본 연구는 식의약 선진화 3개년 계획(식품 중 유해물질 등 재평가 계획)에 근거하여 식품 중 중금속의 주기적 안전성 평가 수행에 필요한 모니터링 자료 생산을 통한 중금속 실태조사 및 위해평가를 실시하였다. 식품 중 중금속 실태조사 관련 국내・외 문헌조사를 실시하였고, 시료채취, 분석법, DB관리 표준화 등 모니터링 시스템 표준화, 인증표준물질(CRM) 등을 통한 분석법 검증 및 측정불확도를 산출하였다.

본 연구는 식의약 선진화 3개년 계획(식품 중 유해물질 등 재평가 계획)에 근거하여 식품 중 중금속의 주기적 안전성 평가 수행에 필요한 모니터링 자료 생산을 통한 중금속 실태조사 및 위해평가를 실시하였다. 식품 중 중금속 실태조사 관련 국내・외 문헌조사를 실시하였고, 시료채취, 분석법, DB관리 표준화 등 모니터링 시스템 표준화, 인증표준물질(CRM) 등을 통한 분석법 검증 및 측정불확도를 산출하였다.
농・수・축산물, 가공식품, 특수용도식품 등에 대하여 중금속 5종(납, 카드뮴, 비소, 수은, 알루미늄) 및 일부시료에 대하여 메틸수은, 주석, 무기비소를 분석하였다. 납, 카드뮴, 비소, 알루미늄, 주석 함량 분석을 위해 microwave장치를 이용해 전처리한 후 ICP-MS 또는 ICP-OES로 측정하는 시험법을 확립하였으며, 수은은 수은자동분석기, 메틸수은은 GC-ECD, 무기비소는 HPLC-ICP-MS를 이용한 분석법을 확립하였다.
모든 중금속의 검량선은 R2값이 0.999 이상이며, 중금속의 검출한계는 대상식품의 매질별로 확인한 결과, 농산물 중 납은 0.0002 ∼ 0.0059 mg/kg, 카드뮴은 0.0001 ∼ 0.0030mg/kg, 비소는 0.0004 ∼ 0.0097 mg/kg, 알루미늄은 0.0116 ∼ 0.2743 mg/kg, 수은은 0.0001 ∼ 0.0005 mg/kg, 무기비소는 0.0004 mg/kg 이었다. 축산물 중 납은 0.0002 ∼ 0.3000 mg/kg, 카드뮴은 0.0001 ∼ 0.4000 mg/kg, 비소는 0.0002 ∼ 1.0000 mg/kg, 알루미늄은 0.0031 ∼ 1.5000 mg/kg, 수은은 0.00003 ∼ 0.0003 mg/kg 이었다. 수산물 중 납은 0.0001 ∼ 0.0250 mg/kg, 카드뮴은 0.0001 ∼ 0.0125 mg/kg, 비소는 0.0002 ∼ 0.025 mg/kg, 알루미늄은 0.0050 ∼ 0.0250 mg/kg, 수은은 0.00004 ∼ 0.018 mg/kg, 메틸수은은 0.0013 ∼ 0.012 mg/kg, 무기비소는 0.011 mg/kg 이었다. 가공식품 중 납은 0.0002 ∼ 0.0500 mg/kg, 카드뮴은 0.00015 ∼ 0.1240 mg/kg, 비소는 0.0005 ∼ 0.8490 mg/kg, 알루미늄은 0.0150 ∼ 0.1452 mg/kg, 수은은 0.00004 ∼ 0.0006 mg/kg, 메틸수은은 0.0058mg/kg, 주석은 0.0002 ∼ 0.0021 mg/kg, 무기비소는 0.0002 ∼ 0.0178 mg/kg 이었다. 표준용액 첨가법과 CRM을 이용하여 회수율을 실험한 결과, 76.0 ∼ 114 %의 양호한 결과를 얻었으며, 중금속 분석법에 대한 측정불확도를 산출하였다.
식품 중 중금속 함량 실태조사 분석을 위하여 납, 카드뮴, 비소는 총 시료수 49,963건(농산물 14,683건, 축산물 10,160건, 수산물 10,680건, 가공식품 14,440건)을 수거하였다.
납의 경우, 전체의 평균 납 함량은 0.048 ± 0.298 mg/kg, 농산물의 평균 납 함량은 0.018 ± 0.085 mg/kg, 축산물의 평균 납 함량은 0.041 ± 0.498 mg/kg, 수산물의 평균 납 함량은 0.124 ± 0.383 mg/kg, 가공식품의 평균 납 함량은 0.025 ± 0.105 mg/kg 이었다.
카드뮴의 경우, 전체의 평균 카드뮴 함량은 0.291 ± 2.618 mg/kg, 농산물의 평균 카드뮴 함량은 0.015 ± 0.058 mg/kg, 축산물의 평균 카드뮴 함량은 0.001 ± 0.005 mg/kg, 수산물의 평균 카드뮴 함량은 1.307 ± 5.545 mg/kg, 가공식품의 평균 카드뮴 함량은 0.023 ± 0.083 mg/kg 이었다.
비소의 경우, 전체의 평균 비소 함량은 1.092 ± 4.065 mg/kg, 농산물의 평균 비소 함량은 0.020 ± 0.235 mg/kg, 축산물의 평균 비소 함량은 0.009 ± 0.033 mg/kg, 수산물의 평균 비소 함량은 4.594 ± 7.403 mg/kg, 가공식품의 평균 비소 함량은 0.353 ± 2.233 mg/kg 이었다.
알루미늄의 경우, 총 시료수 49,427건(농산물 14,147건, 축산물 10,160건, 수산물 10,680건, 가공식품 14,440건)을 수거하였으며, 전체의 평균 알루미늄 함량은 16.971 ± 133.542 mg/kg, 농산물의 평균 알루미늄 함량은 10.352 ± 38.384 mg/kg, 축산물의 평균 알루미늄 함량은 0.783 ± 2.596 mg/kg, 수산물의 평균 알루미늄 함량은 25.783 ± 87.337 mg/kg, 가공식품의 평균 알루미늄 함량은 28.334 ± 231.391 mg/kg 이었다.
수은의 경우, 총 시료수 50,137건(농산물 14,673건, 축산물 10,160건, 수산물 10,855건, 가공식품 14,449건)을 수거하였으며, 전체의 평균 수은 함량은 0.014 ± 0.072 mg/kg, 농산물의 평균 수은 함량은 0.002 ± 0.006 mg/kg, 축산물의 평균 수은 함량은 0.002 ± 0.003 mg/kg, 수산물의 평균 수은 함량은 0.054 ± 0.148 mg/kg, 가공식품의 평균 수은 함량은 0.004 ± 0.012 mg/kg 이었다.
메틸수은의 경우, 총 시료수 1,543건(수산물 1,257건, 수산물 가공식품 286건)을 수거하였으며, 전체의 평균 메틸수은 함량은 0.165 ± 0.362 mg/kg, 수산물의 평균 메틸수은 함량 0.199 ± 0.393 mg/kg 이었으며, 수산물 가공식품의 평균 메틸수은 함량 0.015 ± 0.032 mg/kg 이었다.
주석의 경우, 캔제품 699건에 대하여 평균 주석 함량 1.829 ± 18.339 mg/kg 이었다.
무기비소의 경우, 총 시료수 1,772건(농산물 300건, 수산물 961건, 가공식품 511건)을 수거하였으며, 전체의 평균 무기비소 함량은 0.183 ± 0.911 mg/kg, 농산물의 평균 무기비소 함량은 0.073 ± 0.020 mg/kg, 수산물의 평균 무기비소 함량은 0.293 ± 1.224 mg/kg, 가공식품의 평균 무기비소 함량은 0.038 ± 0.090 mg/kg 이었다.
우리나라 국민 전체의 대상 물질에 대한 노출 수준을 파악하기 위해 위해평가를 실시하여 대상 식품에 대한 일일섭취량과 위해 분포결과를 산출하였다. 대상 식품군은 총 525개 품목으로 농산물 179개 품목, 축산물 48개 품목, 수산물 125품목, 가공식품 173품목에 대한 물질별 위해도를 추정하였다.
납의 경우, 전연령 노출안전력은 1이상이나, 영유아 및 어린이의 노출안전력(MOE) 1에 근접한 것으로 조사되었다. 전연령에서 농산물, 가공식품, 수산물 순으로 노출 기여가 높은 것으로 나타났으며, 두족류는 식품 함유 농도에 의한 노출 기여가 높은 식품으로 평가되었다.
카드뮴의 경우, 전연령의 평균 일일노출량은 0.322 ug/kg-day 수준이었고, PTMI(25 ug/kg-month) 대비 평균 위해도는 38.6%로 나타났다. 전연령에서 수산물, 농산물, 가공식품 순으로 노출 기여가 높게 나타났으며, 두족류, 패류, 조미김은 식품 함유 농도에 의한 노출 기여가 높은 식품으로 평가되었다.
비소의 경우, 전연령의 평균 일일노출량은 2.863 ug/kg-day 수준이었고, PTWI(350 ug/kg-week) 대비 평균 위해도는 5.7%로 나타났다. 전연령에서 수산물, 가공식품, 농산물 순으로 노출 기여가 높게 나타났으며. 일반어류, 두족류, 갑각류, 패류, 해조류, 조미김은 식품 함유 농도에 의한 노출 기여가 높은 식품으로 평가되었다.
알루미늄의 경우, 전연령의 평균 일일노출량은 127.931 ug/kg-day 수준이었고, TDI(300 ug/kg-day) 대비 평균 위해도는 42.6 %로 나타났다. 전연령에서 가공식품, 농산물 순으로 노출 기여가 높게 나타났으며, 면류, 커피류는 높은 식이량으로 인해 상위 노출 식품으로 평가되었다.
수은의 경우, 전연령의 평균 일일노출량은 0.085 ug/kg-day 수준이었고, PTWI(3.7 ug/kg-week) 대비 평균 위해도는 16.0%로 나타났다. 전연령에서 수산물, 가공식품, 농산물 순으로 노출 기여가 높게 나타났으며, 다량어류 및 새치류와 같은 심해성 어류는 식품함유 농도에 의한 노출 기여가 높은 식품으로 평가되었다.
메틸수은의 경우, 전연령의 평균 일일노출량은 0.020 ug/kg-day 수준이었고, PTWI(2.0 ug/kg-week) 대비 평균 위해도는 6.8%로 나타났다. 어류, 다랑어류 및 새치류는 오염도에 의한 기여가 높았으며, 어육가공품은 높은 식이량으로 인해 상위 노출 식품으로 평가되었다.
무기비소의 경우, 전연령의 평균 일일노출량은 0.260 ug/kg-day 수준이었고, PTWI(9 ug/kg-week) 대비 평균 위해도는 20.2%로 나타났다. 즉석편의식품, 과자류는 오염도에 의한 기여가 높았으며, 곡류, 면류는 높은 식이량으로 인해 상위 노출 식품으로 평가되었다.
총 주석의 경우, 전연령의 평균 일일노출량은 0.139 ug/kg-day 수준이었고, PTDI(2,000 ug/kg-day) 대비 평균 위해도는 0.007%로 식품 기준 규격 강화에 의한 위해 관리가 필요한 항목에 부합되지 않았으며, 각 연령군별 위해도도 모두 0.1% 미만의 낮은 수준으로 평가되었다.

This study conducted the research on the status of heavy metals and risk assessment through the creation of monitoring data required for periodic safety assessment of heavy metals in food based on the 3-year plan for food and drug safety advancement (reassessment of hazardous substances etc. in food). A survey of domestic and foreign literature on the status of heavy metals in food was carried out. In addition, analytical method verification and measurement uncertainty were yielded through standardization of monitoring system such as sampling, analysis method, DB management standardization along with certified reference material (CRM).
5 heavy metals (lead, cadmium, arsenic, mercury, aluminum) for agricultural, marine, livestock products, processed food, and special purpose food and methyl mercury, tin and inorganic arsenic for some samples were analyzed. In order to analyze the contents of lead, cadmium, arsenic, aluminum and tin, a test method measured by ICP-MS or ICP-OES was established after pre-treatment through microwave apparatus. The analytic methods such as mercury automatic analyzer for mercury, GC-ECD for methyl mercury and HPLC-ICP-MS for inorganic arsenic have been established.
The R2 value of the calibration curve for all heavy metals was 0.999 or higher. In terms of the detection limit of heavy metals, agricultural products was 0.0002 ∼ 0.0059 mg/kg for lead, 0.0001 ∼ 0.0030 mg/kg for cadmium, 0.0002 ∼ 1.0000 mg/kg for arsenic, 0.0031 ∼ 1.5000 mg/kg for aluminum, 0.00003 ∼ 0.0003 mg/kg for mercury.
Livestock products was 0.0002 ∼ 0.3000 mg/kg for lead, 0.0001 ∼ 0.4000 mg/kg for cadmium, 0.0002 ∼ 0.025 mg/kg for arsenic, 0.005 ∼ 0.025 mg/kg for aluminum, 0.00004 ∼ 0.018 mg/kg for mercury, 0.0013 ∼ 0.012 mg/kg for methyl mercury, 0.011 mg/kg for inorganic arsenic. Marine products was 0.0001 ~ 0.025 mg/kg for lead, 0.0001 ∼ 0.0125 mg/kg for cadmium, 0.0002 ∼ 0.025 mg/kg for arsenic, 0.005 ∼ 0.025 mg/kg for aluminum, 0.00004 ∼ 0.018 mg/kg for mercury, 0.0013 ∼ 0.012 mg/kg for methyl mercury, 0.011 mg/kg for inorganic arsenic.
Processed food was 0.00024 ∼ 0.0500 mg/kg for lead, 0.00015 ∼ 0.1240 mg/kg for cadmium, 0.00052 ∼ 0.8490 mg/kg for arsenic, 0.015 ∼ 0.1452 mg/kg for aluminum, 0.00004 ∼ 0.00057 mg/kg for mercury, 0.0058 mg/kg for methyl mercury, 0.00018 ∼ 0.00208 mg/kg for tin, and 0.00024 ∼ 0.01784 mg/kg for inorganic arsenic. Using method of standard addition and CRM to test recovery rate, we attained good rates of 76.0 ∼ 114 %, and we calculated uncertainty of measurement in our analytical methods for heavy metals.
To analyze heavy metal content in foods, we collected 49,963 samples (14,683 agricultural products, 10,160 livestock products, 10,680 marine products, and 14,440 processed foods) for lead, cadmium, arsenic.
For lead, average lead content of total products was 0.048 ± 0.298 mg/kg, average lead content of agricultural products was 0.018 ± 0.085 mg/kg, average lead content of livestock products was 0.041 ± 0.498 mg/kg, average lead content of marine products was 0.124 ± 0.383 mg/kg, and average lead content of processed foods was 0.025 ± 0.105 mg/kg.
For cadmium, average cadmium content of total products was 0.291 ± 2.618 mg/kg, average cadmium content of agricultural products was 0.015 ± 0.058 mg/kg, average cadmium content of livestock products was 0.001 ± 0.005 mg/kg, average cadmium content of marine products was 1.307 ± 5.545 mg/kg, and average cadmium content of processed foods was 0.023 ± 0.083 mg/kg.
For arsenic, average arsenic content of total products was 1.092 ± 4.065 mg/kg, average arsenic content of agricultural products was 0.020 ± 0.235 mg/kg, average arsenic content of livestock products was 0.009 ± 0.033 mg/kg, average arsenic content of marine products was 4.594 ± 7.403 mg/kg, and average arsenic content of processed foods was 0.353 ± 2.233 mg/kg.

For tin, average tin content of 699 canned products was 1.829 ± 18.339 mg/kg. For inorganic arsenic, average mercury content of total products(1,772) was 0.183 ± 0.911 mg/kg, average inorganic arsenic content of 300 agricultural products was 0.073 ± 0.020 mg/kg, average inorganic arsenic content of 961 marine products was 0.293 ± 1.224 mg/kg, and average inorganic arsenic content of 511 processed foods was 0.038 ± 0.090 mg/kg.
Based on Korean dietary habits, risk assessment were conducted for commonly consumed foods including estimated daily intake. The target food groups were 525 items, and the risk for each item was estimated for 179 items of agricultural products, 48 items of meats products, 125 items of daily products, and 173 items of processed foods.
In the case of lead, all age groups MOE are more than 1, but close to MOE 1 for infants and children. All age groups were found to have higher exposure contribution in the order of agricultural products, processed foods and daily products, and cephalopods were evaluated as foods with high exposure contribution by food containing concentration.
In the case of cadmium, the average daily dose of all age groups was 0.322 ug / kg-day, and the average risk was 38.6% over PTMI(25 ug / kg-month). All age groups were found to have higher exposure contribution in the order of daily products, agricultural products and processed foods, and cephalopods, shellfishes, and seasoned laver were evaluated as foods with high exposure contribution by the food containing concentration.
In the case of arsenic, the average daily exposure of all ages was 2.863 ug / kg-day and the average risk was 5.7% compared to PTWI(350 ug / kg-week). All age groups were found to have higher exposure contribution in the order of daily products, processed foods and agricultural products, and general fish, cephalopods, seaweeds, seasoned laver, and processed marine products were evaluated as foods with high exposure contribution by food containing concentration.
In the case of aluminum, the average daily exposure of all ages was 127.931 ug / kg-day, and the average risk was 42.6% of TDI(300 ug / kg-day). All age groups were found to have higher exposure contribution in the order of processed foods and agricultural products, and noodles, and coffee were rated as high-exposure foods due to high dietary levels.
In the case of mercury, the average daily exposure for all ages was 0.085 ug / kg-day and the average risk was 16.0% of PTWI(3.7 ug / kg-week). All age groups were found to have higher exposure contribution in the order of processed foods and daily products, and deep sea fish such as tunas/billfishes were evaluated as a food with high exposure contribution by the food containing concentration.
In the case of methylmercury, the average daily exposure for all ages was 0.020 ug / kg-day, and the average risk was 6.8% over PTWI (2.0 ug / kg-week).
Fish, tunas and billfishes were higher exposure contribution and fish meat products were rated as high-exposed foods due to high dietary levels.
In the case of inorganic arsenic, the average daily exposure for all ages was 0.260 ug / kg-day, and the average risk was 20.2% of PTWI (9 ug / kg-week). Deep instant food, and confection were higher exposure contribution and Grains and noodles were rated as high-exposed foods due to high dietary levels.
In case of tin, the average daily exposure for all ages was 0.139 ug / kg-day, and the average risk was 0.007% compared to PTDI (2,000 ug / kg-day), and then it did not meet the items requiring risk management by strengthening the food standard, and the risk for each age group was evaluated to be as low as less than 0.1%.
