To date, lots of African yam lines have been used for food without accurate metabolic analysis. The results shown in this study represent the considerable improvement in the metabolic evaluation of African yams and the estimation of useful secondary metabolites including carotenoids, flavonoids, ptheonolic cmompounds and dioscin contents in African yam tubers by FT-IR spectroscopy. The total carotenoids contents from 62 African yam tubers varied from 0.09 to 0.61mg/g dry weight (wt). The total flavonoids and phenolic compounds also varied from 41.9 to 171.0 mg/g and from 1.20 to 3.56 mg/g dry wt. FT-IR spectra confirmed typical spectral differences between the frequency regions of 1,700–1,500, 1,500–1,300 and 1,100-950 cm-1) , respectively. These spectral regions were reflecting the quantitative and qualitative variations of amide I, II from amino acids and proteins (1,700–1,500 cm-1) ), phosphodiester groups from nucleic acid and phospholipid (1,500–1,300 cm-1) and carbohydrate compounds (1,100-950 cm-1) ). Principal component analysis (PCA) and subsequent partial least square-discriminant analysis (PLS-DA) were able to discriminate 62 African yam tuber lines into discrete clusters corresponding to their chemotaxonomic relationship. The quantitative prediction modeling of total carotenoids, flavonoids, phenolic compounds and dioscin contents from African yam tuber lines were established using partial least square regression algorithm from FT-IR spectra. The regression coefficients (R2) ) between predicted values and estimated values of total carotenoids, flavonoids, phenolic compounds and dioscin were 0.83, 0.86, 0.72 and 0.7208, indicating that prediction model had higher or average accuracy. Since these secondary metabolites have diverse medicinal effects, these evaluation systems could be efficiently applied for metabolic estimation of African yam breeding lines. Thus, considering these results we suggest that FT-IR combined with multivariate analysis could be applied as a novel tool for metabolic evaluation and high-throughput screening of African yam lines with higher content of useful secondary metabolites. Furthermore, we also established the in vitro plant proliferation system from African yam tuber explants via tissue culture. The regenerated plants were successfully transferred to soil and grown to normal plant in the green house. These plant regeneration system of African yam tubers could be applied as an novel tool for mass proliferation of useful African tuber lines. In conclusion, we suggest that FT-IR combined with multivariate analysis and plant regeneration system of African yam could be applied for metabolic evaluation and screening of useful yam accessions with high throughput manner and agronomic applications.
