I. Purpose of Research
1. Although various drugs are being developed and used for RA, there are concerns about the low efficacy and side effects following continued use.
2. The use of MSCs as therapeutic approach for arthritis is well under development; however, the immunosuppressive effects of MSCs are variable depending on the investigator. Thus, there is a need for functional improvement of immunosuppressive function of MSCs.
Ⅱ. Research Summary and Scope
1. Establishment of optimal culture conditions for human and mouse BM-MSCs.
2. Establishment of optimal culture conditions for hUCB derived tol-DC, Treg, Tr1 and MDSCs.
3. Establishment of quality verification protocol for human BM-MSCs
4. Improvement of tol-DC functions using Tregs.
5. Evaluation of combined therapeutic effects of mouse MSCs with Treg or Tr1 in a murine CIA model.
Ⅲ. Results
1. Establishment of optimal culture conditions of mouse MSCs, Tr1, Treg, and tol-DC.
2. Development of combined cell therapy to improve immunomodulatory functions of MSCs.
3. Therapeutic effects of MSCs combined with Tr1 in a CIA model.
4. Isolation and culture of human MSCs for arthritis treatment and development of quality verification system.
5. There was no difference in cytokine secretion in cultured MSCs up to P4.
6. P4-cultured MSCs showed multipotent differentiation potential into osteoblasts, chondrocytes, and adipocytes.
7. Manufacturing method of MSCs expressing high levels of kyneurine and IL-1R production that can be applied as pre-treatment procedure for ex-vivo cultured MSCs was established.
8. Establishment of DC induction and expansion methods from hUCB CD34+ cells.
9. Establishment of quality verification standards and investigation of markers of tolDC for quality control.
10. Establishment of culture methods for MDSCs from hUCB-derived CD34+ cells.
11. Establishment of methods for tol-DC functional improvement using Tregs.
12. Induction of hUCB-derived iTregs using TGF-b and retinoic acid.
13. Establishment of induction methods of hUCB-derived Tr1 cells
Ⅳ. Future application of results
It is probable that the combined cell therapy developed from the current study can be commercialized for practical use. The commercialization of combined cell therapy for RA could allow the application of this approach in various autoimmune diseases. This leads to new possibilities and opportunities, especially in incurable diseases where no effective treatment and care is available.
