□ 연구의 목적 및 내용
탄소재료, 단분자 프로브, 또는 탄소재료-단분자 프로브를 조합하여 다양한 분석물질에 대한 센서 시스템을 구축하고자 함. 첫째로, 산화그래핀과 단분자 프로브를 융합하여 정전기적 상호작용 기반의 생체인산 화합물 센서를 개발하고, 생체인산에 대한 그래핀 기반의 전계효과트랜지스터(G-FET) 센서를 개발함. 둘째로, 탄소양자점을 이용한 전기화학발광 및 전기화학센서를 개발함. 셋째로, 정전기적 상호작용 기반의 단분자 프로브를 개발함. 넷째로, 탄소재료 기반 센서를 개발함. 마지막으로, 이리듐 착화합물 기반 전기

탄소재료, 단분자 프로브, 또는 탄소재료-단분자 프로브를 조합하여 다양한 분석물질에 대한 센서 시스템을 구축하고자 함. 첫째로, 산화그래핀과 단분자 프로브를 융합하여 정전기적 상호작용 기반의 생체인산 화합물 센서를 개발하고, 생체인산에 대한 그래핀 기반의 전계효과트랜지스터(G-FET) 센서를 개발함. 둘째로, 탄소양자점을 이용한 전기화학발광 및 전기화학센서를 개발함. 셋째로, 정전기적 상호작용 기반의 단분자 프로브를 개발함. 넷째로, 탄소재료 기반 센서를 개발함. 마지막으로, 이리듐 착화합물 기반 전기화학발광센서를 개발함.
□ 연구결과
(1) 산화그래핀과 단분자 프로브의 정전기적 상호작용을 이용한 센싱 시스템을 개발함. 이 센싱 시스템은 기존의 단분자 프로브에 비하여 PPi와 뉴클레오사이드 삼인산(NTPs)을 탁월하게 구별함. 많은 양의 ATP가 있는 환경에서도 소량의 PPi를 검출할 수 있었고 이 특성을 이용하여 중합효소 연쇄반응의 실시간 모니터링에 성공적으로 적용함.
(2) (Carbon dot)-Nax+ 이온 화합물을 구현하였고 전기화학발광센서 연구에 응용함. 이것은 전기화학 셀 내에서 이온전달 매개체 뿐만 아니라 공반응 화합물 역할을 하여 루테늄의 전기화학발광 현상을 유도하였음. 탄소양자점 이온젤은 합성하였고 전기화학촉매로서의 가능성을 확인하였으나 분석물질의 이온젤 내부로 확산 어려움 때문에 탄소양자점과의 전자교환이 원활하지 않아서 센서로서의 성능은 보이지 못함.
(3) 광산화를 이용하여 플라빈 프로브, 비소화합물에 대한 형광 프로브, 및 Cu(II)/Cu(I) 산화환원 변환을 통한 생체티올 프로브를 개발함.
(4) 탄소 나노튜브 기반 초박형 전자소자와 색 가변 유기발광 다이오우드 기반 디스플레이를 결합하여 심전도 센서를 개발함.
(5) ECL 발광 메카니즘에 기반한 분자 설계를 통하여 황화이온과 호모시스테인에 각각 선택적인 이리듐 착화합물 기반 전기화학발광 센서를 개발함. 실험과 계산을 병행하여 각 프로브의 선택성과 감도의 차이를 설명함.
□ 연구결과의 활용계획
- 현장진단이 가능한 질병진단용 및 독성물질 검출용 센서 개발을 통하여 상용화 가능한 소형 디바이스 제작의 기술력 확보를 가능하게 함.
- 파이로포스페이트 선택적인 센서는 세포생물학 및 의학 연구에 큰 진전을 가져올 수 있고, 조기 암진단 등에 효과적으로 사용될 수 있음.
- 독성물질 센서 개발을 통하여 테러/환경오염 물질의 노출에 대한 위험으로부터 보호함.
- 탄소재료 기반 초박형 전자소자와 단분자 프로브를 결합하여 착용 가능한 센서 시스템을 개발할 수 있음.

We will develop sensing methods for various analytes using carbon materials, molecular probes, and carbon material-molecular probe conjugates. First, we combined molecular probes and carbon materials to develop a new type of probes for phosphate-containing biomolecules. We also developed graphene-based field-effect transistor (G-FETs) probes. Second, we developed electrochemiluminescent (ECL) and electrochemical probes based on carbon dots. Third, we developed molecular probes based on electrostatic interactions. Fourth, carbon materials-based sensing system was developed. Lastly, we developed electrochemiluminescent (ECL) probes using iridium complexes.
□ Result
(1) We developed a new sensing system for PPi using electrostatic interactions between graphene oxide and molecular probes. This system was able to distinguish PPi from large excess of NTPs, and was successfully applied to real-time monitoring of polymerase chain reaction.
(2) We developed a new ionic compound, (carbon dot)-Nax+, which was applied to fundamental research for ECL probes. We synthesized carbon dot-based ionic gels which could be used as electrochemical catalysts, however, not as sensors because of difficulty in diffusion of analytes into ionic gels.
(3) We developed fluorescent “ON-OFF” probes for As(III)-containing toxic compounds, “turn-on” probe for biothiols through Cu(II) to Cu(I) redox conversion, and flavin probes using flavin-mediated photooxidation.
(4) We were able to fabricate wearable electrocardiogram sensor using carbon materials and color-tunable organic light-emitting diodes.
(5) We developed iridium complex-based electrochemiluminescent (ECL) probes for sulfide and homocystein with high selectivity and sensitivity.
□ Expected Contribution
- Our sensing systems will be developed into point of care detection systems for disease diagnostics and detection of toxic materials, which would make it possible to construct small devices for commercialization.
- PPi probes would bring much progress in the field of cell biology and medical research, and also can be used for early diagnosis of cancers.
- Toxic material probes will protect us from exposure to terror/environment-contaminating materials.
- Wearable sensing systems can be developed by combining a carbon material-based thin electronic device and a molecular probe.
