□ 연구의 목적 및 내용
○ 본 연구는 하나의 발생 단계에서 존재하는 줄기세포의 snapshot을 연구하는 기존의 줄기세포 연구의 패러다임을 뛰어 넘어, 배아에서 성체까지 전체 생명과정에서 발생하는 다양한 줄기세포군들의 전사체 변화에 대한 종적 (longitudinal) 추적을 통하여, 배아발생 단계부터 성체조직 재생과 노화 과정에서 줄기세포 중요성과 이에 대한 분자학적 기전을 규명하는 재생의료 연구의 새로운 패러다임을 제시하는 것이 최종 목적임.

□ 연구의 목적 및 내용
○ 본 연구는 하나의 발생 단계에서 존재하는 줄기세포의 snapshot을 연구하는 기존의 줄기세포 연구의 패러다임을 뛰어 넘어, 배아에서 성체까지 전체 생명과정에서 발생하는 다양한 줄기세포군들의 전사체 변화에 대한 종적 (longitudinal) 추적을 통하여, 배아발생 단계부터 성체조직 재생과 노화 과정에서 줄기세포 중요성과 이에 대한 분자학적 기전을 규명하는 재생의료 연구의 새로운 패러다임을 제시하는 것이 최종 목적임.
○ 이를 위하여 배아줄기세포와 성체줄기세포 stemness 조절에 공통적으로 관여하는 신규 후성유전체 인자 발굴하고, 발굴한 후성유전체 인자의 단백질 번역 후 수정(post-translational modification, PTM) 탐색과 조절 네트워크를 규명하였음.
○ 구체적으로 1차년도에서는 배아부터 성체 노화까지 줄기세포군들의 전사체 변화를 분석하여 배아 및 성체줄기세포 공통 신규 후성유전체 인자 발굴 및 발굴한 인자의 기능조절 네트워크 탐색하고, 2차년도에서는 발굴한 배아 및 성체 줄기세포 공통 신규 후성유전체 인자들 네트워크를 활용하여 줄기세포 성상 조절 연구와 함께 치료용 줄기세포의 기능성 증진 기술을 개발하는 연구를 수행함.
□ 연구결과
○ 배아 및 성체조직 유래 줄기세포 (3종) 및 분화세포 (2종)의 총 15개의 전사체 데이터 베이스를 분석하여, 391개의 만능성 줄기세포 특이적 전사인자 발굴하였으며, 심층 연구를 위한 배아 및 성체 줄기세포 공통 전사 인자인 Tfcp2l1을 발굴함.
○ 전자 네트워크와 패스웨이 분석을 완료; 5개 후보 표적유전자 확립
○ 배아 및 성체줄기세포에서 Tfcp2l1의 기능 연구와 함께 Tfcp2l1을 조절하는 네트워크를 단백질 결합체 분석과 특이적 단백질 후 변헝 (PTM) 발굴 및 관련 조절 기전을 규명하였으며, 특히 Tfcp2l1 인산화 (site-X)와 촉매 효소 (pk-X)에 의한 배아줄기세포의 성상 (만능성, 분화능, 세포증식능) 조절 기전을 최초로 규명함.
○ Tfcp2l1 형질 전환 성체줄기세포주 확립과 이와 관련된 생착률 증진 기술을 개발하였으며, 개발된 줄기세포 기능성 강화 기술의 in vivo 유효성을 난치성 질환 모델 2종(폐동맥고혈압 & 간질성방광암)을 통하여 입증하였으며, 환자 시료 분석을 통하여 본 연구에서 도출한 전임상 연구 결과의 임상적 유의성을 입증함
○ 배아 및 성체줄기세포 공통 후성유전제 인자인 Sirt1 단백질의 Sirt1+/+ vs Sirt1-/- 마우스 배아줄기세포 (mESC) 전사체 및 후성유전체 다중오믹스 연구를 통하여, 줄기세포의 후성유전체 안전성 조절을 위한 신규 분자학적 기전을 규명함.
○ 연구 결과들은 SCI 급 논문 출판 7편 : Cell Rep (IF=8.282) & Circ Res (IF=13.965),Sci Rep (IF=4.259), Biochem Biophys Res Commun (IF=2.466), Int J Mol Med (IF=2.341), Histopathology (IF=3.523), Bioorg Med Chem Lett (IF=2.486), 비 SCI급 논문 출판 1편, 국내 특허 출원 2건과 해외 PCT 특허 출원 1건으로 보고함.
□ 연구결과의 활용계획
배아 발생부터 성체 노화까지 다양한 줄기세포군들의 전사체 변화의 체계적인 분석을 통한 배아 및 성체줄기세포 공통 신규 후성유전체 인자 발굴과 기능에 대한 본 연구는
• 체외 배양으로 발생하는 DNA 메틸화이상 등의 후성유전체 변이 제어를 통한 줄기세포 치료제의 안전성을 확보할 수 있는 기술적 근거를 마련함.
• 후성유전체 변이 제어 기술과 줄기세포 기능성 증진 기술은 재생 의학적 치료가 필요한 난치성/퇴행성 질환의 줄기세포치료제 고도화 기술로 활용이 가능하며, 줄기세포치료제에 대한 기능성 강화, 중개·임상연구 활성화와 함께 안전성 확보 기술발전과 줄기세포 치료법의 새로운 적응증을 확대가 가능함.
• 더불어 다양한 파이프라인 개발을 통하여 산업화를 선도하고 있는 국내 줄기세포 연구분야의 글로벌 리더쉽 확보전략으로 활용이 가능할 것으로 기대됨

The core purpose of this project is to identify the novel epigenetic factors which play a crucial role on regulating stem cells developed from embryo development to ageing through the longitudinal analysis of their transcriptome, protein interactome, and post- translational modification network. Thus, this study would not only advance our understanding of epigentic nature of stem cells through whole life, but also provide the novel strategy to develop the next generation stem cell therapeutics
To address this goal, we will first perform the in silico analysis of genome-wide gene expression database established from murine embryonic stem cells (ESCs),Oct4 expressing adult stem cells (VSELs), hematopoietic stem cells (HSCs), differentiated bone-marrow mononuclear cells (BM-MNCs) together their aged cell populations. This developmentally longitudinal analysis for stem cell transcriptome will provide the novel bio-markers, which can characterize the specific stage of stem cell development. To facilitate their functional study, we will develop the live-cell pluri-/multi-potency screening system employing the ESCs, which express the GFP under the lineage-specific promoter activity like gcOct4-GFP ESCs for pluripotency. In addition, we will manufacture the lenti-virus containing over-expressing cDNA or shRNA for the 1st selected genes and they will be applied to the GFP live pluri-/multi-potency screening system. To focus on epigenetic mechanism for stem cell biology, we will select in 1st screening system the bio-markers, which can characteristically regulate the epigenetic status or transcription gene ontology. To investigate their role on stem cell properties, we will manipulate the expression of candidate epigenetic biomarkers in murine embryonic stem cell-line, R1 ESC cells and human mesenchymal stem cells (MSCs) and we examine the effect of gene manipulation on their self-renewal, differentiation potential, cell proliferation, cell death and the molecular signature. To apply these results to developing the efficient ex vivo expansion protocol for adult stem cells,we will test the genetic manipulation of the epigenetic biomarkes in murine Oct4 expressing adult stem cells (VSELs) and human MSCs.
□ Result
Employing the transcription analysis and gene functional assays, we identified Tfcp21l as a transcription factor (TF) which is commonly involved in stemness of embryonic and adult stem cells. Ectopic expression and silencing of Tfcp2l1significantly affected the pluripotency, cell proliferation, developmental potency of murine ESC. The Tfcp2l1 protein strongly interacted with pluripotency assoicated TFs through N-terminal domain and also interacted with several proteins which regulate cell cycle, Particularly, the phosphorylation of Tfcp2l1 at site X is mediatedby protein kinase-X (PK-X) and this type of post-translational modification (PTM) of Tfcp2l1 regulated the stemness of murine ESCs. To extend the role of Tfcp2l1 into adult stem cells, we over-expressed human TFCP2L1 in human bone-marrow and umbilical cord derived MSCs and investigated their therapeuctic effects for treating pulmonary arterial hypertension which was induced by sub-cutaneous injection ofmononcrotaline. Furthermore, the beneficial therapeutic effects was also validated in an animal model of interstitial cystits which is an intractable bladder dysfunction disorder.
□ Expected Contribution
We expect that this proposal will not only advance our understating the epigenetic landscape of stem cell population but also contribute to establish the clinically best-suitable stem cell population. In addition, the knowledge from this proposal can be expanded to other biological research such as cancer biology and ageing researches.
