연구의 목적 및 내용
최근 10여년 동안에 한국 연안에서는 독성해파리의 출현이 크게 증가하는 추세에 있고, 그와 함께 해파리 쏘임의 피해로 때때로 심각한 문제가 되고 있으나 이들 해파리의 독소 성분에 대한 정보가 크게 부족하여 적절한 대처가 매우 어려운 실정이다. 따라서 본 연구에서는 한중일 해역에서 가장 큰 피해를 주고 있는 노무라입깃해파리로 부터 venom metalloproteinase를 비롯한 독소 성분의 분리 동정 및 그 독소 유전자를 cloning하고 독소단백질의 구조와 작용의 기작을 규명하고자 한다. 이 연구는 향
연구의 목적 및 내용
최근 10여년 동안에 한국 연안에서는 독성해파리의 출현이 크게 증가하는 추세에 있고, 그와 함께 해파리 쏘임의 피해로 때때로 심각한 문제가 되고 있으나 이들 해파리의 독소 성분에 대한 정보가 크게 부족하여 적절한 대처가 매우 어려운 실정이다. 따라서 본 연구에서는 한중일 해역에서 가장 큰 피해를 주고 있는 노무라입깃해파리로 부터 venom metalloproteinase를 비롯한 독소 성분의 분리 동정 및 그 독소 유전자를 cloning하고 독소단백질의 구조와 작용의 기작을 규명하고자 한다. 이 연구는 향후 독성해파리 쏘임에 의한 피해를 예방하거나 치료할 수 있는 방안을 제시할 수 있을 것이라 생각한다. 더 나아가 해파리 독소를 이용한 항혈전치료제 및 항암제 개발의 가능성을 모색할 계획이다.
연구결과
우리나라 연근해의 주요종인 노무라입깃해파리를 이용하여 이의 단백 독소에 대한 연구를 진행하였다. 특히 우리나라에 출몰하는 노무라입깃해파리의 단백 활성을 통해 Chymotrypsine, Phospholipase A2 (PLA2)및 Matrix Metalloprotease(MMP)의 존재가 확임됨에 따라 이들에 관련되어 있는 유전자 및 게놈 염기서열을 밝혔다. 노무라입깃해파리 serine protease의 속해있는 Chymotrypsin like-1 (CTRL)은 게놈의 길이가 2,434 뉴클레오티드를 가지며 4개의 exon으로 연결된 256개의 아미노산으로 구성되어 있었다. 노무라입깃해파리 Phospholipase A2 계열에서는 acidic PLA2 및 PLA2
PA3A/PA3B/PA5의 유전자 및 게놈 염기서열을 밝혔다. 우선 acidic PLA2의 경우 1,667 뉴클레오티드로, 3개의 exon으로 연결된 483개의 아미노산으로 구성되어 있었으며, PLA2 PA3A/PA3B/PA5는 1,366 뉴클레오티드의 3개의 exon으로 구성되어 있었다. 또한 498개의 아미노산으로 하나의 단백질을 만드는 것으로 나타났다. MMP의 경우 MMP17-1 및 MMP17-2 유전자의 염기서열을 완전히 분석하였는데, MMP17-1의 경우 1611 뉴클레오티드로 구성된 cDNA를 가지고 있었으며 537개의 아미노산로 구성되어 있었으며, MMP17-2는 1,578 뉴클레오티드와 525개의 아미노산을 가지고 있었다. 세균 및 곤충세포를 이용한 재조합단백질은 serine protease 효소 활성을 측정하여 발현을 확인하였다.
연구결과의 활용계획
지구온난화와 함께 해수 온도의 상승에 따라 우리나라에도 앞으로 많은 독성해파리의 출현이 예상되고 그로 인한 피해가 지속적으로 증가할 것으로 생각된다. 따라서 이들 해파리 독의 작용기작에 대한 연구가 필요하다. 더불어 해파리독의 독성작용을 조절할 수 있는 방법에 대한 연구를 진행할 예정이다. 이러한 연구결과는 독성해파리 쏘임에 의한 중독 작용에 대한 이해를 높여서 해수욕객의 쏘임 피해를 감소하거나 예방하는데 매우 긴요하게 활용할 수 있을 것이다. 또한 향후에는 해파리독을 이용한 항암제 후보물질 개발 등의 연구를 추진함으로써 바다 유해생물의 유익한 활용방안을 모색할 계획이다. 그럼으로써 해양생물의약 신소재 자원의 공동 DB 구축과 개발․생산 기반 구축을 통한 생물의약 기술의 축적으로 국가 경쟁력 제고와 더불어 연구조직의 활성화와 인프라 강화에 도움을 줄 것으로 생각된다.

However, a molecular mechanism of action of this jellyfish venom has been poorly understood. Our research goal is to identify a major toxic component (e.g., venom metalloproteinase) and its cDNA cloning from N. nomurai jellyfish venom. The illumination of its mechanism of action and protein structure can probably help preventing the intoxication or curing the stinging patients.
Furthermore, this study may pave way for the development of novel therapeutics, such as antithrombotic agent and anticancer agent using jellyfish venoms in the near future.
Result
N. nomurai venom, such as chymotrypsine, phospholipase A (PLA), and matix matalloprotease (MMP) activity showed by protein activity assay, inculde amidolytic, hemolysis etc. N. nomurai had chymotrypsine like-1 (CTRL-1) gene and they composed with 2,434 nucelotides for genome, 4 exons. 256 amino acid of CTRL-1 gene had chymotrypsine activity. N. nomurai venom contained that two isozymes of phospholipase A2 named acidic PLA2 and PLA2 PA3A/PA3B/PA5. acidic PLA2 had 1,667 necleotides for genome, 3 exons and 483 amino acid. PLA2 PA3A/PA3B/PA5 had 1,366 necleotdes for genome, 3 exons and 498 amino acid. In addition, N. nomurai venom had MMP, named MMP17-1 and MMP17-2. MMP17-1 composed with 1,611 necleotides for cDNA and 537 amino acid. MMP17-2, 1,578 neceleotides for cDNA and 525 amino acid. Recombinant proteins of CTRL-1 gene of N. nomurai has been successfully generated and confirmed using bacterial and insect cell expression system.
