연구의 목적 및 내용:
EMT는 개체 발생과정 중 상피 기원의 외배엽에서 간엽 조직이 형성되는 세포학적 현상으로, 상피 기원 암세포의 침윤성 성장 및 전이를 설명하는 세포생물학적 기전이며, 나아가 줄기세포 의 특성으로 인정되고 있음. EMT를 조절하는 다양한 인자는 알려져 있으나 EMT 조절 경로는 잘 알려져 있지 않음. E-cadherin의 전사억제자인 Snail 발현 조절이 EMT와 암세포의 전이 과 정에서 결정적인 것으로 알려져 있으며, Wnt 신호전달은 beta-catenin을 통한 암 발생기전의 가장 중요한 원인으로서
연구의 목적 및 내용:
EMT는 개체 발생과정 중 상피 기원의 외배엽에서 간엽 조직이 형성되는 세포학적 현상으로, 상피 기원 암세포의 침윤성 성장 및 전이를 설명하는 세포생물학적 기전이며, 나아가 줄기세포 의 특성으로 인정되고 있음. EMT를 조절하는 다양한 인자는 알려져 있으나 EMT 조절 경로는 잘 알려져 있지 않음. E-cadherin의 전사억제자인 Snail 발현 조절이 EMT와 암세포의 전이 과 정에서 결정적인 것으로 알려져 있으며, Wnt 신호전달은 beta-catenin을 통한 암 발생기전의 가장 중요한 원인으로서, E-cadherin 전사억제자인 Snail의 인산화와 반감기를 조절함으로써 암 세포의 침윤성 성장과 전이를 조절함이 밝혀짐.
본 연구팀은 (1) Wnt 신호 전달에 의한 Snail 발현 및 EMT 조절, (2) p53에 의한 Snail 발현 및 EMT 조절 기전, (3) Snail에 의한 miRNA 발현 조절 기전 규명과 EMT 조절 miRNA 발굴에 대한 연구를 통해 사람의 암에서 가장 흔한 암 발생 인자 중에 하나인 p53 유전자와 주요 암발생 신호전달체계 중 하나인 Wnt 신호 전달에 의한 Snail 발현 조절 기전을 규명하고, 나아가 Wnt/Snail axis를 표적화하여 암 전이를 억제할 수 있는 기술 개발을 목표로 함
연구결과:
1. Wnt 신호 전달에 의한 Snail 발현 및 EMT 조절:
: EMT 주요유도자인 Snail 단백이 glycosylation되어 stabilization됨을 밝혀, Snail이 glucose metabolism과 EMT를 잇는 중요한 연결 고리일 가능성을 제시함. 또한, Snail이 DNA double-strand breaks를 조절하는 DNA-PKcs와 결합하여 kinase activity를 감소시킴을 밝혀 암세포가 방사선 및 항암 치료 내성을 띄는 원리를 규명함. Wnt 신호전달에 의해 Hippo-YAP axis가 조절됨을 밝혀 암세포의 분열과 이동의 조절기전을 밝힘. 또한 위암의 원인인자로 알려 진 Helicobacter pylori의 oncoprotein인 CagA가 GSK3와 결합하여 Snail에 의한 EMT를 촉진 시키는 기전을 밝혀 박테리아에 의한 위암진행 기전을 밝힘. 이러한 연구결과는 EMBO Journal (2010), Cell Death & Disease (2013) 등의 일련의 논문으로 발표였고, ‘Helicobacter pylori CagA promotes Snail-mediated epithelial mesenchymal transition through the depletion of endogenous GSK-3β’ (Nature Communications 2014) 논문은 현재 revision 중에 있음
2. p53에 의한 Snail 발현 및 EMT 조절 기전:
: 사람의 암에서 가장 흔한 암 발생인자 중 하나인 p53 암억제 유전자가 microRNA-34 family를 이용하여 Wnt 신호전달 및 Snail 발현과 EMT를 조절하는 것을 밝혀 p53 암억제 유전자가 암발생뿐만 아니라 침윤성 암 진행도 조절함을 밝히고, miRNA가 p53 암억제 유전자와 암발생 신호전달의 link 역할을 함을 밝힘. 이러한 연구결과는 Journal of Cell Biology (2011), Science Signaling (l 2011), Cell Cycle (2012) 등 일련의 논문으로 발표함
3. Snail에 의한 miRNA 발현 조절 기전 규명과 EMT 조절 miRNA 발굴:
: EMT 조절 과정에서 Axin2/GSK3 결합부위의 조절이 암진행에서 중요함을 밝힌 선행 연구를 바탕으로, p53 암억제 유전자는 miRNA-34를 통해 Axin2에 의한 GSK3 핵외이송을 억제하여 암진행을 억제함을 밝힘. Axin2/GSK3 결합을 표적화하는 약물의 개발이 항전이 억제제의 Mode of action으로서 타당함을 밝힘. 이러한 연구결과는 ‘세포핵에서 세포질로의 GSK3의 이동을 억제하는 화합물을 함유하는 세포핵에서 세포질로의 GSK3 이동에 의해 발생되는 질환의 치료 또는 예방용 약햑적 조성물’ (국내특허 및 PCT 출원, 2010) 및 Cell Cycle (2013) 등 일련의 특허 및 논문으로 발표함
연구결과의 활용계획:
(1) miRNA를 통한 p53 암억제 유전자와 Wnt 신호전달간의 상호 조절 기전을 통해 암진행의 새로운 패러다임을 제공하여 EMT 조절 RNA network 후속 연구로 활용될 수 있음
(2) 암환자의 가장 큰 사망 원인인 재발과 전이를 유도하는 EMT의 세포생물학적 기전을 규명함
(3) 암세포의 전이를 억제할 수 있는 분자 표적 및 실험적 근거를 확보로 전이억제제에 대한 chemical screening을 통해 전이억제제 후보를 도출할 수 있을 것임

EMT (epithelial-mesenchymal transition) is a cellular phenotypic changes from ectoderm-derived epithelium to mesenchyme during the developmental process, and EMT is also regarded as a cellular mechanism on invasive program and metastatic progression of carcinoma cells. Although several factors regulating EMT were described, the regulatory cascade of these are not well-known. The expression of Snail, an E-cadherin repressor, is known to be regulated by Wnt signaling thereby resulted in invasive program of carcinoma cells.
This study, therefore, is aimed to elucidate the roles of p53 tumor suppressor and Wnt signaling regulating Snail and EMT, and to discover the potential target of Wnt/Snail axis for development of new cancer therapeutics through the elucidation of (1) Regulation of Snail and EMT by Wnt signaling, (2) Regulation of Snail and EMT by p53 tumor suppressor, (3) Identification of EMT regulating miRNAs those are transcriptionally repressed by Snail.
Result:
1. Regulation of Snail and EMT by Wnt signaling:
: We discovered that Snail1 is stabilized by O-GlcNAc modification, which suggest that Snail constitute a molecular link between cellular glucose metabolism and the control of EMT. Results have published in EMBO Journal (2010). We discovered that Snail binds with DNA-PKcs, the important kinase in non-homologous end joining (NHEJ) of DNA repair, and inhibits the kinase activity of DNA-PKcs and induces radiation resistance. Results have published in Cell Death & Disease (2013). We also discovered that Helicobacter pylori CagA promotes Snail-mediated EMT through the depletion of endogenous GSK3, which results have been submitted and the manuscript is in revision at Nature Communications.
2. Regulation of Snail and EMT by p53 tumor suppressor:
: We discovered that tumor suppressor p53 regulates Wnt/Snail axis and EMT through transactivation of microRNA-34 (miR-34) family, which suggest that p53 and Wnt signaling pathway are tightly linked and plays a important roles in tumor progression as well as tumor development. and microRNA intrinsically links p53 tumor suppressor and oncogenic Wnt signaling pathway. Results have published in Journal of Cell Biology (2011), Science Signaling (2011), Cell Cycle (2012).
3. Identification of EMT regulating miRNAs those are transcriptionally repressed by Snail:
: We have validated the Proof of Concept(PoC) of Axin-GSK3β interaction site as a therapeutic target of Wnt/EMT axis.
