펄스 전기장 기술의 산업화 적용을 위한 품질기준 마련과 공정 연구를 통해 우유 살균의 산업화 가능성을 확인하였다. 우유의 PEF 살균의 국내외 현황을 검토하고 미국이나 유럽의 주스살균 제품의 유통을 확인하여 우유의 살균 가능성을 확인하였다. 우유의 살균을 위한 최적의 PEF 조건을 설정하기 위해 기기 및 공정 연구를 수행하였으며, 그에 따른 실험실적 기기와 공정을 완성하였다. 제품의 관리 규격 기준 및 규격 표준화를 위해 축산물 위생관리기준 등을 활용한 성분규격 기준을 마련하였고, 우유 가열에 의한 변성 물질 및 변성도에 대한 표준화를 마련하였다. 주요 타깃 미생물은 일반세균, 대장균군, 진균으로 선정하였으며, 기능성 단백질은 유청 단백질, 카제인, 면역 단백질 등을 선정하여 각 규격에 대한 평가법을 확립하였다. 초기 우유 살균 연구에서 PEF 조건은 20 L/h의 유속으로 70% of out voltage, 25 us of pulse width에서 처리하여 최종 우유의 온도가 PEF 처리를 통해 50, 65℃로 처리되는 공정을 세팅하였다. 살균 제어 및 영양원 보존을 위한 최적 공정 설계를 통해 공정 최적화 및 단계별 scale up 연구를 진행하였고, 상품화를 위한 연구를 위탁기관과 공동연구로 수행하였다. 공정을 최적화하기 위해 표준용액을 이용하여 연속식 처리를 10 kV/cm of Field strength에서 100, 150, 200, 250 kJ/L of energy의 PEF 공정변수를 주어 최적의 공정변수를 찾고자하였다. 위에서 설정한 공정변수에 기존의 가열살균공정(LTLT, HTST)을 적용하여 시판하는 저지방우유에 타깃 미생물 4종(S. cerevisiae, E. coli, L. brevis 및 L. plantarum)을 접종 후 적용하는 연구와 원유에 적용하는 연구를 통해 단백질 보존과 미생물의 저감을 위한 최적의 공정으로 가열살균(LTLT, HTST)과 PEF 조건을 200, 250 kJ/L로 하여 병용하는 공정으로 설정하였다. 마련된 최적의 PEF 및 가열살균공정에 대해 시생산을 통한 상업화 및 법제화 추진을 위해 시생산 공정 라인적용 연구를 실시하였다. 선정된 가열살균공정은 HTST였으며, 이에 따른 PEF의 시생산 최적의 공정은 100 L/h of flow rate, 16 kV/cm of field strength 및 250 kJ/L of energy로 처리하는 공정이었다. 원유를 위 공정과 같이 처리 시 4℃의 원유가 승온을 통해 30℃로 상승하고 PEF 처리를 통해 75℃ 까지 상승한다. 그 후 HTST 처리 후 냉각 및 포장이 되어 냉장 보관 상태로 저장성 실험결과 일반 HTST 처리 우유에 비해 미생물의 저장성은 더 우수하며, 기능성 단백질의 보존은 HTST 처리 우유와 유의적인 차이가 없었음을 확인할 수 있었다. 또 유제품으로 저지방우유, 초코우유 및 아메리카노의 시생산 공정라인의 최적화 공정을 마련하였다.
펄스전기장 기술을 우유 및 유제품의 살균에 적용하기 위한 기초 및 응용연구를 수행하였다. 펄스전기장 처리의 변수로는 Electric field strength (kV/cm), Pulse width (μs), Pulse frequency (Hz), Treatment time (s) 가 있으며, 이들 변수의 조합을 통해 다양한 펄스 처리가 가능하다. 펄스처리가 우유의 부패 미생물에 미치는 영향을 확인하기 위해 Saccharomyces cerevisiae, Escherichia coli , Lactobacillus brevis, Lactobacillus plantarum을 모델 균주로 선정하고 Batch chamber 에서 펄스전기장 조건에 따른 사멸특성을 확인하였다. S. cerevisiae 와 Lactobacillus 의 경우, 펄스전기장 처리에 의해 보다 쉽게 사멸되어 10kV/cm 이상의 전기장에서 빠르게 사멸 되는 것이 확인되었으나, E.coli 의 사멸에는 경우 15 kV/cm 이상의 전기장 조건이 요구되었다. Batch 조건에서 확인된 펄스처리 기술의 산업화 적용을 위해 연속식 처리가 가능한 파일럿 스케일의 실험을 수행하였다. 펄스처리 파일럿 장치는 30 L/h 의 유속을 처리할 수 있도록 구성되었으며 온도조절 장치를 통해 펄스처리 초기온도의 설정이 가능하였다. 파일럿 처리 장치는 Batch 처리장치에 비해 상대적으로 낮은 전기장 세기인 10 kV/cm 를 사용하였으나 pulse frequency의 증가를 통해 시료에 펄스 에너지를 250 kJ/L 까지 가해줄 수 있었다. 저지방 우유에 모델 균주인 Saccharomyces cerevisiae, Escherichia coli , Lactobacillus brevis, Lactobacillus plantarum 를 접종하고 연속식 펄스처리를 수행하였을 경우 150 kJ/L 이상에서 모델 균주의 사멸이 관찰되었으며 200 kJ/L 이상의 에너지 수준에서는 모든 균주의 생균수가 4 decimal reductions 이상 감소하였으며, 4°C에서 15일간 저장 시 균수의 증가도 억제된 채로 유지되어 처리초기온도 30°C, 펄스에너지 200 kJ/L 의 조건이 펄스전기장을 이용한 저지방 우유 살균조건으로 제시되었다. 제시된 펄스 처리조건을 활용하여 커피우유, 딸기우유, 바나나우유 등의 가공유 제품살균에 펄스전기장 처리를 적용하였다. 펄스전기장 적용 시 호기성 미생물은 최대 1.0 decimal reduction, Yeast 의 경우 최대 3.0 decimal reductions 수준의 감균 효과를 나타냈다. 감균 효과가 미생물 접종 후 처리한 이전 실험에 비해 상대적으로 낮은 이유는 이들 가공유의 원부재료인 탈지분유와 커피, 과일 농축액 제조 공정이 높은 열에너지를 동반하는 공정으로 구성되어 자연적으로 존재하는 미생물 중 내열성이 강한 부분이 살아남아 과점하고 있기 때문으로 판단되었다. 펄스전기장 처리에 따른 가공유 제품의 물리, 화학, 향기성분은 일정하게 유지되었다.
비가열 살균법인 펄스 전기장 기술을 활용한 우유 내의 기능성 단백질의 보존성 확보 기술을 구축하기 위하여 살균 공정 별로 우유를 처리하였을 때 나타나는 우유 단백질의 변화를 파악하고 분석하였고, 기존의 가열 살균과 PEF를 병용처리 하였을 때 우유 내의 기능성 단백질의 보존성을 연구하였다. 원유, 탈지유 HTST 우유 및 LTLT 우유를 PEF 처리한 후 우유 중에 존재하는 단백질의 변성 여부를 DSC, SDS-PAGE 및 HPLC를 이용하여 조사한 결과 PEF의 처리 조건(50℃, 65℃)에 관계없이 PEF 처리가 우유 중에 존재하는 단백질을 변성시키지 않았다. 또한 살균 공정별로 PEF 처리한 우유의 탁도시험에 의해서도 PEF 처리가 우유 단백질을 변성시키지 않았고, pH, 산도, alcohol test, 색도 및 갈색화 반응을 분석한 결과 LTLT나 HTST 및 UHT 처리와 비교할 때 PEF 처리가 우유의 물리화학적 성질을 크게 변화시키지 않았다. PEF의 에너지가 증가할수록 IgG, IgA, β-lactoglobulin, lactoferrin의 농도는 유의적으로 감소하였으나, α-lactalbumin이나 TGF-β1의 감소는 유의적이지 않았다. PEF와 LTLT를 병용처리한 우유를 16일간 저장하였을 때, IgG, β-lactoglobulin, lactoferrin, α-lactalbumin은 각각의 농도는 저장기간에 따라 크게 영향을 주지 않는 범위 내에서 감소하였으나, IgA만이 PEF 에너지의 양에 관계없이 그 농도가 유의적으로 감소하였다. 같은 가열처리 방식이라도 면역단백질의 농도에 있어서 장시간 가열하는 LTLT 살균방식이 단시간 가열 살균인 HTST보다 면역단백질의 손상에 영향을 끼치는 것으로 판단할 수 있었다. PEF 처리 시, 여러 가지 생리활성이 높은 lactoferrin은 기존의 살균방법보다 면역활성이 높았고, α-lactalbumin에 큰 영향을 끼치지 않는 것으로 확인하였다. 저장 중 원유의 면역단백질의 농도는 처음 1주일에 1차로 급격한 감소율을 보였고, 1개월이 경과한 후 2차로 급격한 감소율을 보였다. 반면에 PEF를 처리한 시료나 기존 살균방법의 단독처리나 PEF 병용처리 시료의 경우 점진적으로 면역단백질의 농도가 감소하였고, 1개월이 경과한 후는 원유를 비롯한 다른 시료간의 차이가 크지 않았다. PEF 처리에 따른 우유 내의 면역단백질은 기존의 살균방법인 LTLT, HTST를 병용처리하였을 때 그 효과가 초기에는 PEF를 단독으로 처리한 우유보다 좋지 않았으나, 기존의 살균방법과 병용처리한 우유가 단백질의 보존성에 있어 더 좋은 결과를 얻을 수 있었다.
우유는 단백질, 탄수화물, 비타민, 무기질 등을 함유하고 있어 미생물이 성장할 수 있는 이상적인 조건을 가지고 있으며 저장성을 향상시키고 우유의 변질을 초래하는 효소를 파괴하기 위해 열처리방식의 살균공정을 실시하고 있으나 열에 의해 발생되는 영양성분의 파괴, 향미의 손실, 색택의 변화, 조직감의 변화가 발생하며 이러한 문제를 해결하기 위하여 저온살균공법(LTLT) 및 감마선 조사 등 우유에 열을 적게 가하는 방법을 이용한 살균법에 관한 시도가 이루어져 왔으나, 아직까지는 기존의 살균 방법인 UHT를 대체할 수 있는 효과적인 저온 살균법에 관한 뚜렷한 해결책은 찾지 못하고 있다. 이에 한 가지 대안으로 PEF 비가열적 살균방법을 사용하여 3종의 시작품을 제작하여 고전압 처리할 경우 온도가 거의 상승하지 않으며, 처리시간이 짧고 대부분 연속처리가 가능하며, 처리 후에도 식품의 물리적, 화학적 및 영양학적 특성들을 거의 변화시키지 않는 것을 확인하였다.

Application of pulsed electric field technology(PEF) industrialization confirmed the possibility of milk sterilization, through the quality standards established for the industrialization process research. The potential of PEF sterilization of milk was confirmed through reviewing the status of the distributed juice pasteurized products of US or Europe. The apparatus and process study was carried out to establish the optimum conditions for the sterilization of milk PEF, which resulted in the completion of the process and laboratory apparatus accordingly. For standardized management specification standards and specifications of the product, standard for denatured and denatured material also by heating milk were prepared. The main target microorganisms were selected for bacteria, coliform bacteria, and fungi, the functional protein wass whey protein, casein, and immune protein, and such as an evaluation method was established for the respective standards. Initial studies in the sterilization of milk by PEF treatment conditions in 70% of out voltage, 25 us of pulse width at a flow rate of 20 L / h was set to process the temperature of the milk to be processed by the end 50, 65 ℃ through the PEF treatment. The scale up research through the optimal process design for insect control, and nutrient conservation, research institutions was proceeding step-by-step process optimization with collaboration. In order to optimize the continuous process, using the standard solution, in 10 kV/cm of field strength, in 100, 150, 200, 250 kJ/L of energy of given process parameters, the PEF was to find the optimal process parameters. By heat sterilization processes in process parameters set above (LTLT, HTST) low-fat milk, innoculated by four kinds of target microorganisms (S. cerevisiae, E. coli, L. brevis and L. plantarum) , the effect after vaccination optimal process for protein and reduction of microorganisms through applying the heat sterilization (LTLT, HTST) and PEF 200, to 250 kJ / L, were set by the process used in combination. For legislation promoting the commercialization, prototype prodruction were applied with the optimum PEF and heat sterilization processes. As the results, heat sterilization process is selected HTST, thus the process of the optimum PEF prototype prodruction was 100 L / h of flow rate, 16 kV / cm of field strength, and 250 kJ / L of the step of treatment with energy. The temperature of milk was raised to 30 ℃ of 4 ℃ during processing as shown in the above process, and raised to 75 ℃ through the PEF treatment. After HTST treatment then cooled and packaged sample compared to the results in general HTST treatment of milk was better in cold storage, and in the view of preservation of functional protein, the HTST treatment of milk and HTST-PEF milk were no significant difference . In addition to low-fat dairy milk, the optimization process of prototype prodruction line of chocolate milk and Americano were prepared.
Pulsed electric field (PEF) technolgy was applied to pasteurize milk and dairy products. The processing parameters of PEF were electric field strength (kV/cm), pulse width (μs), pulse frequency (Hz), and treatment time (s). Saccharomyces cerevisiae, Escherichia coli, Lactobacillus brevis, Lactobacillus plantarum were selected as test microorganisms for PEF treatment. In batch type experiments, Saccharomyces cerevisiae and Lactobacillus were more susceptible to PEF treatments, and inactivation of them were initiated by the electric field strength of 10 kV/cm. Escherichia coli required 15 kV/cm for inactivation. The microbial inactivation by PEF treatment was confirmed by continuous, pilot scale experiments. The PEF pilot could process sample flow of 30 L/h. Low fat milk was innoculated with test microorganisms and PEF was applied to the samples with different energy level. The total energy inputs were from 100 to 250 kJ/L. The microbial inactivation was increased as the increase of energy inputs. We suggest the PEF energy of 200 kJ/L with inlet temperature of 30°C as industrial processing condition. The suggested processing condition reduced the microbial counts of test microorganisms more than 4 decimal reductions. The suggested PEF processing condition was applied to 3 different dairy products such as coffee milk, strawberry milk, and banana milk. PEF treatments showed restricted effects against normal flora of aerobic counts, but normal flora of yeasts were reduced by 3 decimal reductions. The application PEF treatments on dairy products results in little changes in color, pH, particle size distribution, and flavor profiles.
To construct the preservative technology of functional proteins in bovine milk using pulsed electric fields (PEF) technology as a non-thermal sterilization, the changes of bovine milk proteins were examined when bovine milk was heat-sterilized, and the preservation of functional proteins of bovine milk was studied when the milk was treated with the conventional thermal sterilization process in combination with PEF. When the denaturation of proteins in PEF-treated raw milk, skim milk, HTST- and LTLT-milk were examined by SDS-PAGE, differential scanning calorimetry (DSC) and HPLC, bovine milk proteins did not denature regardless of PEF's treatment condition (50 and 60℃). PEF treatment did not denature milk proteins in PEF-treated-milk at different sterilization process and physicochemical properties of milk proteins were not considerably changed compared with LTLT-, HTST- or UHT-sterilization when turbidity, pH, titratable acidity, alcohol test, and browning test were carried out. As an energy of PEF treatment increased, the amounts of IgG, IgA, β-lactoglobulin and lactoferrin significantly decreased, but the amounts of α-lactalbumin or TGF-β1 were not significantly decreased. When PEF and LTLT combination milk was stored at 16 days, the amounts of IgG, β-lactoglobulin, lactoferrin and α-lactalbumin decreased a little during the storage period. However the amount of only IgA significantly decreased regardless of an energy of PEF treatment. The damage of immunoproteins was affected by the thermal sterilization process with long time, a LTLT, more than the thermal sterilization with short time, a HTST. When milk was treated with PEF, lactoferrin which has various multifunctional activity retained higher immune-activity than when treated with the conventional sterilization methods. Also PEF treatement did not effect on the immune-activity of α-lactalbumin. The amounts of immunoproteins of raw milk rapidly decreased at the first 1 week, and secondly these rapidly decreased over a month. On the other hand, the amounts of immunoproteins of PEF-treated-milk, only treatment of conventional sterilization or PEF combined with the conventional sterilization methods milk gradually decreased and there were little difference between the other milks, including raw milk, after a month. These results showed that the effect of PEF combined with the LTLT- or HTST-milk was worse than only PEF treatment milk, but the effect of preservation of immunoproteins, the conventional sterilization combined with the PEF treatment milk was better.
Milk contain protein , carbohydrate , vitamins, minerals and a lot of minerals, but it is vulnerable to heat damage during pasteurization processes. The pasteurization or sterilization process can destroy the enzyme which results in the deterioration of milk such as destruction of nutrients, loss of flavor, change of colors and texture change. Various means have been used to extend the shelf life of dairy products such as high-pressure processing, irradiation, ohmic heating and pulsed electric field(PEF) technologies. of these technics, We have prepared three kinds of prototypes using PEF in the high voltage and very short time treatment conditions. and very reasonable results were obtained. PEF technologies will be potential alternatives to traditional thermal milk pasteurization, owing to their advantages in minimizing sensory, physico-chemical and nutritional damage.
