유전체, 생약, 식물분류팀의 협력 연구를 통해 차세대 유전체 분석 (NGS) 기술을 이용하여 5가지 생약 (인진호, 한인진, 포공영, 한련초, 홍화자)의 기원종에 해당하는 기원식물 10종 (인진호:사철쑥, 한인진: 더위지기; 포공영: 민들레, 흰민들레, 서양민들레, 털민들레,감지포공영; 한련초: 한련초; 홍화자:잇꽃)과 근연종식물 3종(큰비쑥, 가는잎한련초, 산민들레) 포함 전체 13종에 대해 엽록체와 핵내 ITS영역을 포함하는 45S rDNA 전체서열을 완성하고 기원종과 근연식물 및 위품 가능한 식물종을 비교하여 기원식물을 안정적으로 판별할 수 있는 바코드 마커 시스템을 구축하였다. 더불어 국내 생산되는 생약 재료의 생물주권확보를 위한 정보와 DB 구축자료를 마련하고 유전체, 대사체 정보 상호 비교를 통해 근연종과 동일 기원종의 다양한 샘플을 구별하는 안정적이고 신뢰할 수 있는 실용적 바코드 마커를 개발하여 기원종의 구분에 대한 체계를 제공하고자 하였다. 본 연구에서는 NGS 기술을 이용하여 5가지 생약 품목에 해당하는 기원식물 13종 식물 17 점(4종은 2개의 종내변이 계통, 즉 다른 지역에서 수집된 계통 포함하였음)에 대해 엽록체와 핵내 ITS영역을 포함하는 45S rDNA 전체서열을 완성하였다. 비교유전체방법을 이용하여 대상 기원종 식물내 변이 여부를 확인하고 동일 종내 변이는 없으면서 다른 종을 식별할 수 있는 기원식물 특이 마커를 제작하였다. 인진호/사철쑥과 비교종인 큰비쑥을 구분하는 마커 7종, 포공영 기원식물과 비교종인 산민들레 구분마커 6종, 한련초와 가는잎 한련초 구분마커 3종, 국내산과 중국산 홍화 구분 마커 2종을 개발하였다. 선별된 마커를 이용하여 최종 선발하여 식약처에서 제공받은 검체에 적용, 안정성을 확인하였다.
또한, 대상품목의 대사체 프로파일링을 수행하였는데, 인진호와 한인진, 포공영의 경우 국내에서 생산되는 여러 종류의 기원식물에 대해 대사체 비교 분석을 수행하였다. 11종의 쑥과 5종의 민들레에서 대사체 분석을 수행하였으며, 각각의 종특이 대사체 마커를 선정하였다. 유전체 분석과 대사체 분석을 통해 포공영의 기원식물들과 비교종인 산만들레가 가까운 근연관계임을 확인하였다.

In this study, we developed authentification system to discriminate five herbal medicine (Artemisiae Capillaris, Artemisiae Iwayomogii, Taraxaci, Ecliptae Herba, and Safflower) from those medicinal species and allied species using genetic and metabolomic analysis. We first selected thirteen species as origin and allied species such as Artemisiae capillaris, A. gmelinii Weber ex Stechm, Taraxacum platycarpum H. Dahlstedt, T. officinale Weber, T. coreanum Nakai, T. mongolicum Handel-Mazzetti, T. borealisinense Kitam, E clipta prostrata Linné, Carthamus tinctorius Linné,A. fukudo Makino, T. ohwianum Kitam, and E. alba L. Hass. Then we assembled those complete chloroplast genomes and analyzed 45s nrDNA sequences. Through comparative genomic analysis,we developed chloroplast-based molecular markers, which are able to discriminate inter-species variations without intra-species variations. Total seven InDeL markers were developed against three Artemisiae species and three molecular markers were developed against two Eclipta species. Two molecular markers were able to distinguish origin of C. tinctorius derived from Korea and China. Combinations of six molecular markers were able to identify six Taraxacum species. Also the developed molecular markers were applied to herbal medicine materials.
Total 5 Taraxacum and 11 Artemisiae species were performed to metabolite profiling, and identified metabolic markers derived from each species. Finally, genetic diversity among 13 species including 10 medicinal plants and 3 allied species were identified using genetic analysis such as those chloroplast genome sequence and 45s nrDNA and those metabolites profiling analysis.
This authentification system using genetic and metabolomics studies provide comprehensive information to discriminate medicinal plants and genetic diversity.
