(1) 혈관 생성 모사시스템 구축 및 고혈당 조건에서 혈관 생성 능력 및 세포간의 신호 전달 메커니즘 연구
혈관 내피 전구세포 (ECFC)는 말초 혈액에서, 혈관벽 전구세포 (MPC)는 골수에서 분리한 다음, 정상 혈당 (5mM, NG) 또는 고혈당 (30mM, HG)에서 7일간 배양 후 실험이 사용하였음. 실험 결과, 고혈당에서 배양한 ECFC는 세포 증식 능력과 혈관 생성 능력이 모두 유의하게 감소되었으나, 흥미롭게도 MPC와 혼합 배양된 ECFC는 고혈당 조건에서도 혈관 생성 능력이 정상 상태에서 배양된 ECFC와 유사하
(1) 혈관 생성 모사시스템 구축 및 고혈당 조건에서 혈관 생성 능력 및 세포간의 신호 전달 메커니즘 연구
혈관 내피 전구세포 (ECFC)는 말초 혈액에서, 혈관벽 전구세포 (MPC)는 골수에서 분리한 다음, 정상 혈당 (5mM, NG) 또는 고혈당 (30mM, HG)에서 7일간 배양 후 실험이 사용하였음. 실험 결과, 고혈당에서 배양한 ECFC는 세포 증식 능력과 혈관 생성 능력이 모두 유의하게 감소되었으나, 흥미롭게도 MPC와 혼합 배양된 ECFC는 고혈당 조건에서도 혈관 생성 능력이 정상 상태에서 배양된 ECFC와 유사하게 유지됨을 관찰하였음. 더불어서, 혈관 생성을 조절하는 인자들의 발현 변화를 측정한 결과, 고혈당 배양에서 감소되는 것으로 측정된 thrombopoietin, VEGF, VEGF-D, MCP-1의 발현이 MPC와의 혼합 배양에서는 다시 정상 상태로 회복됨이 관찰하였음.
(2) 허혈 부위에 혈관 생성 및 혈류 회복을 목적으로 two-cell system의 세포 치료법으로서의 가능성 연구 하지 허혈 근육 조직에 ECFC, MPC, ECFC+MPC를 주입한 결과, ECFC+MPC에서 혈류의 회복이 ECFC, MPC 단독 주입과 비교하여 유의적으로 증가되었고, 혈관의 개수도 유의적으로 증가하였음. Flow cytometric analysis 분석에서는 myeloid cells, neutrophils, macrophages, monocytes 등 다양한 면역세포들이 ECFC+MPC가 주입된 허혈 근육에 다량 유입되는 것을 관찰하였고, anti-Gr-1 antibody를 투여하여 동물의 면역 세포를 고갈시켜 면역 세포의 유입을 저하시킨 경우에는 ECFC+MPC에 의한 혈류회복 효과가 사라지는 것을 관찰하였음.
고혈당 상태에서 배양된 ECFC+MPC를 당뇨쥐에 주입한 결과, 정상쥐에 정상 혈당에서 배양한 ECFC+MPC를 주입한 대조군과 비교하여 비슷한 정도의 혈관 생성 능력을 보였음. 이와 같은 결과들은 two-cell system (ECFC+MPC)가 혈관 손상으로 야기되는 허혈 조직에 혈관의 재생과 혈류의 회복에 임상 적용될 수 있는 치료법으로서의 가능성을 강력하게 밑받침하고 있음.

ECFC were isolated from human peripheral blood, and MSc were isolated from human bone marrow. ECFC and MSC were cultured in each growth mendium containing normal glucose (5 mM;NG) or high glucose (30 mM;HG) for 7 days. The proliferation rate and tube formation ability of ECFC were significantly reduced in HG compared to NG. Interestingly, ECFC coincubated with MPC showed similar degree of tube formation between HG and NG. The reduced thrombopoietin, VEGF, VECF-D, MPC-1 in ECFC were also restored to the normal level when coincubated with MPC.
(2) vasculogenic ability of two-cell system within ischemic tissues of animal models
ECFC+MPC, ECFC alone, MPC alone, or vehicle alone were injected into the hind limb ischemic muscle. At day 5, hind limbs injected with ECFC+MPC showde greater blood flow recovery as well as microvessel density compared with ECFC, MPC, or vehicle. Flow cytometric analysis of ischemic muscles at day 2 revealed increased myeloid lineage cells including myeloid cells, neutrophils, macrophages, and monocytes in ECFC+MPC-injected muscles compared to vehicle-injected muscles.
Systemic myeloid cell depletion sith anti-Gr-1 antibody blocked the improved blood flow observed with ECFC+MPC. Myeloid cell depletion also reduced ECFC and MPC retention. Our data suggest that ECFC+MPC delivery could be used to reestablish blood flow in ischemic tissues, and this process may be enhanced by coordinated recruitment of host myeloid cells.
In another animal experiment, diabetic condition was induced into the mouse by streptozitocin injection, and HG-treated ECFC+MSC were mixed and injected into the diabetic mouse subcutaneously. After 7 days, the number of perfused blood vessels formed by human ECFC+MPC in diabetic mouse was comparable with vessels formed by ECFC+MPC in normal mouse. Our results demonstrate that vasculogenic capability of ECFC+MPC is intact in hyperglycemic condition, which suggests that ECFC delivery in combination with MSC can apply to the regeneration of functional microvessel network in diabetic ischedmic region.
