본 연구에서는 “식중독균/축산식품” 조합에 대해 risk profile 개발, “식중독균/축산식품” 조합 위험순위 결정, 축산식품에서 식중독균의 증식/생존특성 분석, 주요 축산식품 HACCP 공정효과 검증 등을 통해 도축 후 소비단계까지 안전한 미생물관리 옵션(기준규격, 저감화 목표치) 개발의 과학적 근거자료를 제시하여 안전한 축산식품 유통에 기여하고자 하였다. 본 연구결과를 통해 축산식품의 중장기적 위해평가 방향 및 우선관리순위를 제시하였다. 또한 국내 축산식품을 대상으로 “식중독균/축산식품” 조합의 위험 순위 결정 도구개발을 위
본 연구에서는 “식중독균/축산식품” 조합에 대해 risk profile 개발, “식중독균/축산식품” 조합 위험순위 결정, 축산식품에서 식중독균의 증식/생존특성 분석, 주요 축산식품 HACCP 공정효과 검증 등을 통해 도축 후 소비단계까지 안전한 미생물관리 옵션(기준규격, 저감화 목표치) 개발의 과학적 근거자료를 제시하여 안전한 축산식품 유통에 기여하고자 하였다. 본 연구결과를 통해 축산식품의 중장기적 위해평가 방향 및 우선관리순위를 제시하였다. 또한 국내 축산식품을 대상으로 “식중독균/축산식품” 조합의 위험 순위 결정 도구개발을 위한 입력변수 및 주요지 표점수를 설정하였으며 주요 연구결과는 다음과 같다.
● 위해물질/식품 조합의 위해평가 및 관리를 위한 배경정보를 제공하기 위하여 국내·외 문헌 자료 분석을 통해 다음 6개 식중독균/축산식품조합 risk profile을 개발하였음: ① Campylobacter spp in poultry ② Salmonella in poultry and egg ③ Pathogenic E coli (EHEC) in meat and ground meat ④ Cl. Perfringens in livestock products ⑤ L. monocytogenes in milk and egg © S. aureus in milk and egg.
● 축산식품 식중독발생 통계자료, 전문가 설문조사, risk ranger 결과를 종합하여 식중독 발생 가능성을 고려하고 위해평가 및 관리가 필요한 우선순위에 따라 세 그룹(I, II, III)으로 분류함. 그룹 I 에는 Salmonella spp./식용란 및 알가공품, Campylobacter spp./계육, Pathogenic E. coli/식육, 분쇄가공육, 그룹 II에는 Cl. perfringens/식육, 소시지 및 햄류, S. aureus/식육, 소시지 및 햄류, Salmonella spp./식육, E. sakazaki/조제분유, 그룹 III 에는 L. monocytogenes/식육, 소시지 및 햄류, Norovirus/육류조리식품, Shigella spp./식육, S. aureus/알가공품, 우유, L. monocytogenes/알가공품, 우유, 치즈류, B. cereus/조제우유, 육류조리식품 등이 포함되었음.
● 축산식품에서 식중독균의 증식/생존특성 분석연구결과 훈제오리, 치킨너겟에서 C. jejuni, 일반/저지방우유, 메추리알에서 L. monocytogenes, S. aureus, Cl. perfringens, 알가열성형제품 (오믈렛, 계란구이, 계란찜 등)에서 Salmonella(S. Typhimurium & S. Enteritidis), L. monocytogenes, E. coli, 냉동 분쇄가공육에서 EHEC에 대해 초기오염수준, 보관온도에 따라 증식/생존가능성을 분석한 결과, 훈제오리 및 치킨너겟의 경우 현재의 유통 조건인 냉장 및 냉동조건에서 C. jejuni 증식가능성이 없으나 C. jejuni의 생존 위험성을 고려할 때 식육/식육가공품에서 C. jejuni의 위해평가를 통해 C. jejuni에 대한 기준규격 설정이 필요함. 반면 냉장온도에서 유통되는 우유, 메추리알에서 L. monocytogenes 및 S. aureus의 증식위험은 매우 커 불검출로 관리가 필요하나 10°C에서 유통·판매되는 우유 및 메추리알에서 Cl. perfringens에 증식위험성은 없는 것으로 판단되어 현재의 불검출 기준을 완화하여도 무리가 없을 것으로 사료됨.
● 또한 알가열성형제품으로 오믈렛(치즈, 참치, 떡갈비, 피자), 계란구이, 계란찜 등에서 냉장보관 시 Listeria 증식, Salmonella 생존/증식을 고려 반드시 음성기준으로 관리되어야 함. 특히 Salmonella는 알가열성형 제품에서 증식속도에 균주의 차이를 보였는데 S. Enteritidis는 S. Typhimurium보다 더 빠르게 증식하여 알 가공품에서 S. Enteritidis의 리스크가 높은 것으로 확인됨. 장출혈성대장균(EHEC)의 경우도 냉동분쇄육에서의 생존력을 고려할 때 반드시 불검출 기준규격으로 관리되어야하며 공정 최종단계의 성과목표도 0.04 CFU/g 이하로 설정이 필요하며 분쇄가공육 조리 시소비자들에게 적절한 조리방법을 반드시 준수하도록 식품표시 사항에 강조할 필요성이 있음.
● 분쇄가공육 제품에서 HACCP 공정효과를 검증한 결과 분쇄가공육의 경우 CCP의 살균공정에서 살균온도와 시간에 대한 업체의 기준설정이 적절함을 검증했으나, 냉동분쇄가공육에서 장출혈성 대장균(EHEC)의 베로톡신 1(VT1, 햄버거 패티, 떡갈비)과 베로톡신 2(VT2, 돈가스)가 양성으로 검출되고 냉동 시 매우 강한 생존력을 보여 분쇄가공육에서 장출혈성대장균 (EHEC)에 대한 기준 설정이 필요함. 또한 축산 식품 중 연구가 많이 진행되지 않은 액란가공공정의 HACCP 공정을 검증한 결과 액란 살균 공정에서의 살균온도와 시간에 대한 업체의 기준설정이 미생물을 관리하기에 적절하지 못한 것으로 판단되었으며 향후 액란 공정의 CCP에서 온도와 시간 관리를 재검토 하는 연구가 필요할 것으로 사료됨.
● 국내 축산식품 관련 연구자료, 국내외 축산식품관련 식중독 통계자료를 종합 분석하고 국외 risk ranking tool의 입력변수를 비교 분석하여 국내 축산식품 위해순위 결정 도구 변수 및 주요지표점수를 결정하였음. 결정된 변수는 ①식중독 발생가능성, ②위해심각성, ③인구집단 민감성, ④식품섭취빈도, ⑤식품소비인구집단 비율, ⑥유통단계에서 유해 미생물 오염발생빈도(원재료, 가공식품 구분), ⑦유해미생물 성장가능성, ⑧유통관리의 효과, ⑨소비자의 최종 처리방법을 포함하여 모두 9개의 변수를 설정하고 각각의 변수에 대해 변수세부항목 및 각 항목의 중요성에 따라 주요지표점수(score)를 결정하여 축산식품에서 위해순위 결정을 위한 도구 개발의 실용화가 가능하도록 하였음.
● 결론적으로 본 연구결과는 축산식품 위해평가에 대한 중장기적인 방향을 제시하고 우선관리 대상을 결정하는데 유용한 정보를 제공할 것으로 사료됨.

The objective of this study was to provide the scientific evidence for microbiological control guideline for safe distribution of livestock products from farm to table. In this study, we have developed risk profiles of six microbial hazards and determined risk ranking of the combination of pathogen-livestock products. The result of this study will provide a useful information for the risk management prioritization of the combination of pathogen and livestock product. The characteristics of growth and survival of pathogens in livestock product were also evaluated and HACCP plan was verified. In addition, we have determined the input variables and criteria score for development of risk ranking tool for the combination of foodborne pathogen-livestock products in Korea. The major results of the study are summarized as follows.
● We have prepared six risk profiles of foodborne pathogen-livestock product pairing throughout the literature review of national and international studies as follows; ① Campylobacter spp. in poultry ② Salmonella in poultry and egg ③ Pathogenic E coli (EHEC) in meat and meat products ④ Clostiridum perfringens in livestock products ⑤ Listeria monocytogenes in milk and egg © Staphylococcus aureus in milk and egg (refer to supplement).
● We have classified the combination of pathogen-livestock products into group I, II, and III according to the extent of likelihood and severity of risk after review foodborne outbreak data related to livestock products, experts' opinion about the risk of pathogen-livestock product pairing and the outcome of risk ranger(semiquantitative risk ranking tool), Group I includes Salmonella spp./egg and egg products, Campylobacter spp./poultry, pathogenic E. coli/meat and processed ground meat. Group II includes Cl. perfringens/meat, sausage and ham, S. aureus/meat, sausage and ham, Salmonella spp/meat, E. sakazaki/ infant formula, and Group III includes L. monocytogenes/meat, sausage and ham, Norovirus/cooked meat products, Shigella spp./meat, S. aureus/processed egg, milk, L. monocytogenes/processed egg, milk, and cheese, B. cereus/formula milk, cooked meat products.
● According to the result of HACCP verification of frozen, processed ground meat, critical limit at the CCP was sufficient for intended purpose. However, microbial criteria for Enterohemorrhagic Escherichia coli (EHEC) in processed ground meat is needed due to its great survival ability during frozen storage and high contamination rate of EHEC verotoxin 1(VT1) and 2(VT2) in frozen pattie, dduk-galbis and pork cutlet. In addition, critical limit (temperature and time) was not sufficient to control the microbial hazard of processed liquid egg according to the monitoring data of microbial hazard in sample after CCPs. Thus a scientific or technical review of the critical limits is necessary in the future study to verify that the specifications that are set are adequate to control the hazards in processed liquid egg.
● According to the results of characteristics of growth and survival of pathogens in various livestock products, the growth of C. jejuni growth was not observed in smoked duck and chicken nugget during refrigerated and frozen storage, but microbial criteria of C. jejuni is needed in poultry and poultry processed products due to the risk of its survival and low infectious dose. L. monocytogenes and S. aureus in milk and quail’s egg must be controlled as "negative" because they grew so rapidly at refrigerated temperature, but no growth of C. perfringens in milk and quail’s egg was observed at the temperature below 10°C. Listeria and Salmonella in egg products such as omelet, baked egg, and steam egg should be also controlled as "negative” in consideration of their growth and survival at refrigerated temperature. More rapid growth of S. Enteritidis than S. Typhimurium was observed in egg processed products, indicating the greater risk of S. Enteritidis in egg processed products. Enterohemorrhagic Escherichia coli(EHEC) in frozen processed ground meat must be controlled as "negative" as well, due to its great survival ability and thus its performance objective at the end of the manufacturing process should be less than 0.04 CFU/g.
● We have determined input variables and criteria score for development of "risk ranking tool" for the combination of foodborne pathogen-livestock product in Korea and thus the risk ranking tool can be practicalized. The determined variables include ①likelihood of occurrence, ②hazard severity, ③susceptible population, ④frequency of consumption, ⑤ proportion of population consuming the product, ⑥prevalence of contamination at retail (raw meat or processed meat), ⑦growth potential, ⑧effectiveness of post processing control system, and ⑨consumer practices. The criteria score was varied according to the importance and impact of each variables.
● In conclusion, the results of this study will provide the direction of mid-and long-term planning for risk assessment and risk management prioritization of livestock products
