A. Development of anti-allergic foods using natural materials
○ To discover and develop natural functional materials that can improve allergy, natural medicine specimens (about 280 kinds), naïve food materials (about 290 kinds), marine plant materials (56 kinds), food-derived plant materials (54 kinds) and food materials (133 kinds) were acquired. The target mechanism underlying continuous allergic induction was established, and related cell tests were established. (1) test for the activity that inhibits the permeation of allergens into intestinal epithelial cells by using Caco-2 cells, (2) test for the activity that regulates T cells by using splenocytes of Th2 response-induced mice, (3) test for the activity that induces the differentiation of regulatory T cells by using naive T cells of mice, (4) test for the activity by which B cells inhibit IgE production by using U266B1 cells IgE, and (5) test for the activity that inhibits degranulation by using mast cells were performed. Anti-allergic activities of about 800 kinds of natural products were evaluated through each established cell test system. About 50 kinds of primary anti-allergic functional materials were finally discovered.
○ Allergic disease animal models (allergic contact dermatitis, food allergy, atopic dermatitis, allergic rhinitis and allergic asthma) were established for about 50 kinds of materials selected based on anti-allergic activity tests at the cellular level, and anti-allergic functionality was evaluated for the primarily selected materials. 8 kinds of materials (rosa multiflora fruit, pepper, scutellaria baicalensis, fenugreek, poria cocos bark, naïve citrus Hongkyool leaves, achyrantis radixs, and turmeric) were finally selected.
○ Among the 8 kinds of finally selected materials, rosa multiflora fruit, which was discovered as a top priority anti-allergic material, showed an excellent efficacy in various allergic diseases such as allergic rhinitis, asthma, atopic dermatitis, allergic contact dermatitis and food allergy. For example, when rosa multiflora fruit extract was administered for 8 weeks to a model of atopic dermatitis induced by house dust mite, a typical allergen, symptoms such as erythema, keratin and edema were reduced compared with the control group, and IL4s as mite-specific IgE and Th2 cytokines was also reduced compared with the control group. In addition, when immune cells infiltrated into the lung in an OVA-induced asthma model were analyzed with the bronchoalveolar lavage fluid (BALF), infiltrated immune cells (eosinophils, neutrophils, lymphocytes, macrophages) were found to be inhibited by the intake of rosa multiflora fruit.
○ The results of animal experiment were reproduced in a clinical trial. When patients with rhinitis were given rosa multiflora fruit 1g daily for 2 – 4 weeks, TNSS (Total Nasal Symptom Score) and the symptoms of rhinitis such as watery rhinorrhea were alleviated, and such a symptom alleviating activity lasted for up to 2 weeks after the end of its administration, showing excellent activity in clinical studies.
○ Up to date, standardization studies and safety studies together with functional studies on rosa multiflora fruit have been conducted. The technology transfer licensing with regard to those results was completed, and in collaboration with the technology transfer licensee, it is planned to register it as an individual authorized raw material for improving nasal conditions associated with hypersensitive immune response.
B. Development research on anti-allergic food using probiotics
○ To confirm anti-allergic activity of about 100 kinds of microorganisms derived from traditional fermented foods, anti-inflammatory cytokine secretion activity, regulatory T cell induction activity, Th2 cytokine secretion inhibitory activity, and IgE secretion inhibitory activity were evaluated, from which 5 kinds of strains with anti-allergic activity were finally selected. The selected strains were evaluated for their anti-allergic activity using four allergic disease animal models (food allergy, atopic dermatitis, contact dermatitis and asthma), and Lactobacillus pentosus KF340 (LPKF340), which has excellent activity for improving atopic dermatitis and contact dermatitis, was discovered.
○ LPKF340 showed excellent activity for atopic dermatitis. When LPKF340 was administered for 8 weeks to an atopic dermatitis model induced by a typical allergen, house dust mite, symptoms such as erythema, keratosis and edema were reduced compared with the PBS-treated group, and IL-4, a Th2 cytokine, was also reduced compared with the PBS-treated group. The degree of reduction was similar to that of dexamethasone, an immunosuppressant, confirming that the LPKF340 strain had excellent inhibitory activity for atopic dermatitis. In comparison with 3 functional foods that are commercially available for improving skin condition associated with hypersensitive immune response, the LPKF340 showed similar or excellent activity. It was confirmed that LPKF340’s inhibitory activity for atopic dermatitis was induced by Breg which secretes IL-10, an anti-inflammatory cytokine, and was high when administered alone or in the state of live bacteria.
○ After establishing optimum medium/culture conditions for the LPKF 340 strain, experimental products of 3 batches were produced by applying the mass production process. Through that process, the quality standards, which meet the standards and specifications of hazardous substances set by the Ministry of Food and Drug Safety, were established.
○ When the safety and stability of LPKF340 were evaluated using experimental products, the results showed that LPKF340 is safe, its storage stability at 4℃ was excellent and its survival rate under intestinal tract condition was excellent. Based on the above mentioned results, technology transfer for LPKF340 was completed.
