1. 젖산균 분리 및 수집
가. 원유에서 젖산균 분리
서울우유 관할 원유검사소(경기도 및 강원도 일부), 전북 축산시험연구소 및 경북 가축위생시험소에서 지원받아 1,222개의 균주를 분리하였으며, 발효유에 적합하기 위해서는 skim milk를 응고시킬 수 있는 산생성 능력이 있어야 하므로 10% 환원탈지분유에 분리균주를 접종한 후 37℃에서 18시간 및 24시간 배양하여 응고된 균주가 598개 균주이었다.
나. 신생아 분변에서 젖산균 분리

IV. 연구개발 결과
1. 젖산균 분리 및 수집
가. 원유에서 젖산균 분리
서울우유 관할 원유검사소(경기도 및 강원도 일부), 전북 축산시험연구소 및 경북 가축위생시험소에서 지원받아 1,222개의 균주를 분리하였으며, 발효유에 적합하기 위해서는 skim milk를 응고시킬 수 있는 산생성 능력이 있어야 하므로 10% 환원탈지분유에 분리균주를 접종한 후 37℃에서 18시간 및 24시간 배양하여 응고된 균주가 598개 균주이었다.
나. 신생아 분변에서 젖산균 분리
순천향병원 및 한강성심병원 산부인과 신생아실에서 지원받아 32명의 분변으로부터 분리하였다. bifidobacteria는 43개 균주를 수집, 보관하였고, 젖산균은 86개를 수집하였다.
다. 성인 분변에서 젖산균 분리
한국식품연구원 직원 및 가족에서 지원받아 47명의 분변으로부터 분리하였다. bifidobacteria는 52개 균주를 수집, 보관하였고, 젖산균은 187개를 수집하였다.
2. 비만억제능 균주 선발
원유에서 분리된 젖산균 중 10% 환원탈지유에 응고반응을 보인 598개와 분변에서 분리한 젖산균 273개, bifidobacteria균 95개를 대상으로 anti-lipase activity 실험결과 lipase 억제활성이 높은 균주 12종을 선발하여, 3T3-L1 cell을 이용한 Anti-adipogenic activity 실험을 실시하였다.
3. 선발 젖산균주 동정
Gram염색, 현미경 관찰, 포자생성, 호기적 및 혐기적 성장, catalase 생성, 15℃ 및 45℃에서의 생장, glucose로부터 가스생성, arginine으로부터 ammonia 생성 등의 시험, 당발효 시험, 16SrDNA sequencing 등을 분석한 결과 선발된 12종의 젖산균 중 Enterococcus 균주가 7종, Lactobacillus 균주가 2종, Weissella 균주가 3종으로 동정되었다.
4. 선발 젖산균의 특성조사
1차년도에서 비만억제능이 높은 균주 12종 중 산 생성 속도와 비만억제능을 감안하여 3종의 젖산균을 선발한 다음, 발효유의 종균으로서 적합한지의 여부를 실시하였다.
Weissella sp. F22와 L. plantarum Q180 균주는 40℃가, E. faecalis MD366 균주는 37℃가 적정온도로 나타났다. Weissella sp. F22 균주는 다른 2종의 균주에 비해 Streptomycin, Lincomycin에 대하여 내성이 강한 반면 Bacitracin에 대해서는 내성이 약한 것으로 나타났다. Enterococcus faecalis MD366 균주는 다른 2종의 균주에 비해 내성이 강한 것으로 나타났으며, 특히 Gentamycin, Kanamycin, Neomycin, Vancomycin에 대하여 내성이 강한 것으로 나타났다. Lactobacillus plantarum Q180 균주는 특히 Lincomycin과 Penicillin-G에 대해 내성이 약한 것으로 나타났다. 젖산균 3종 모두 Benzopyrene을 발암성 물질로 전환시키는 발암효소인 β-glucuronidase의 경우에는 효소활성이 없는 것으로 나타나 안전성을 확인할 수 있었으며, 또한 3종 모두 담즙에 대한 내성이 있었다. Weissella sp. F22 균주와 L. plantarum Q180 균주가 E. faecalis MD366 균주보다 pH에 대한 내성이 강한 것으로 나타났으며, E. faecalis MD366 균주가 다른 2종의 젖산균보다 항균력이 있는 것으로 나타났다.
5. 최적 배양기술 확립
비만억제능이 우수한 L. plantarum Q180 균주의 반응표면실험 결과 비만억제능 젖산균 최적배양조건은 환원탈지유 농도 9.54%, 배양온도 37.16℃, 배양시간 28.25h이었으며, 그에 따른 예상값은 pH 4.47, 55.55%의 anti-lipase activity, 20.48%의 anti-adipogenic activity로 나타났다.
6. 발효산물 분리동정
발효유로부터 ODS AQ colume, Vydac C18 column, Superdex peptide HR column을 이용하여 순차적으로 분리, 정제한 peptide의 N-말단으로부터 아미노산 배열은 Asp-Thr-Ile-Ser-Ala-Gln로 나타났으며, lipase 억제활성은 IC50이 2,817ug/mL로 나타났다.
7. 체내 균총 변화 및 지방 흡수 저해효과
가. 균총변화 추적 (pyroseguencing with 16S rRNA)
일반식이 대조군인 A군은 실험시작 전에 비해 6주 후 Bacteroidetes보다 Firmicutes가 증가하였고, 고지방식이 대조군인 B군은 Bacteroidetes가 약간 증가하였다. 상업균주 식이군인 C군은 실험시작 전에 비해 6주 후 Firmicutes보다 Bacteroidetes가 크게 증가하였다. L. plantarum Q180 식이군(108 CFU/day)인 D군과 L. plantarum Q180 식이군(109 CFU/day)인 E군은 다른 균주에 비해 가장 Bacteroidetes가 증가하였다. Weissella sp. F22 식이군인 F군은 Bacteroidetes가 약간 증가하였고, E. faecalis MD366식이군인 G군 역시 Bacteroidetes가 증가하였지만 크게 증가하지는 않았다. 일반식이 대조군 및 상업균주군에 비해 L. plantarum Q180 식이군이 비만억제효과가 있는 것으로 나타났다.
나. 체내 지방흡수 저해효과
비만 억제능이 우수한 균으로 선정된 Weissella sp. F22, E. faecalis MD366, L. plantarum Q180 균주의 체내 지방흡수 저해효과 실험을 실시하였다. 0시간의 Triglyceride(TG) 함량에 대한 시간 별 TG 함량의 비율을 나타낸 결과, L. plantarum Q180로 제조한 발효유를 투여한 D군이 3시간까지 0시간 대비 38.4%의 증가를 보이다가 5시간 후 0시간대보다 17.7% 감소를 나타내어 가장 우수한 지방흡수 저해효과를 갖는 것으로 나타났다.
다. 쥐 무게와 혈중 지질농도 변화
사육 6주 후 일반식이 대조군인 A군의 평균 무게는 26.30g, 고지방식이 대조군인 B군의 평균 무게는 28.84g, ABT-3 (109 CFU/day)균주를 경구투여한 C군은 28.74g, Lactobacillus plantarum Q180 (108 CFU/day)균주를 경구투여한 D군은 29.57g, Lactobacillus plantarum Q180 (109 CFU/day)균주를 경구투여한 E군은 28.29g, Weissella sp. F22 (109 CFU/day)균주를 경구투여한 F군은 28.44g, Enterococcus faecalis MD366 (109 CFU/day)균주를 경구투여한 G군은 27.95g으로 A군과 D군을 제외하고는 유의적인 차이가 없었지만(p<0.05), 고지방식이 대조군과 비교하여 E군은 13.70%, F군은 11.29%, G군은 18.10% 체중 증가량이 감소하였다. 부고환 지방무게의 경우 E군과 G군이 다른 군에 비해 적게 나타났으나 고지방식이 대조군과의 유의적 차이는 없었다. 간 효소치인 AST와 ALT는 각군의 값의 차이는 보여지나 유의적인 차이는 없는 것으로 나타났다. 혈청의 중성지방(TG)의 농도는 A군의 경우 81.00±12.77 mg/dL 이었고, 이에 비해 고지방식이대조군(B)은 110.00±21.34 mg/dL으로 정상식이군에 비해 증가하여 나타났다. C군과 D군, F군 그리고 G군은 고지방식이대조군과 비슷한 값을 나타내었으나, E군은 88.67±5.35mg/dL으로 정상식이군과 비슷한 수준을 나타내었다.
라. 부고환 지방조직 세포 크기
부고환 지방세포의 크기를 측정하여 지방세포 크기의 분포를 본 결과 B군을 제외한 모든 군에서 2,000m2 크기의 지방세포가 가장 많이 분포하였으며, 고지방식이 대조군인 B 군에서는 5,000m2 이상 크기의 큰 지방세포가 가장 많이 분포였다. H&E 염색액으로 염색된 지방세포를 현미경으로 관찰해 보았을 때 역시 육안으로도 지방세포의 크기가 차이나는 것을 알 수 있었다.
8. 최적 배합조성물 개발 및 상업적 이용 위한 젖산균 분말화
가. 제품 개발 및 이화학적 특성 조사
원유 80.682%와 탈지분유 3.318%를 첨가하고 65℃에서 배합하여 완전히 녹인후 90℃에서 5분간 살균하였으며, 40℃로 냉각시킨 다음 상업균주인 ABT-3 균주를 0.1% 접종하고, 최종 pH 4.4으로 감소할 때까지(40℃, 5시간) 배양한 액에 Lactobacillus plantarum Q180 균주(108 CFU/mL)를 혼합한 후 딸기잼 또는 블루베리잼과 올리고당 5%, 액상과당 2%를 첨가하고 교반하여 호상발효유를 제조하였다. 딸기쨈 4%를 첨가한 발효유(A), 6%를 첨가한 발효유(B), 8%를 첨가한 발효유(C), 블루베리쨈 4%를 첨가한 발효유(D), 6%를 첨가한 발효유(E), 8%를 첨가한 발효유 (F)의 이화학적 성분을 조사한 결과 pH는 4.3이었으며 고형분함량은 21.46~24.65%, 젖산균수는 7.33×109~4.66×109 CFU/mL, 적정산도는 0.95~1.05%이었다.
나. 최적 배합조성물 개발
개발제품의 배합비를 보면, 원유 80.682%, 탈지분유 3.318%, 올리고당 5%, 액상과당 2%, 블루베리잼과 8% 정제수 1%이었다
다. 젖산균 동결건조 분말화
1) Lactobacillus plantarum Q180 배양조건
L. plantarum Q180을 70 L Bio-fermentor 시스템에 대량 배양하는 조건으로 MRS 생장배지 30리터에 0.1%량의 L. plantarum Q180 배양액을 접종하여 38℃의 온도에서 배양한 결과, 1.48 × 1010/mL의 생균을 얻을 수 있었다. L. plantarum Q180의 생장데이터를 분석한 결과 1개 세포의 중량은 6.65 x 10-8 g/cell 이었으며, 1g당 생균수는 8.09249E+12, 비증식속도는 0.0366/min 이며, 배양조건하에서 세대시간(generation time)은 18.9515분, 생장유도기(lag time)는 21.65시간이었다. L. plantarum Q180 MRS 배양액을 3,500rpm으로 원심분리하여 총 487g의 wet cell을 회수하였으며, cell 회수율은 0.974% 이었으며, wet cell의 수분함량은 73.3%, 생균수는 2.24 × 1011/mL 이었다.
2) 동결건조 스타터 개발
Cell pellet의 동결상해를 줄이기 위하여 cryogenic agents로서 특수조제 분말 (SIMP)을 첨가하여 혼합한 후, 저온동결기 (Freezer, Operon 사, 한국)에서 동결 온도 –27℃로 동결시켰다. 동결된 L. plantarum Q180의 cell을 진공건조기 (수분증발용량 50kg, Operon 사, 한국)에서 냉열기 온도 –50℃, 냉열판 온도 –25℃, 4,500 mtorr의 진공조건 하에서 24시간 동결 건조한 결과, 수분함량 5.1%의 스타터 건조 분말(cell pellet) 1.75g을 생산할 수 있었다. Cell pellet의 동결상해를 줄이기 위한 cryogenic agents로서 SIMP를 첨가한 cell pellet의 결과, 동결건조에 의한 세포 생존률은 99% 이상인 것으로 나타났다.
3) 요구르트 배양 및 저장 중 변화
상업용 DVS 형 요구르트 스타터인 ABT-4, L. plantarum Q180 스타터를 각각 0.1% 단독 또는 두 균주를 0.05%씩 혼합 접종하여 요구르트 발효 베이스를 제조한 결과, 최종 생균수에 도달하는 시간에 차이는 있었으나, 6시간 경과 시 ABT-4는 생균수가 최대에 이른 후 감소하기 시작한 반면 L. plabtarum Q180과 혼합 또는 단독 접종한 요구르트의 배양 중 생균수는 15시간까지 계속 증가하였다. 15시간 후 배양을 종료한 3개 처리구(ABT, ABT+LPL, LPL)의 요구르트를 2℃로 저장하여 12시간 및 24시간 경과 후 생균수를 측정한 결과, 24시간 경과시ABT-4 는 생균수가 감소한 반면 L. plabtarum Q180(LPL)과 혼합 또는 단독으로 배양한 요구르트의 배양 중 생균수는 계속 유지하였다.
9. 비만억제 작용기작 규명
Western blotting 방법을 이용하여 지방산 산화에 관여하는 PPARγ, C/EBPα 의 mRNA 발현차이를 비교한 결과 3T3-L1 세포의 지방분해시에 L. plantarum Q180(108 CFU/g) 균주를 400 ug/mL 농도로 처리한 세포에서 무처리구 대비 PPARγ의 발현은 46.07%, C/EBPα의 발현은 35.16% 감소하였다.
10. 동물모델을 이용한 발효유의 비만억제 효능평가
가. 실험동물의 식이 효율 및 체중 변화
실험동물을 군 당 6마리씩 일반식이 대조군(A), 생리식염수를 경구투여한 고지방식이 대조군(B), 상업균주인 ABT-3(CHR HANSEN)균주와 Lactobacillus plantarum Q180 균주를 혼합배양한 발효유를 경구투여한 군(C), ABT-3균주와 Lactobacillus plantarum Q180 균주를 혼합배양하고 가르시니아 캄보지아를 첨가한 발효유를 경구투여한 군(D), Lactobacillus plantarum Q180을 이용한 발효유를 경구투여한 군(E), Lactobacillus plantarum Q180로 배양하고 가르시니아 캄보지아를 첨가한 발효유를 경구투여한 군(F) 등 6군으로 나누고 8주간 사육한 결과 고지방식이 대조군인 B군의 체중 증가량이 293.46g으로 가장 많은 증가를 보였으며 E군, C군, D군, F군, A군 순으로 체중 증가량이 감소하였다. C군은 고지방식이군 대비 체중이 5.14% 감소하였으며, D군은 6.5%, E군은 3.36%, F군은 10.81% 감소하여 L. plantarum Q180 균주로 배양하고 가르시니아 캄보지아를 첨가한 발효유를 경구투여한 군에서 체중 감소율이 가장 뛰어났다.
나. 혈액분석
혈액 분석 결과 ALT수치는 각 군간의 값의 차이는 보여지나 유의적 차이는 없었으며 AST 수치는 C군에서 가장 작은 수치를 나타내었다. 혈청의 중성지방 (TG)의 농도의 경우 정상식이군과 프로바이오틱스 급여군에서 모두 고지방식이 대조군인 B군에 비해 유의적으로 적은 수치를 나타내었으며, 아디포넥틴의 경우 각 군간의 유의차를 보이지 않았고, 렙틴의 경우 고지방식이 대조군인 B군에서 가장 많이 증가하였으며 프로바이오틱스 급여 군은 정상식이 대조군인 A군보다는 증가하였지만 B군 대비 유의적으로 감소함을 보였다.

1) Isolation of lactic acid bacteria from raw milk
Raw milk was collected from several farms under support of Raw milk testing lab within the jurisdiction of Central, North, Western guidance division (part of Kyoung Gi Do and Gang Won Do)in Seoul Dairy Cooperation, Provincial institute for livestock promotion(Jeonbuk, Kyoungbuk, Jeju). 1,222 bacteria were isolated from modified MRS media. 598 bacteria were further isolated after the microbes were inoculated on 10% the reconstituted skim milk to check acid production ability which can coagulate skim milk followed by incubation at 37℃ for 18 and 24hr.
2) Isolation of lactic acid bacteria from infant feces
32 new-born infant feces were collected under support of Soon Chun Hyang Hospital and Hangang Sungshim Hospital. 43 bifidobacteria and 86 lactic acid bacteria were isolated after incubation at 37℃ for 18 and 24hr.
3) Isolation of lactic acid bacteria from adults feces
47 adult feces were collected from staffs and their family members of KFRI. 52 bifidobacteria and 187 lactic acid bacteria were isolated.
2. Collection of lactic acid bacteria with anti-obesity ability
Lactic acid bacteria isolated from raw milk were further isolated after the microbes were inoculated on 10% the reconstituted skim milk followed by incubation at 37℃ for 18 h and 24 h, respectively. 598 strains were selected from them. Anti-lipase activities of 598 lactic acid bacteria isolated from raw milk and 273 lactic acid bacteria and 95 bifibacteria were tested. Based on the anti-lipase activity experiment’s result, 12 strains with strong anti-lipase activity were investigated anti-adipogenic activity.
3. Identification of selected lactic acid bacteria
7 Enterococcus sp, 2 Lactobacillus sp and 3 Weissella sp was identified by experiment of gram staining, fractography, spore creation, aerobic and anaerobic growth, catalase creation, growth at 15℃ and 45℃, gas formation from glucose, ammonia formation from arginine, glucose fermentation and 16S rDNA sequencing from 12 selected lactic acid bacteria.
4. Characteristics of selected lactic acid bacteria
Sitability as starter culture for the fermented milk production has surveyed through the anti-obesity ability and acid formation speed of 3 selected lactic acid bacteria from 12 strains which have strong anti-obesity ability. The optimum temperature of Weissela sp. F22 and L. plantarum Q180’s was 40℃ and E. faecalis MD366’s optimum temperature was 37℃. Weissella sp. F22 was weaker resistant to the Bacitracin, but it was stronger resistant to the Streptomycin, Lincomycin than L. plantarum Q180 and E. faecalis MD366. Enterococcus faecalis MD366 has shown stronger resistance, especially Gentamycin, Kanamycin, Neomycin and Vancomycin than Weissela sp. F22 and L. plantarum Q180. Lactobacillus plantarum Q180 has shown weak resistant to the Lincomycin and Penicillin-G. Weissela sp. F22, L. plantarum Q180 and E. faecalis MD366 could confirm safety through no β-glucuronidase activity that Benzopyrene is converted to carcinogenic substance and has bile tolerance. Weissela sp. F22 and L. plantarum Q180 had stronger pH tolerance than E. faecalis MD366. E. faecalis MD366 shown stronger antibacterial activity than Weissela sp. F22 and L. plantarum Q180.
5. Establishment of optimum culture technology
The optimum culture condition of anti-obesity lactic acid bacteria was 9.54% skim milk, 37.16℃ culture temperature and 28.25 h culture time through response surface experiment’s result of L. plantarum Q180 which has strong anti-obesity ability. According to response surface experiment’s predictive value was pH 4.47, anti-lipase activity 55.55% and 20.48% anti-adipogenic activity.
6. Separation identification of fermented product
Amino acid sequence from N terminus of gradually separated and refined peptide by using ODS AQ colum, Vydac C18 column and Superdex peptide HR column from fermented milk was Asp-Thr-Ile-Ser-Ala-Gln and anti-lipase activity (IC₅₀) was 2,817 μg/mL.
7. Contrification of selected lactic acid bacteria for commercial exploitation and development of optimum mix formula
1) Tracking of variation of intestinal microbial flora (pyrosequencing with 16srRNA)
After 6 weeks, A group, control group fed with general diet, increased Firmicutes more than Bacteroidetes and B group, control group fed with high-fat diet, more increased before starting experiment. Also, C group, fed with commercial strain diet, increased Bacteroidetes more than Firmicutes before starting experiment. D group, fed with L. plantarum Q180 (10⁸ CFU/day), and E group, fed with L. plantarum Q180 (10⁹ CFU/day), increased Bacteroidetes more than groups fed with another strains. F group, fed with Weissella sp diet, slightly increased Bacteroidetes. G gorup, fed with E. faecalis MD366, also slightly increased Bacteroidetes. Therefore, D group, fed with L. plantarum Q180 (10⁸ CFU/day), and E group, fed with L. plantarum Q180 (10⁹ CFU/day), more effected anti-obesity than control group fed with general diet and group fed with commercial strain diet.
2) Absorption inhibition effect of internal deposit fat
It has implemented that absolution inhibition effect of internal deposit fat with L.plantarum Q180, E. faecalis MD366 and Weissella sp. selected lactic acid bacteria showing strong anti-obesity. The results showed Triglyceride (TG) content ratio classified by time about TG content on 0 h. Consequently, D group, fed with fermented milk with L. plantarum Q180, showed the strongest absorption inhibition effect of internal deposit fat as D group increased 34.8% TG content until 3 h and decreased 17.7% TG content after 5 h compared to 0h.
3) Variation of body weight and blood lipid concentration in rat
After rearing 6weeks, A, B, C, D, E, F, G group's average body weight was 26.30, 28.84, 28.74, 29.57, 28.29, 28.44, 27.95g. These groups were no significant difference except A and D group (p<0.05). However, compared with group B, weight increase amount in group E, F and G was respectively decreased 13.70%, 11.29% and 18.10%. In case of epididymal weight, E and G group had less weight compared to other groups but no significant difference with control group fed with high fat diet. The AST and ALT had difference each group but no significant difference. A group's TG concentration of serum was 81.00±12.77 mg/dL. On the contrary, B group was 110.00±21.34 mg/dL increased compared to control group. The C, D, F and G roup was similar to group fed with high fat diet but E group was similar to control group.
4) Variation of epididymal adipocyte size
The result of adipocyte size's distribution to measure epididymal adipocyte size was much spread 2,000μm² of adipocyte size on all groups and B group was much spread more than 5,000μm² of adipocyte size. When observing dyed adipocyte size with H&E dye solution through microscope, it showed difference of adipocyte size with the naked eye.
8. Contrification of selected lactic acid bacteria for the commercialization and development of optimum mix formula
1) Investigation of physiochemical property
80.682% of raw milk and 3.318% of skim milk was mixed and dissolved at 65℃, and pasteurized at 90℃ for 5 minutes and controlled to 40℃ and inoculated ABT-3 strain as commercial strain and mixed L. plantarum Q180 (10⁸ CFU/mL) in culture solution until pH up to 4.4, followed by the addition and agitation of strawberry or blueberry jam, 5% of oligosaccharide and 2% of liquid fructose. The result of physiochemical property of the fermented milk added 4% of strawberry jam (A), the fermented milk added 6% of strawberry jam (B), the fermented milk added 8% of strawberry jam (C), the fermented milk added 4% of blueberry jam (D), the fermented milk added 6% of blueberry jam (E), the fermented milk added 8% of blueberry jam (F) was 4.3 of pH, 21.46~24.65% of solid content, 7.33×10⁹~4.66×10⁹ CFU/mL and 0.95~1.05% optimal acidity.
2) Development of optimum mix formula
Compounding ratio of development product was 80.682% of raw milk, 3.318% of skim milk, 5% of oligosaccharide, 2% liquid fructose, 8% of blueberry jam and 1% of purified water.
3) Confrication of selected lactic acid bacteria through freeze drying
a. Fermentation condition of Lactobacillus plantarum Q180
On the condition of mass culture in 70 L Bio-fermentor system, L. plantarum Q180 culture was inoculated in the 30 L of MRS growth medium and incubated at 38℃. 1.48 × 1010/mL of viable bacteria was obtained. Growth data of L. plantarum Q180 was analyzed. The weight of one cell was 6.65 x 10-8 g/cell, and the number of viable bacteria per 1 g was 8.09249E+12. Specific growth rate was 0.0366/min, and generation time was 18.9515 minutes under culture condition. Moreover, the growth lag time was 21.65 hours. b. Development of freeze-drying starter
To reduce the freezing injury of cell pellet, especially prepared powder (SIMP) was added as the cryogenic agents. It was frozen in the low temperature freezer (Freezer, Operon Inc., Korea) at -27℃. The Freezed cell of L. plantarum Q180 was freeze-dried in vacuum drier (50kg of water evaporative capacity, Operon Inc., Korea) at –50℃ of chiller and –25℃ of the chiller plated cell, under 4,500 mtorr of vacuum condition, for 24 h. 1.75 g of dried powder starter was produced and water content of the stater was 5.1%. Cell survival rate of the cell pellet added SIMP, as cryogenic agents for reducing freezing in jury, was showed over 99%.
c. Fermentation of yogurt and change during storage
Three yogurt fermentation bases were produced using ABT-4 starter, commercial DVS-type yogurt stater, and L. plantarum Q180 starter. 0.1% of ABT-4 or L. plantarum Q180 was inoculated and 0.05% of ABT-4 and L. plantarum Q180 were inoculated. Among the three yogurt bases, there was no difference in time to reach the final number of bacteria. However, the number of viable bacteria in yogurt base produced by ABT-4 was started to reduced after 6 h. The number of viable bacteria in yogurt bases produced by L. plantarum Q180 or mixed culture was continually increased until 15 h. After 15 h, the three yogurt (ABT, ABT+LPL, LPL) terminated the fermentation were stored at 2℃ for 12 h or 24 h. As the results of count of the viable bacteria, the number of viable bacteria of the yogurt fermented by ABT-4 was decreased. However, it was maintained in the yogurt fermented by L. plantarum Q180 or mixed culture.

To understand the body weight gain of 6 kinds of treatment was tried to the Spraue-Dawley rat. The treatment was control group fed with normal diet(A), control group fed with high-fat diet administered orally physiological saline(B), a group fed with fermented milk made with L. palnatarum Q180 and ABT-3(CHR HANSEN)(C), a group fed with fermented milk made with L. palnatarum Q180 and ABT-3(CHR HANSEN) added garsinia canbogia (D), a group fed with fermented milk made with L. palnatarum Q180(E) and a group fed with fermented milk made with L. palnatarum Q180 added garsinia canbogia(F). After raising 8 weeks, B group, control group fed with high-fat diet administered orally physiological saline, increased 293.46 of body weight gain and decreased in order of E, C, D, F, A group. Compared with group B, weight increase amount in group C, D, E, and F was respectively decreased 5.14%, 6.5%, 3.36%, and 10.81%. Weight reduction ratio is the highest in group F, the rat fed with fermented milk made with L. palnatarum Q180 added garsinia canbogia.
2) Blood analysis
In case of ALT, there was no significant difference but showed different values of each group. In case of AST, there was the lowest in C group. In the concentration of blood serum TG, a group fed with normal diet and a group fed with probiotics showed the lowest value compared to B group. In case of adiponectin, there was no significant difference. Leptin was the biggest increase in B group fed with high-fat diet. In case of group fed with probiotics, it was increased compared to A group but decreased compared to B group.
