연구의 목적 및 내용
과채류의 재배 환경 인자와 과채류 표면 미생물과의 상관관계를 통하여 식품 유래 병원균의 생육 환경을 이해함으로써, 과채류에 존재하는 잠재적인 식품 유래 병원균 오염을 예측하고자 함.
▪ 과채류 표면의 식중독 원인균 검출을 위해 재배 환경(일반 & 유기농, 사계절, 수확 전과 후에 따른 과채류 시료 수집
▪ 과채류 표면 박테리아 및 진균의 메타지놈 유전체 분석을 통한 표면 미생물 분석
▪ 과채류 종류에 따른 식중독 병원균, 표면 박테리아 군집, 재배 방식 간의 상관성 분석 및 연관 네트워크
연구의 목적 및 내용
과채류의 재배 환경 인자와 과채류 표면 미생물과의 상관관계를 통하여 식품 유래 병원균의 생육 환경을 이해함으로써, 과채류에 존재하는 잠재적인 식품 유래 병원균 오염을 예측하고자 함.
▪ 과채류 표면의 식중독 원인균 검출을 위해 재배 환경(일반 & 유기농, 사계절, 수확 전과 후에 따른 과채류 시료 수집
▪ 과채류 표면 박테리아 및 진균의 메타지놈 유전체 분석을 통한 표면 미생물 분석
▪ 과채류 종류에 따른 식중독 병원균, 표면 박테리아 군집, 재배 방식 간의 상관성 분석 및 연관 네트워크 구축
▪ 구축된 식중독 병원균, 표면 미생물, 환경 인자와의 네트워크를 기반으로 선택적 제어법을 탐색
연구결과
▪ 과채류 표면 미생물체 탐색을 위해 22개 지역농장을 통해 총 66개 시료를 일반 & 유기농, 수확 전후, 재배지역, 질병발생에 따라 수집함.
▪ 과채류 표면 미생물의 메타지놈 유전체로부터 16S rRNA gene을 증폭, 대규모 염기 서열 분석법 454 pyrosequencing을 통해 염기서열 확보
▪ Gammaproteobacteria를 중심으로, Actinobacteria, Firmicutes, Bacteroidetes에 속하는 다양한 과채류 표면 미생물 종이 존재함을 확인함. 선택배지를 통해 3가지 주요 대장균군 및 식중독 원인균의 정량, 정성 분석을 실시함.
▪ 재배 방식(일반 & 유기농), 수확 전후, 병해충 발생에 따른 과채류 표면 미생물 군집 구조가 차이를 보였48으며, 내생/외생 인자 특이적 미생물 군 및 종 다양성이 확인됨.
▪ 과채류 종류별 표면 미생물체 비교를 통해, 과채류 특이적 미생물체 형성을 확인하고, 이에 영향을 미치는 재배환경 요인을 제시함.
▪ 다양한 환경인자 속에서도 주요 식중독 원인균은 검출한계 이하로 존재함을 확인함.
▪ 과채류 표면으로부터 식중독 병원균 증식을 제한하는 선택적 제어법을 탐색함.

We investigate the impact of agricultural factors and surrounding microbial communities on the development of food-borne pathogens on the surface of fresh produce, and predict food-borne microbial contamination of fresh produce. We further focus on selective and specific applications for chemical and biological sterilization of food-borne microbial contaminants using the networks among food-borne pathogens, agricultural factors, and surrounding microbial communities.
Result
▪ We collected 66 broccoli samples from 22 local farms, and classified according to farming region, plant growth, disease occurrence and farming practice.
▪ We extracted metagenomic DNAs, and obtained 16S rRNA gene sequence data using 454-pyrosequencing.
▪ The bacterial communities on broccoli leaves comprised Gammaproteobacteria, Actinobacteria, Firmicutes and Bacteroidetes. E. coli coliforms and foodborne pathogens were cultured using selective media, but foodborne pathogens were below limit of detection.
▪ We found that bacterial communities on broccoli leaves varied on farming regions, plant growth and disease occurrence, and identified specific bacterial taxa contributing to geography and host-related variations.
▪ We compared bacterial communities on broccoli leaves with those of11 fruits and vegetables, and found fresh produce-specific bacterial communities. Proximity to ground may be attributed to fresh produce-specific bacterial communities.
▪ We examined organic acids from microbial fermentation to reduce thw growth of foodborne pathogens on fresh produce as sanitizing agents.
Expected Contribution
▪ Descriptions of microbial communities on fresh produce according to environmental and host-related factors are needed to understand the development and alteration of phyllosphere microbiota.
▪ Investigation of microbial communities on fresh produce supports to understand the underlying mechanisms of specific growth of foodborne pathogens on fresh produce.
▪ Ultimately, microbe-microbe interactions could be further used to enhance food safety and quality.
▪ Considering an increase of organic agricultural industry, phyllosphere microbiota associated with fresh produce is further examined to predict influence on human health.
