○ 돼지 심장 특이적 프로모터 개발 및 기능 분석

Ⅳ. 연구개발결과
○ 돼지 심장 특이적 프로모터 개발 및 기능 분석
- 심장에서 특이적으로 발현되도록 하는 프로모터의 발굴은 심장에서의 유전자 기능을 분석하는데 매우 중요한 역할을 한다. 본 연구를 통해서 심장에서 특이적으로 발현하는 유전자를 발굴하여 그 유전자의 프로모터 영역을 분석한 다음 정상적으로 기능을 하는지 확인하였다.Gene Expression Omnibus (GEO) 프로그램을 통해 마우스와 사람의 심장에서 특이적으로 발현하는 것으로 확인된 여러 유전자들 중에서 다음의 MYH6, TNNI3 및 MYBPC3를 결정하였다.
- 여러 조직에서 RT-PCR을 통해 MYH6, TNNI3 및 MYBPC3 유전자의 심장 특이적 발현을 분석한 결과 마우스와 사람과 마친가지로 돼지에서도 심장에서만 특이적으로 발현하는 것으로 확인되었다.
- 유전체정보학(bioinformatics) 분석을 통해 예측한 결과 이들 유전자들의 프로모터 염기서열에서 여러 접합 부위(binding sites)를 포함하고 있는 심장발달관련 전사요소(transcription factor) 들이 있다는 것을 확인하였다.
- 이들 유전자들의 프로모터 염기서열을 분석한 결과 사람과 돼지 간에 높은 보존 영역(conserved region)이 존재한다는 것을 확인하였다.
○ 사람 면역 관련 유전자 과발현 벡터 개발 및 형질전환 체세포주 개발
- 프로모터의 기능이 정상적으로 작동하는지 확인하기 위하여 체세포에 선별마커인 green fluorescence protein (GFP)를 포함시킨 벡터를 형질전환시켜 발현을 분석한 결과 세 유전자의 프로모터에서 모두 GFP 발현이 확인되어 세 유전자의 프로모터가 포함된 벡터가 정상적으로 기능을 한다는 것을 확인하였다.
- 여러 조직 및 장기를 이용하여 발현을 확인한 결과 심근세포 유래 형질전환 세포에서만 GFP가 발현하는 것이 확인되어 심장 특이적 발현임을 확인하였다.
- 위 MYH6, TNNI3 및 MYBPC3 벡터를 기본으로 하여 형질전환 세포주를 개발하였다.
○ 바이오장기용 형질전환 복제난자 생산 효율 향상 연구
- 돼지의 체외성숙을 향상시키기 위하여 Hh-Ag1.5를 배양액에 첨가하여 배양을 실시한 결과 25 nM Hh-Ag1.5를 첨가한 처리군에서 높은 성숙율을 나타내었다. 반면 100 nM Hh-Ag1.5를 첨가한 경우 현저하게 저하되는 결과가 나타났다.
- 25 and 50 nM Hh-Ag1.5를 처리한 처리군에서 유의적으로 높은 배반포 발달율과 세포수를 얻을 수 있었으나 역시 100 nM을 처리할 경우 역효과가 나타나는 것을 확인하였다.
- 25 nM Hh-Ag1.5를 처리한 처리군에서 세포사 유발(pro-apoptotic) 유전자인 Bax의 발현이 낮게 나타났으며, 세포사 억제(anti-apoptotic) 유전자인 Bcl-XL 유전자의 발현은 높게 나타나는 결과를 얻었다.
- 난자를 질적으로 평가할 수 있는 유전자인 Tert와 Zfp42 유전자의 발현을 확인한 결과대조군과 25 nM 처리군에서 유의적으로 높게 나타났다.
- 체외 성숙율은 CM-25% 그룹에서 대조구보다 유의적으로 높았으나(P<0.05), 다른 SM 또는 CM 처리구와는 차이가 없었다. 배반포 형성율은 CM-25% 그룹(29.2%)에서 대조구(20.7%), SM-50%(19.6%) 및 CM-50%(23.66%) 처리구보다 유의적으로 높았다.
- 배반포에서의 세포수 및 세포사 비율은 SM-25% 그룹이 대조구에 비하여 유의적인 차이가 나타났다(P<0.05).
- 난자의 질과 연관되어 있는 유전자들(Oct4, Klf4, Tert 및 Zfp42)의 발현은 CM-25% 그룹에서 대조구보다 유의적으로 증가되었다(P<0.05).
○ 바이오장기용 형질전환 복제돼지 생산 및 기능 검증
- MYH6-GFP가 형질전환된 세포를 이용하여 복제돼지를 생산한 결과 1마리에서 MYH6 유전자가 포함된 벡터가 삽입되었음을 확인하였다. 하지만 이 돼지의 장기를 분석한 결과 GFP가 발현하지 않았다.
- 하지만 이 돼지의 체세포를 이용하여 iPSc를 구축한 다음 심근세포로 분화를 유도한 결과 GFP가 발현되는 것을 확인하였다.

The identification of differentially expressed genes and their promoters, which control gene expression in a tissue-specific manner, is of great importance for the investigation of transgene function and generation of transgenic animals for xenotransplantation.Identification of promoter sequences that can drive heart-specific expression of transgenes is essential to investigate gene function in the heart. The objectives of the present study were to identify differentially expressed genes in the mouse and human heart, evaluate their expression in pig hearts, analyze their promoter sequences for the regulation of heart-specific expression, and investigate expression of a target gene (green fluorescence protein, GFP) in pig primary heart cells under the regulation of the promoters. Gene expression profiles in the Gene Expression Omnibus (GEO) repository have been integrated and utilized to identify MYH6, TNNI3, and MYBPC3 as common heart-specific genes in the mouse and human. RT-PCR further confirmed heart-specific expression of the genes among various tissues in pigs as well as in the mouse and humans. Bioinformatics analysis predicted that their promoter sequences contain multiple binding sites for transcription factors involved in cardiogenesis and the promoter sequences were substantially conserved between pigs and humans. In addition, in vitro analysis showed that expression of green fluorescence protein (GFP) reporter gene under the regulation of promoter sequences of MYH6, TNNI3, and MYBPC3 was detected in primary pig heart cells but not in other primary cells. These findings, along with the physiologic similarities between humans and pigs, suggest that these novel promoters may be valuable candidates for the regulation of heart gene expression in both humans and pigs used for biomedical purposes. Using MYH 6 transgenic donor cells, the transgenic cloned piglet was successfully produced.
The present study was performed to investigate the effect of Hh-Ag1.5, a small-molecule chemical agonist of SMOothened receptor, on the in vitro maturation and development of in vitro fertilized (IVF) embryos in pigs. Oocytes or fertilized embryos were cultured in a maturation or embryo culture medium supplemented with 0 (control), 25, 50 or 100 nM of Hh-Ag1.5, respectively. Although the maturation rate were not different among treatment groups, the blastocyst formation rate in the group treated with 25 nM Hh-Ag1.5 was significantly increased compared to other groups (P<0.05). While the highest dose of Hh-Ag1.5 (100 nM) did negatively affect to the embryo development and cell number in blastocysts compared to other groups (P<0.05), the apoptotic cell index in blastocysts was significantly lower in 25 and 50 nM groups than in control and 100 nM groups (P<0.05). The mRNA expression of the pro- apoptotic gene Bax and the ratio of Bax/Bcl-XL decreased in among treatment groups compared to control (P<0.05). The embryo quality related genes, Tert and Zfp42, were significantly decreased in 50 and 100nM groups compared with control and 25 nM groups (P<0.05). In conclusion, the addition of 25 nM Hh-Ag1.5 to in vitro maturation and culture medium can enhance the developmental potential as well as quality of IVF embryos in pig.
The addition of growth factors and cytokines to in vitro culture (IVC) media could affect embryo development and the quality of the resulting blastocysts. The present study was performed to investigate the effect of porcine induced pluripotent stem cell (piPSC)-culture conditioned medium (CM) on the in vitro maturation (IVM) and development of parthenogentic embryos (parthenotes) in pigs. Cumulus-oocyte complexes (COCs) or activated oocytes were cultured in IVM or IVC medium supplemented with 0 (control), 25, or 50% of stem cell medium (SM) or CM, respectively. The maturation rate of CM-25% group was significantly improved when compared with control group (P<0.05), but that was not different among SM or CM groups. Blastocyst formation rate was significantly higher in CM-25% group (29.2%) than that of control (20.7%), SM-50% (19.6%) and CM-50% (23.66%, P<0.05). Cell number and the apoptotic cell index in blastocysts was significantly lower in SM-25% than in CM-25% group (P<0.05). The embryo quality related genes, OCT4, KLF4, TERT and ZFP42, were significantly increased in CM-25% group compared with control (P<0.05). In conclusion, the addition of 25% of CM to IVM and IVC medium positively influences not only the developmental potential also quality of parthenotes in pig. Transgenic pigs have been produced using the somatic cell nuclear transfer (SCNT) technique with MYH6-GFP cells, but unfortunately the GFP signal was not detected in heart. Induced pluripotent stem cells (iPSCs) originated from MYH6-GFP piglet were established. To analyze their differentiation capacity, the MYH6-GFP iPSc were induced to cardiac muscle cells and expressed stem cell markers.
