개발 목적 및 필요성
생활환경 및 산업환경으로부터 납, 카드뮴을 비롯한 다양한 유해중금속에 노출되는 사례가 국내외적으로 빈번해지면서, 유해중금속 납과 카드뮴의 관리, 규제 방안, 저감기술, 독성평가 기술 개발 등 국제규제 대응 방안이 절실히 요구되고 있는 실정이다. 본 연구진은 유해 중금속의 위해저감기술 개발을 통해 유해중금속 관련 국제규제에 대해 적절한 대응방안을 제시하고, 국내 유해중금속 저감기술 발전에 기여하고자 하였다.

생활환경 및 산업환경으로부터 납, 카드뮴을 비롯한 다양한 유해중금속에 노출되는 사례가 국내외적으로 빈번해지면서, 유해중금속 납과 카드뮴의 관리, 규제 방안, 저감기술, 독성평가 기술 개발 등 국제규제 대응 방안이 절실히 요구되고 있는 실정이다. 본 연구진은 유해 중금속의 위해저감기술 개발을 통해 유해중금속 관련 국제규제에 대해 적절한 대응방안을 제시하고, 국내 유해중금속 저감기술 발전에 기여하고자 하였다.
연구개발 결과
○ 유해 중금속 통합안전성 정보시스템 구축
- 유해 중금속 납, 카드뮴 관련 문헌조사를 통해 사용량 및 유통 정보와 특허 동향을 분석 및 수집하였다. 또한 국내 기업체를 대상으로 납, 카드뮴 사용에 대한 설문 조사를 시행하여, 이를 기반으로 납과 카드뮴의 사용 현황 및 그에 따른 인식에 대한 데이터베이스를 구축하였다. 이 데이터베이스를 기반으로 웹 기반의 통합 안전정보 시스템을 구축하여 유해 중금속 납, 카드뮴 위해성 정보를 제공할 수 있도록 하였으며, 이는 향후 유해중금속의 유통 및 활용에 대해 신속하고 효율적인 관리를 가능케 하고 현재 중금속 독성 및 노출 관리에 소요되는 막대한 시간과 예산을 효과적으로 감소시킬 수 있을 것으로 사료된다.
○ 유해중금속 납, 카드뮴 저감 및 대체기술 개발
- 전기분해 공정을 기반으로 산세폐액 내 중금속 제거 공정기술을 개발함과 동시에, 친환경적인 산화철 입자 제조기술을 개발하여 이를 통해 유해중금속 대체 소재인 산화철 안료를 생산하였다. 열처리 공정 및 첨가물 투입을 통해 불순물을 제거하고 산화철 안료의 색상을 다양하게 구현하였으며, 안료 회사와의 논의를 통해 제조한 산화철 안료의 적용성 연구 및 제조/공정 기술의 사업화 및 기술 이전 가능성을 확인하였다.
본 기술을 통해 고유해성 물질의 대체물질 개발 및 저감 기술과 관련된 국가 경쟁력을 향상시킬 수 있으며, 오염물질 사용 및 배출을 최소화함으로써 작업환경 개선과 오염방지 시설 설치 및 운영에 소요되는 비용을 절감할 수 있을 것으로 사료된다.
○ 유해중금속 납, 카드뮴 특이 독성평가 기술 개발 및 대체기술로 제조된 산화철의 독성저감효과 평가
- 전기분해 기술로 제조한 산화철 안료의 유해 중금속 납, 카드뮴 대비 독성 저감효과 검증을 위하여 인간유래세포주 및 동물모델을 이용한 유해중금속 특이 독성 평가 및 산화철 안료의 독성저감효과를 평가하였다.
효율적인 검증을 위하여 통합적 독성평가 플랫폼을 제시함으로써 다양한 수준에서의 독성평가 시스템을 구축하였다. 또한 환경시험생물종을 이용하여 유해중금속 특이적인 바이오모니터링을 위한 기반 연구를 수행하였다. 이러한 유해중금속 특이 독성평가기술의 개발을 통해, 유해중금속 특이적인 건강영향평가 기반마련 및 관련 분야에 대한 신산업 및 전문 인력의 창출에 크게 기여할 수 있을 것으로 판단되는 바이다.
활용계획
본 연구에서 개발된 유해중금속 저감기술들은 환경보건학적, 사회적, 경제적 관점에서 중요한 의미를 가질 것으로 판단되며, 특히 유해중금속의 국제규제 대응기술력 향상에 기여할 수 있을 것으로 사료된다.

(1) Establishing an system of integrated safety information system on heavy metal
Compared to the USA, information about domestic distribution and emissions of lead and cadmium are relatively scarce in the majority of situations here. Therefore, the available information about domestic distributions, emissions and usage of lead and cadmium was examined. In the case of lead, distributions amounted to 263,000t. Emissions were highest in the manufacturing combustion sector and production processes. Lead usage was 413,000t and was highest in electrical equipment manufacturing. In the case of cadmium, distributions amounted to 413t.
Emissions were highest in the manufacturing combustion sector and production processes. Cadmium usage was 76.3t and was highest in the electronics industry such as n manufacturing computers and communication equipment. Our investigation about the prevalence of products containing lead and cadmium found that batteries, ammunition, electronics, cosmetics, and especially children’s goods most frequently exceeded standards. This result indicates that heavy metal-related regulation should be reinforced, and that of heavy metal reduction/substitution technologies are needed.
A list of companies using lead and cadmium was established. A total of 1,435 of companies were manufacturers of lead and cadmium, and 897 companies were distributors. Replying to questionnaires related to heavy metal regulation and self-regulation by each companies, almost all companies indicated that the first reduction target should be lead or cadmium. Moreover, they also respond about requiring safety assessments of children’s goods and establishing a manual related to heavy metal accidents.
Using the accumulation of literature data collected from above, and, the company survey data, we established a database and web-based ‘system of integrative safety information on lead and cadmium’ including inventory information, research trends, safety information, statistical data, reduction/substitution trends and trends of policies related to lead and cadmium. The database is continuously updated with users’ requirements and recent trends of related studies. A web-based survey was added to collect domestic companies’ opinions about lead and cadmium usage.
(2) Development of technology for reduction/substitution of heavy metal lead and cadmium
Diverse manufacturing processes including wet corrosion, microwave, and electrolysis were compared to develop technology for reduction/substitution of the heavy metals, lead and cadmium. An electrolysis method was utilized to develop eco-friendly iron oxide particles because of its high efficiency. Pickling waste fluid from the steel industry contains diverse harmful heavy metals. Because these heavy metals can be removed by an electrolysis process, we tried to eliminate four heavy metals (cadmium, lead, chrome and mercury) using electrolysis. To establish an electrolytic process for purifying iron oxide particles from pickling waste fluid, the optimal conditions such as concentration of electrolyte, current density, etc. were investigated. Changing some conditions in the electrolysis process resulted in differences in products. In addition, the concentrations of raw materials or heat treatment conditions were assessed to determine the proper conditions for applying pigments containing iron oxide particles. The colors were analyzed according to numbers of iron oxide particles. We confirmed that the color of iron oxide particles depends on the regulation of waste fluid acidity and particle numbers. The heavy metals were effectively eliminated as the electrolysis time increased. Thus it was confirmed that an electrolysis method is appropriate to eliminate harmful heavy metals from waste fluid. Based on these results, we established electrolysis as a process to manufacture alternatives to heavy metal pigments.
These developments for reduction/substitution of the heavy metals, lead and cadmium, are advanced technologies in terms of waste recycling and environmental protection. Thus, we discussed business possibilities and transfer of skills for the above technologies with a domestic pigment manufacturing company. Various aspects such as economic, technical and environmental health were discussed, and a positive response was obtained from the company.
(3) Development of technology for evaluation of heavy metal-specific toxicity.
We analyzed the research trends in studies of heavy metal toxicity to suggest a course of study about lead/cadmium-specific toxicity and oral administration. We also conducted in vivo and in vitro toxicity evaluation studies of lead/cadmium exposure.
The in vivo long-term and low-level exposure studies, and detected lead/cadmium-induced genotoxic effects. The reduction of DNA repair activity and induction of apoptosis due to lead/cadmium exposure in rat tissue were also investigated. In the acute and semi-acute test using comet assay, MN assay, GSH/GSSG assay, APE activity assay, MT1 westernblotting and H&E staining method, we significant biological damage was detected in the group exposed to lead/cadmium. In vitro studies also demonstrated significant toxic effects due to lead/cadmium exposure. An integrated analysis of all toxicity results was performed to reduce the bias of the in vivo experiments.
In addition, an evaluation of iron oxide safety as an alternative material lead and cadmium was conducted. In the acute and semi-acute test using the same procedure as the heavy metal toxicity test, significantly less damage was observed in the iron oxide exposed group. Moreover, the integrated toxicity scoring process was also applied for reduction of bias in this in vivo study. As a result, it was confirmed that iron oxide particles are significantly safer than lead/cadmium pigments. The gene list which is differentially expressed by long term, low level exposure to lead/cadmium was obtained through microarray techniques, and analysis of their biological pathways. In particular, a lead-specific gene list was identified using RNA-seq and pathway analysis, and candidates for lead/cadmium specific biomarkers were suggested. Moreover, we developed a heavy metal sensor using transformed yeast, and confirmed the possibility of an early detection system for prevention of heavy metal exposure.
To evaluate the toxicity of cadmium and lithium as aquatic environment pollutants, Daphnia magna (D. magna) was used as is recommended by Organization for Economic Co-operation and Development (OECD). We obtained the gene lists through microarrays using RNA from cadmium/lithium exposed D. magna, and applied BLAST tools in NCBI for comparative genomic analysis between human and D. magna genes. Through pathway analysis with them, as a result, we suggested 23 candidates of cadmium-specific biomarkers and 53 candidates for lithium-specific biomarkers.
