro, the inhibitory effect of ceramide and PS against melanin formation and tyrosinase activity in melanoma cell line, B16F10. Though there was no dose-dependent activity, ceramide showed higher inhibitory effect on melanin biosynthesis compared to positive control, arbutin. The tyrosinase activity was also inhibited by 17.2% and 17.6% at the level of 1 and 2.5 μg/mL of ceramide, respectively, demonstrating that ceramide has higher inhibitory effect against tyrosinase compared to positive control, kojic acid at the same dose level. To dissect the mechanism of melanin biosynthesis inhibition by ceramide, we measured the protein level of tyrosinase, TRP-1 and TRP-2 in melanoma cell line, B16F10 on Western blotting analysis, resulting that TRP-2 level was not affected but the level of tyrosinase and TRP-1 was reduced by ceramide treatment. On the other hand, PS showed relatively less whitening effect compared to ceramide.
The anti-wrinkle effect of ceramide and PS in vitro was evaluated by analysing collagen synthesis, and the expression level of MMP-1, MAPKs and AP-1 after treating ceramide and PS to human dermal fibroblast. PS increased the collagen biosynthesis compared to control. At the level of 10 μg/mL, PS greatly decreased MMP-1 expression level activated by UVB irradiation on the Western Blot. To investigate the affected signaling pathway by PS resulting in MMP-1 down-regulation, the inhibitory effect of PS on phosphorylation of MAPKs (JNK, ERK, P38) was analyzed on the Western blot. The results showed that PS could dose-dependently reduce phosphorylation on all kinds of MAPKs which was drastically increased by UVB irradiation, and the phosphorylation on ERK was the most greatly inhibited by PS. The inhibitory effect of PS on AP-1 activation by UVB was also analyzed by measuring the expression level of c-Jun and c-Fos on the Western Blot, showing similar result with MAPKs study. Ceramide also showed the similar anti-wrinkle effect compared to PS by greatly increasing the collagen biosynthesis.
The effect of long-term oral administration of ceramide and PS on wrinkle formation by UV irradiation was investigated in hairless mouse model. At week 10, the skin moisture content of PS group was significantly increased by 56.3±7.6∼72.1±4.2% of normal control group, while that of UV control group was decreased by 40.0±5.0% of normal control group. The skin thickness of PS group was also significantly decreased compared to that of UV control group while UV control group showed increased skin thickness compared to normal control group. Moreover, PS group showed reduced wrinkle depth and much less wrinkle number, unlike UV control group showing deepened wrinkle depth and increased wrinkle number compared to normal control group. The wrinkle area of UV control group was 123.8±12.6 mm2, which was significantly increased that of normal control group, 53.5±7.5 mm2. However, the wrinkle area of PS group was 67.0±2.5∼83.3±15.3 mm2, showing great decreased compared to that of UV control group. In the histopathological analysis, the thickness of epidermis and dermis was observed to be increased in UV control group compared to that in normal control group. However, the thickness of epidermis and dermis in PS group was dramatically decreased in dose dependent manner.
Therefore, the results suggest that long-term oral administration of PS may promote skin barrier function as well as bring improvement in wrinkle formation and elasticity decrease induced by structural damage of skin tissue on UV exposure. In case of ceramide, the skin moisture content of ceramide group was increased by 60.5±8.9∼78.6±7.1% of normal control group, while that of UV control group was decreased by 63.2±7.3% of normal control group. The skin thickness of ceramide group was also significantly decreased compared to that of UV control group while UV control group showed increased skin thickness compared to normal control group. The wrinkle area of UV control group was 32.7 mm2, which was significantly increased that of normal control group, 18.3 mm2. However, the wrinkle area of ceramide group was 16.2∼29.6 mm2, showing great decreased compared to that of UV control group. In the histopathological analysis, it was observed that the epidermis thickness was increased and collagen content was decreased in UV control group compared to that in normal control group. However, the epidermis thickness was decreased and collagen content was increased in ceramide group in dose dependent manner.
In this study, we investigated the effect of oral administration of PS and ceramide on skin moisturizing using hairless mouse. In control group, tape stripping (TS) induced acute increase of transepidermal water loss (TEWL) which was slowly recovered later. On the other hand, ceramide and PS group showed lower damage on skin barrier resulting in lower TEWL value after TS application. Especially, low-dose ceramide group showed persistent increase in recovery rate as well as lower skin damage. Moreover, histopathological study using H&E staining revealed that the epidermis thickness was recovered as thin as that of normal control group by oral administration of ceramide and PS, which was increased in TS control group compared to normal control group. Therefore, it is suggested that oral administration of ceramide and PS may effectively prevent and/or recover skin from sensitization or skin barrier damage.
We also evaluated the anti-inflammatory effect of glucosylceramide (GlcCer) and phosphatidylserine (PS), made from agricultural by-products, in vitro and in vivo . First of all, the effects of GlcCer on the production of IL-1β and TNF-α in LPS-stimulated RAW264.7. Treatment with GlcCer alone or with LPS did not cause any significant cell viability change, indicating that GlcCer was non-cytotoxic to RAW 264.7 upto 100 μg/ml.
Exposure of RAW264.7 to LPS (1 μg/ml) increased their release of IL-1β and TNF-α, which was significantly inhibited by GlcCer in a dose-dependent manner. Although PS also inhibited IL-1β and TNF-α, PS showed relatively less effect compared to GlcCer. GlcCer significantly inhibited MMP-1 as well as proinflammatory mediators such as IL-6, IL-8 and PGE2 in a dose-dependent manner in IL-1β-stimulated RA-FLS. On the other hand, PS showed relatively less effect compared to GlcCer. Next, to test the whitening effect of ceramide and PS in vitro, we investigated the effects of GlcCer and PS on proliferation, cell migration, and tube formation in BAEC. Both GlcCer and PS had no effects on proliferation, cell migration, and tube formation.
The analgesic and antiarthritic activities of GlcCer were investigated in rat models of carrageenan-induced arthritis by measuring weight distribution ratio and knee circumference. In WDR test, before the carrageenan injection (day 0), the mean WDR did not differ significantly among the experimental groups (the WDR was 50:50, thus controls carried 50% of the weight on each hind paw). However, significant changes in the ratio were observed on day 1 after the carrageenan injection, and the weight carried by the affected leg in the vehicle-treated arthritic group reached 20% . Distinct increase of WDR was observed in groups that received GlcCer. Reduction in knee circumference demonstrated that oral GlcCer administration suppressed the carrageenan-induced edema.
Histological staining showed that GlcCer significantly reduced the inflammatory area and the infiltration of inflammatory cells in the arthritic joints. PS also showed the similar anti-arthritic effect compared to GlcCer.
To investigate anti-atopic effects of GlcCer, we used chronic mouse model of trimellitic anhydride (TMA)-induced atopic dermatitis (AD). Chronic (repeated) TMA treatment on the dorsum of both ears caused typical morphological changes of the inflamed skin such as erythema, scarring/dryness, and erosion, which was significantly inhibited by GlcCer. Also, GlcCer inhibited the strong cutaneous cell infiltration into the inflammed ear.
Furthermore, GlcCer also significantly reduced the levels of mRNA of IL-1β, TNF-α, IFN-γ and IL-4 increased in TMA-applied mouse ears and lymph node. On the other hand, PS showed relatively less effect compared to GlcCer.
In another animal model for inflammatory skin disease, mouse model of oxazolone-induced contact hypersensitivity, GlcCer significantly inhibited typical morphological changes of the inflamed skin, such as erythema, scarring/dryness, and erosion, and strong cutaneous cell infiltration into the inflammed ear. In oxazolone-induced ICD mice, GlcCer significantly inhibited irritant-related scratching behavior and dehydration of the stratum corneum, and decreased aquaporin-3 expression.
Overall, GlcCer and PS has antitinflammatory effect in vitro and in vivo, suggesting that they can improve chronic and inflammatory diseases such as arthritis, atopy and contact dermatitis.
Human clinical test for the efficacy evaluation of moisturizing activity and anti-wrinkle activity of soy ceramide and phosphatidylserine was performed.
A placebo-controlled double-blind ingestion study of a soft capsule type dietary supplement containing soy-derived ceramide and soy phosphatidylserine was performed in 103 normal subjects by 12 week ingestion test, and the following results were obtained: (1) Dermatological diagnosis by physicians and by instrumental analysis showed that the supplement significantly improves anti-wrinkle activity.
. (2) On measurement of water content in the skin, the supplement was shown to significantly increase the water content in the skin. The above findings showed that 3 months ingestion of a supplement soy-derived ceramide and phosphatidylserine are effective in moisture-retention and anti-wrinkle activity, and thus, is an effective skin-beautifying food.
