● 중심체는 세포내 미세소관망의 중심점 역할을 담당하는데, 특히 세포분열 시에는 방추체로서 염색체를 딸세포로 끌고 간다. 섬모는 중심체로부터 유래된다.
● 중심체는 세포분열분야 뿐 아니라 줄기세포 유지 및 분화, 신경세포에서 섬모기능 등 다양한 분야에서 관심을 가지고 연구되고 있다.
● 본 과제에서는 세포생물학, 구조생물학, 생화학 및 개체생물학적 연구방법을 동원하여 중심체의 구조와 기능을 이해한다.

연구개발내용
● 중심립 유리 기전 연구: 중심체 유리와 연관된 유전자 발굴하고, 이를 생화학 및 세포 생물학적 방법으로 연구한다. 또한 중심립 유리에 관여하는 단백질 분해효소인 separase의 기질을 동정한다.
● 중심체의 세포주기 조절기전 연구: 중심체 기능을 조절하는 염색체복제 단백질들 및 상호작용하는 단백질을 탐색하여 생화학/구조생물학적 등의 기법을 사용하여 작용기작을 밝히고 중심체 내외부로의 신호전달 과정을 분석하고, 중심체, 세포주기와 염색체복제 조절의 상호연관성을 밝힌다.
● 중심체 단백질 분해기전 연구: 세포주기 의존적인 중심체 단백질들의 분해 기전을 연구하고자 한다. 또한 다양한 생리적 상황에서 중심체 단백질의 분해기전이 어떻게 조절되는지를 연구할 예정이다.
● 중심체 연결 단백질 구조 연구: 중심체는 단백질 중합체에 의해 연결되며 분리된다. 본 과제에서는 중심체 연결 중합체의 구성 단백질들을 생화학적, 세포학적, 구조생물학적, 그리고 개체생물학적 관점에서 접근할 계획이다. 또한 섬모형 신경세포에서 발현되는 특이 단백질 조절 인자를 규명하고 그 기능을 규명하고자 한다. 특히 dauer 단계에서의 기능에 초점을 맞추고자 한다
연구개발 성과
● 중심립 유리 기전 연구: Separase의 새로운 기질로서 pericentrin을 동정하고 중심립 유리과정에 기여도를 확인 한바, 새로운 기질 탐색 및 특성 연구를 완료함. PLK4의 새로운 기질로 CP110를 동정함. 논문 4편
● 중심체의 세포주기 조절기전 연구: DNA 복제 조절 단백질인 Cdc6 의 중심체로의 이동 확인함. TopBP1의 항체 제작관련 기술이전 라이센싱 체결함 (Millipore). 논문 2편, 기술이전 1건
● 중심체 단백질 분해기전 연구: CHFR 이라는 E3 ubiquitin ligase가 중심체 단백질의 분해를 조절하는 기전을 연구했음(AUKA, PLK1, NEK2). 섬모형성과정에서 탈아세틸화 효소의 중요성 확인. ISRIB drug을 이용한 eIF2α 인산화, stress granule 형성, 그리고 apoptosis의 상관관계 규명. 논문 4편
● 중심체 연결 단백질 구조 연구: 중심체 단백질의 결정화를 위해 최적의 조건을 연구 중이며, 세포생물학적 및 생화학적 방법을 통해 중심체로의 위치 및 중심체 연결 단백질 기능에 중요한 도메인을 확인하였음. 개체생물학적 관점에서 중심체 연구를 위해, C. elegans의 dauer시기 섬모형성과 관련한 다양한 돌연변이 제작을 완료함. 논문 2편

● Centrosome is a major microtubule organizing center and functions as a spindle pole during mitosis. Cilia can be formed from the centrosome in interphase cells.
● Importance of the centrosome is acknowledged not only in the cell biology but also in other research fields, such as stem cell biology and neurobiology.
● We propose a series of centrosome research from diverse aspects such as cell biology, protein biochemistry, structural biology, and integrative biology.

● Mechanism of centriole disengagement: To identify the genes related in the centriole disengagement, approaches of cell biology and biochemistry will be employed. In addition, a novel substrate of the separase can be revealed as a key molecule involved in centriole disengagement.
● Cell cycle-dependent regulation of the centrosome: Some proteins that play a role in DNA replication, also localize at the centrosome. Therefore, their roles in the centrosome might be related for the centriole replication to be limited to occur only in the S phase. Biochemical/ structural approaches would help to understand the mechanism of this relevant signaling pathway and how these proteins regulate the centrosomes.
● Mechanism of degradation of the centrosomal proteins: Degradation of cell cycle-dependent centrosomal proteins will be studied. Additionally, the regulatory mechanism of degradation under various circumstances would be examined.
● The structure of the centrosomal proteins responsible for centrosome cohesion: A pair of the centrosomes are linked to each other by polymeric protein networks, This project will explore the protein network using biochemical, cell biological, structural and ontological methods. Especially, the dauer-stage C.elegans will be the subject of the study to find the function of neuronal cilia.
Developement results
● Mechanism of centriole disengagement: Two of SCI pspers were published in 2015. In the papers, the pericentrin cleavage by separase was proved to be regulated by PLK1 and thus this event is critical for centriole disengagement. Secondly, upon Eg-5 inhibition, cells arrested at M phase were reported to show the centriole disengagement independently of the separase activity.
● Cell cycle-dependent regulation of the centrosome: TopBP1, which is a DNA damage response protein, has been reported to localize at the centrosome and the anti-serum of TopBP1 was licensed to Millipore, one of the major antibody companies.
● Mechanism of degradation of the centrosomal proteins: A new E3 ubiquitin ligase, CHFR is being examined for its role in regulating the centrosomal proteins. The ligase was proved to interact with many important kinases such as PLK1, AUKA, Nek2, which are known to regulate the centrosome function.
● Mechanism of degradation of the centrosomal proteins: Rootletin is the most prominent centrosomal linker protein. Throughout biochemical and cell biological studies, the domain responsible for the function as a centrosomal linker was found. Furthermore, the crystallization of the Rootletin domain is on optimization process.
