본 연구는 화장품 중 배합한도 성분인 파라벤, 세트리모늄브로마이드(클로라이드), 트리에탄올아민 및 화장품 중 배합금지 성분인 자일렌에 대한 분석법을 확립하는 것을 목적으로 하였다. 파라벤은 화장품에서 단일성분으로 최대 0.4%, 혼합성분으로서 0.8%까지 사용할 수 있으나 최근 파라벤의 인체 유해성 논란이 지속되면서 파라벤을 사용하지 않았다는 “무파라벤” 광고를 하는 제품이 많은 상황이다. 제품에 특정성분이 들어있지 않다는 “무(無)○○”표현을 사용하기 위해서는 시험분석자료로 이를 입증해야 하는데 기존의 함량시험으로는 비의도적으로
본 연구는 화장품 중 배합한도 성분인 파라벤, 세트리모늄브로마이드(클로라이드), 트리에탄올아민 및 화장품 중 배합금지 성분인 자일렌에 대한 분석법을 확립하는 것을 목적으로 하였다. 파라벤은 화장품에서 단일성분으로 최대 0.4%, 혼합성분으로서 0.8%까지 사용할 수 있으나 최근 파라벤의 인체 유해성 논란이 지속되면서 파라벤을 사용하지 않았다는 “무파라벤” 광고를 하는 제품이 많은 상황이다. 제품에 특정성분이 들어있지 않다는 “무(無)○○”표현을 사용하기 위해서는 시험분석자료로 이를 입증해야 하는데 기존의 함량시험으로는 비의도적으로 혼입된 성분의 존재유무를 확인하기 어려운 상황이다. 따라서 미량 파라벤 분석을 위한 시험법 개발이 필요하다. 세트리모늄브로마이드(클로라이드)는 두발용 제품을 제외한 화장품에 최대 0.1%까지, 트리에탄올아민은 사용 후 씻어내지 않는 제품에 최대 2.5%까지 사용할 수 있다. 식품의약품안전평가원이 제공하는 화장품 중 배합한도 분석법 가이드라인에 위의 두 성분의 분석법이 없어 새롭게 분석법 확립이 필요한 실정이다. 한편, 기존에 공산품으로 관리되던 물휴지가 화장품으로 분류되면서 기존의 품질기준을 반영하여 자일렌이 화장품 중 배합금지 성분으로 신설되었다. 다만, 자일렌이 화장품 원료의 제조공정에서 용매로 사용되었으나 완전히 제거할 수 없는 잔류용매로서 제품 중 0.002%(손발톱용 제품류는 0.01%) 이하인 경우는 제외이다. 그러므로 화장품에서 잔류용매로서 자일렌 함량을 분석하기 위한 시험법 개발이 시급한 실정이다. 파라벤 6종, 세트리모늄브로마이드(클로라이드), 트리에탄올아민 및 자일렌 분석법 확립을 위해 액체크로마토그래피, 액체크로마토그로피 질량분석기, 가스크로마토그래피 질량분석기를 이용하였다. ‘의약품 등 시험방법 밸리데이션에 대한 가이드라인’에 따라 밸리데이션을 수행한 결과, 다섯 성분의 분석법 모두 상관계수(R2) 0.999 이상의 직선성과 우수한 분리능 및 선택성을 보여주었다. 외부기관 밸리데이션에서을 통한 교차검증에서도 일관성 있는 결과를 확인하였다. 또한 유통화장품 116(216)종에 대한 모니터링을 수행한 결과 다섯 성분 모두 제품에서 사용한도 이내로 검출되거나 검출되지 않았다. 확립된 네 성분의 분석법은 화장품 중 배합한도 및 금지 성분 분석법 가이드라인을 통해 관련 업계에 배포해 화장품 품질관리에 활용될 예정이다. 본 연구에서 확립된 사용한도 성분의 분석법은 유통 화장품 품질관리 및 안전성 평가의 기초자료로 활용되어 국민들이 안심하고 화장품을 사용하는 데 기여할 것으로 기대된다.

This research aims to set up methods of analyzing the restricted ingredients of the cosmetic products, i.e., paraben, cetrimonium bromide(chloride), and the prohibited ingredient of xylene. The maximum permissible paraben content of cosmetics is up to 0.4% as single substance and 0.8% as mixed substances. Since there has been controversy about safety of parabens persistently, many of products have been insisting they didn't have paraben ingredients by using "Paraben Free” advertising phrase. In order to use the marketing phrase of “Free” about a certain substance, the company should prove that the product really does not contain the substance in trouble with the basis of scientifically rational analysis data. However, It is difficult to prove that certain ingredients which are mixed unintentionally are existent or not in cosmetic products by using current content analysis method. Therefore further studies are needed to develop analysis method for identifying micro-content of paraben. Cetrimonium bromide(chloride) can be safely used up to 0.1% to any cosmetic products except hair products. Triethanolamine is limited to a maximum content of 2.5% for the live on products. The current guidelines on the analysis of cosmetic ingredients published by the NIFDSE does not provide analyzing method for cetrimonium bromide(chloride) and triethanolamine. Therefore a new way to analyze such substances needs to be developed. Meanwhile, since wet wipes became categorized as cosmetic products from industrial products, xylene became newly prohibited. Altho㎍h xylene is prohibited ingredients of the cosmetic products, but it may be allowed to a maximum content of 0.002%(when used in nail care products, it is limited up to 0.010%) when it was used as a solvent during the manufacturing process and remains in cosmetics as residual solvent. In this regards, it is timely needed to develop analyzing method for the content of xylene as residual solvent.
In this study, we have developed HPLC, LC-MS/MS, and GC-MS methods for the determination of parabens (6 types), cetrimonium bromide(chloride), triethanolamine, and xylenes. These assays have been fully validated under the “Guideline for the validation of dr㎍ analysis procedure”. Thro㎍h the validation results, analysis methods for all 5 ingredients showed linearity with correlation coefficients (R2) more than 0.999 and have shown outstanding selectivety and resolution. Cross-validation of these analysis methods conducted by outside agencies showed consistent results.
In addition, according to the monitoring for 116(216) different cosmetic products in the market, all 5 ingredients have not been detected or detected within the allowed limitation. These analysis methods will be offered as "Guideline for analysis method of restricted(prohibited) ingredients of cosmetics" to related industry. We expect that the results of this study can be usefully applied in QC and the safety assessment of commercially-available cosmetic products helping customers use the products more safely.
