NCBI GenBank로부터 선정된 유전자 마커의 염기서열을 검색 하였으며, GenBank에 정보가 없는 경우는 직접 염기서열을 분석하여 확인하였다. 비교 대상종 간의 염기서열 정보를 비교·분석하여 종 특이적인 영역을 선별 하였으며, 선별된 염기서열에서 종 특이 프라이머를 설계하였다. 개발된 종 특이 프라이머는 NCBI Primer-BLAST에서 제공하는 “프라이머 특이성 검색” 기능을 활용하여 대상종 이외의 종과의 염기서열 유사성을 확인했고, 종 특이 프라이머를 사용하여 최종 유전자 분석법을 개발하였다. 종 특이 polymerase chain reaction (PCR)의 최적 조건을 확립하기 위해 (1) 결합온도 실험(50∼65℃), (2) PCR cycle 수 실험(30, 35, 및 40 cycles) 및 (3) 검출한계 실험(5∼0.00005 ng/㎕ DNA)을 수행하였다. 개발된 종 특이 PCR법은 대상 종에서만 종 특이적 PCR 산물(100∼300 bp)을 생성하였으며 비교종에서는 교차 반응을 보이지 않았다. 따라서 이번 연구를 통해 개발된 45종의 유전자 분석법은 육안 확인이 어려운 식품원료의 진위판별에 사용될 수 있으며, 정책 제안 등을 통해 불량식품의 제조·유통을 차단하여 소비의 식품에 대한 불안감 해소에 크게 기여할 것으로 기대된다.

We finally selected 22 animal species (Mustela putorius, Meles meles, Arctoscopus japonicus, Sardinops melanostictus, Buccinum striatissimum, Polinices didyma, Sardinella zunasi, Sciaenops ocellatus, Pennahia argentata, Larimichthys crocea, Acanthopagrus schlegelii, Fenneropenaeus chinensis, Marsupenaeus japonicus, Fenneropenaeus merguiensis, Penaeus monodon, Pandalus hypsinotus, Lates calcarifer, Makaira mazara, Lampris guttatus, Pangasius santitwongsei, Litopenaeus vannamei, Macrobrachium rosenbergii) and 23 botanical species (Stachys affinis, Lycopus lucidus, Maclura tricuspidata, Robinia pseudoacacia, Hippophae rhamnoides, Eleutherococcus senticosus, Kalopanax septemlobus, Aralia elata, Hovenia dulcis, Houttuynia cordata, Saururus chinensis, Momordica charantia, Suaeda japonica, Suaeda glauca, Achyranthes japonica, Ganoderma applanatum, Tremetes versicolor, Acer palmatum, Acer pictum, Acer pictum, Piper nigrum, Petroselinum crispum, Ocimum basilicum) that could be used for food frauds through expert group meetings. Species-specific primers were designed based on Cytochorme B and Cytochrome oxidase subunit I genes for animal ingredients and trnL intron region and internal transcribed spacer for botanical materials as a genetic marker. Sequences of each marker were searched through NCBI GenBank, in case of no sequence information, the sequences were confirmed by sequencing the marker genes from our standard materials. Alignment of marker gene sequences of similar species revealed the species-specific regions where species-specific primers were designed. Specificity of the species-specific primer pair against other species was confirmed by using NCBI Primer-BLAST. The optimal conditions for species-specific polymerase chain reaction (PCR) were tested under three criteria; (1) annealing temperature (55∼65℃), (2) PCR cycles (30, 35, 40), and (3) Detection limit (5∼0.00005 ng/㎕ DNA). Under the optimized conditions, species-specific PCR produced 100∼300 bp PCR products, and no cross-reaction was observed among similar species. Thus, PCR analysis developed for the 45 species in this study can verify the authenticity of food ingredients. In addition, our screening method can be utilized to prevent the distribution of economically motivated adulteration food and to improve consumer's right.
