○ 총 19종의 인간 배아줄기세포주(CHA-hES 43~61)를 수립하고, 이중 16종의 인간 배아줄기세포주를 줄기세포 등록제에 등록 완료함.
○ 성광의료재단의 미국 연구팀 및 하버드대학 연구팀와의 공동연구를 통하여 총 14종의 인간 체세포복제배아줄기세포주를 수립하고 이중 12종을 국내의 줄기세포 등록제에 수입줄기세포주로 등록을 완료함.
○ 총 54건의 인간 배아줄기세포주 및 체세포복제배아줄기세포주 분양과 총 23건의 인간 유도만능줄기세포주 분양을 하였고, 줄기세포 등록제와 관련하여 질병관리본부에 총 35건의 인간 배아줄

○ RNA-seq.을 이용한 배아줄기세포주 특성 분석을 위하여 30종 이상의 인간 배아줄기세포주 샘플을 준비하고, 일차적으로 배아줄기세포주 2종과 유도만능줄기세포주 1종, 동일한 체세포에서 확립된 체세포복제배아줄기세포주 2종과 유도만능줄기세포주 2종, 체세포 2종에서 RNA-seq.을 이용한 total RNA 발현 양상을 비교함.
○ 생쥐 정원줄기세포와 중간단계 및 다분화능을 가지게 된 정원줄기세포 간의 comparative transcriptome analysis를 통해 전분화능 조절인자 후보 유전자군을 발굴하고, 이들 중 PC4와 Ruvbl2 유전자의 기능을 분석함.
○ 인간 전분화능 줄기세포에서 일반적인 염색체검사(G-banding) 및 MLPA, SNP-array 등을 이용한 유전적 안정성 확인 기법을 확립함.
○ 인간 유래 지지세포주 개발을 위하여, 1종의 인간 제대혈 유래 세포와 5종의 인간 정소 유래 세포(human testicular stromal cell, hTSC), 총 9종의 피부 유래 세포 (dermal skin fibroblast cell, hDF)를 primary culture를 통하여 배양하여 동결 보관함.
○ 효율적인 인간 배아줄기세포 배양방법과 동결방법을 개발하여 인간 배아줄기세포주를 동결 보관함.

Purpose : This study is carried out to the following purposes: establishment of human embryonic stem cells verified by standardized genetic stability test and characterization test, development of efficient culture protocol for human embryonic stem cells, improvement of human embryonic stem cell cryopreservation protocol, mass production of qualified human embryonic stem cells, analysis of gene expression profiles among various hESC lines using RNA seq., immunological characterization of human embryonic stem cells between undifferentiation and differentiation state, and maintenance and distribution of quality-controlled human embryonic stem cells.

Results
○ A total of 19 human embryonic stem cell lines (CHA-hES 43 ~ 61) were established for 5 years and 16 of them were registered in the stem cell registry.
○ A total of 14 human SCNT-ESC lines were established through a research collaboration with the US research team of SungKwang Medical Foundation and/or Prof. Yi Zhang of Harvard University. And, 12 of them were registered as imported stem cell lines in the stem cell registry (CHA-hES NT2, NT4, NT5~NT8, NT10, NT13, NT15, NT16, NT101, NT102).
○ Fifty four distribution cases of hESCs and SCNT-hESCs. Twenty three distribution cases of hiPSC. Thirty five offer cases of hESCs related to register stem cells in the stem cell registry .
○ Publication of 9 research papers (1-domestic journal (non-SCI); 1-domestic journal (SCI); 7-international journals (SCI) including 'Cell Stem Cell')
○ Preparation of more than 30 samples of hESC lines for RNA-seq. Comparative analysis of gene expression profiles among 2 hESC lines, 1 hiPSC line, two somatic cells, and two SCNT-hESC lines and 2 hiPSC lines established from the same somatic cells using RNA-seq.
○ A comparative transcriptome analysis among mouse spermatogonial stem cells, intermediate spermatogonial stem cells, and multipotent spermatogonial stem cells in order to identify candidate genes for the pluripotency regulation. Among the candidate genes, the functions of PC4 and Ruvbl2 genes were analyzed.
○ Application of MLPA on genetic stability test during hESC cultivation. Confirmation of the trisomy rescue phenomenon, which trisomy turns into normal disomy, during cultivation of hESCs.
○ Comparative analysis of genetic stability among SCNT-hESCs, iPSCs and somatic cells with same genetic background using SNP-array.
○ Differentiation and characterization of retina pigment epithelium (RPE) cells from SCNT-hESCs and iPSCs derived from the same somatic cells including AMP patient
○ Establishment and cryopreservation of 15 human fibroblast cells derived from donated umbilical cord, testis and dermal skin tissue as human feeder cells

Expected Contribution
○ Establishment and distribution of human embryonic stem cells verified by standardized genetic stability test and characterization test would be brought to establish the infrastructure of stem cell research and to prepare to industrialize.
○ Mass production of qualified hESCs by standardized system for hESC cultivation and cryopreservation would facilitate stem cell-related research in Korea.
○ SCNT-hESCs and hiPSCs with the same genetic background are very useful materials to comparative study for the pluripotency and reprogramming mechanism.
○ The genetic stability of hESCs can be confirmed and verified by the evaluation system of hESC genetic stability using molecular genetic methods.
○ The results from comparative analysis for characteristics of the various hESC lines is possible to use hESCs more suitable for the purpose of the study and to increase the efficiency of the study.
○ Human ESCs are the most effective materials for in vitro toxicity and efficacy test in the development of cell therapy and in the development of new drugs.
