- 마비성패류독소를 생산하는 와편모조류 4종을 요각류 Tigriopus japonicus 에게 먹이로서 제공후 독성반응을 관찰한 결과, 4종 중 Gymnodinium catenatum에 대해 T. japonicus의 행동학적 이상반응 확인함.
○ 마비성패류독류(PSPs) 독소평가 방법 확립
- Gymnodinium catenatum에 대한 생물검정법으로서 T. japonicus의 특정 endpoint로서 행동학적 이상반응을 정량화하기 위해 행동추적 소프트웨어를 사용함으로서 T. japonicus의 행동학적 검정법이 PSP 독성평가를 위한 새로운 기법으로 가능성이 있음을 확인함.
2. 해양생물독의 확보/분석방법개발
가. 마비성패류독 생산방법 및 분석법 연구
○ Pre-column oxidation-FLD를 이용하여 국내에서 주로 발생하는 마비성 패류독소 6종에 대한(STX, GTX5, C1,2, dcSTX, GTX2,3, dcGTX2,3) 정성, 정량 분석법 확립함.
○ 2017년 4월에 송도, 다대포, 거제도 채집된 진주담치와 2018년 4월에 영도에서 채집된 진주담치와 따개비에서 GTX2,3과 STX가 주로 발생하는 마비성 패류독소임을 확인함.
○ 배양된 와편모조류 Alexandrium catenella에서 GTX2,3과 STX, GTX5가 존재함을 확인하고, ion-exchange column을 이용하여 소량의 STX, GTX2,3 분리함.
○ PSP 표준시료 확보의 생물의존성을 탈피하기 위해 시도된 화학합성을 통해 1-chloropentan-4-one으로부터 여러 가지 삭시톡신 합성 중간체 확보함.
나. 유독해양생물확보 및 생물독 분리
○ 국내 및 열대 유독해양생물 확보
- 국내 유독 함유 추정 복족류 해양생물 총 6개 과에서 19종 채집함.
○ 신규 국내 유입 유독 아열대 종 탐색
- 제주 해역에서 국내 미보고종인 아열대 모래말미잘류인 Palythoa mutuki 군락 발견함.
- Palythoa 종은 팔리톡신이라는 맹독 성분을 함유하고 있는데 복어가 가진 테트로도톡신 보다 70배 이상 높은 독성이며, 근육통, 호흡곤란, 경련 등의 증상을 일으키며 최악의 경우 사망에 까지 이르게 함.
- 최근 Palythoa 종이 댕기열 바이러스 치료에 효과적이라는 보고가 있으며, 이는 제주 해역에서의 Palythoa mutuki의 발견이 향후 연구에 있어 매우 의미 있을 것으로 판단됨.
○ 생물독 분리
- Conotoxin을 함유하고 있어 맹독성을 나타내는 것으로 알려진 Conus pulicarius로부터 저분자 화합물 분리하고 분광학적 방법을 통해 구조를 규명. 3종의 신규 steroidal glycoxide 화합물을 포함한 5종의 steroid계 화합물을 확인함.
- 육식성 포식생물로 알려진 제주 연안 서식 패류 Reishia clavigera로부터 저분자 화합물을 분리하고 분광학적 방법을 통해 구조를 규명. 6종의 기지 화합물을 확인함.
○ 해양생물독 리스트작성
- 해외 마이크로네시아 축, 코스래 지역과 국내 연안으로부터 확보된 유독 해양생물은 3계 7문 10강 17목 23과 48종을 확보하였음.
3. 유독해양생물로부터 유용독 생산/생합성기술개발
가. 유독해양생물의 분리/확보 및 최적배양조건 규명
○ 해양 미세조류
- 한국 연안에서 나타나는 마비성 패독(PSP; Paralytic Shellfish Poisoning)의 원인 종을 확보하고 대량 배양 시스템을 구축하기 위하여, 확보된 종에 온도와 염분에 대한 성장 반응을 평가함.
- 형태적, 분류계통학적 특성 분석을 바탕으로 5종의 Alexandrium 종과 Gymnodinium catenatum을 배양주로서 확보 했고, 대량 배양 가능성을 확인함.
- 목표종의 대량배양을 위해 rolling incubator가 적용된 대량 배양 시스템을 구축함.
○ 해양 미생물
- 독소를 생산하는 해양미생물의 선별을 위하여 베트남, 필리핀, 마이크로네시아 등의 열대지방과 한국 근해의 다양한 해양환경에 서식하는 해양 동·식물, 해양퇴적토, 해수로부터 분리된 다양한 해양미생물들을 스크리닝하여 3종의 독소 생산 해양미생물을 선정하였음.
- 선정된 해양미생물을 배양하여 얻은 추출물로부터 각종 크로마토그래피, 분광학적 방법을 이용하여 2종의 기지 독소 및 2종의 신규 독소를 분리하였음.
- 유용 독소의 대량생산을 위한 배양조건 탐색을 진행하여 신규 독소를 생산하는 168ST-16.1 균주의 최적배양조건을 규명하였음.
4. 해양생물독으로부터 유용산업소재개발
가. 해양생물독의 의약소재 및 기능성소재로써 활용성 평가
○ 분리한 기지 및 신규 독소의 유용산업소재로의 활용 가능성을 검토하기 위해 생리활성 탐색을 진행하였음. 기지 독소인 아스텔톡신(asteltoxin)은 뇌의 면역세포인 microglia의 활성화에 의한 염증을 억제하는 효과를 나타냈음. Ophiobolin 계열의 신규 독소 2종의 경우, 암세포의 성장을 억제하는 뛰어난 효과를 보여 항암소재로써의 개발 가능성을 보여주었음.
○ 열대 청자고둥 Conus pulicarius에서 분리된 5종의 steroid 유도체를 인간 백혈병 세포주(K562)에 대한 세포독성 실험결과, 5종 중 3종의 신규 glycoside 화합물이 높은 수준의 세포독성을 나타냄을 확인함.

○ Assesment of toxicity of four dinoflagellates
- Four dinoflagellates were supplied to Tigriopus japonicus as a prey to assess the toxicity. Of these, Gymnodinium catenatum caused a decreased mobility of T. japonicus.
○ Establishment of evaluation method of PSPs toxin
- We quantify the mobility of T. japonicus on the presence of G. catenatum using a behavioral tracking software.
- The behavioural tracking method of T. japonicus has a potentiality to detect the PSP causing organisms as a new toxicity test.
2. Collecting marine biotoxins/developing analyzing methods
A. Investigation on methods for producing and analyzing of PSP toxins
○ The established HPLC-FLD method can be extended to development of the simple and efficient isolation method of PSP from poisonous marine creatures.
○ Isolation of a trace amount of PSPs from the cultivated dinoflagellate would provide an opportunity to develop a sustainable production for the analytical standard.
○ An efficient synthetic route to PSP can be developed utilizing the synthetic intermediates that were prepared through this study.
B. Collecting the toxic marine organisms and separation of biotoxin
○ Collecting the Korean and tropical marine organisms containing toxins
- We collected marine gastropoda that is expected to have poison in Korea. A total of 6 families and 19 species were secured.
○ Investigation of subtropical toxic species in Korea
- Palythoa mutuki known to the subtropical species was newly found within zoanthid community in Jeju Island.
- Palythoa species contain a toxic ingredient called palytoxin and is known to be more than 70 times more toxic than tetrodotoxin in pufferfish.
- The discovery of Palythoa mutuki in the Jeju waters is considered to be very meaningful in toxic studies.
○ Separation and identification of toxins in marine organisms
- Isolation and identification of five steoid compounds including three novel steoidal glycosides from tropical Conus pulicarius which is known to secrete highly neurotoxic conotoxins.
- Isolation and identification of six small molecule metabolites from the Reishia clavigera which is known as a carnivourous predator inhabited in the coast of Jeju island.
○ Compilation of toxic marine organisms list
- Forty-eight toxic species belonging 3 kingdoms 7 phyla 10 classes 17 orders 23 families were collected from Chuuk and Kosrae in Micronesia, coastal waters of Korea.
3. Development of synthetic methods for useful toxin from marine toxic organisms
A. Collection of marine toxic organisms and its culture conditions
○ Marine microalgae
- To collect the PSP(paralytic shellfish poisoning) producing dinoflagellates, phytoplankton community was investigated from Korean coastal water.
- As a result, six species composed of Alexandrium species (Alexandrium pacificum, A. affine, A. catenella, A. fraterculus, and A. pseudogonyaulax) and Gymnodinium catenatum, which are known to produce the saxitoxin, were established based on morphological and phylogenetic data, and these species were deposited as strains in the Library of Marine Samples, KIOST.
- The effects of temperature and salinity on the strains were examined to clarify the environmental conditions favorable for the growth, and based on the obtained data, the mass culture system was constructed using the rolling incubator.
○ Marine microorganism
- Various marine microorganisms isolated from marine invertebrates, seaweeds, sediments and seawater collected at Korea and tropical areas such as Vietnam, Philippine and Micronesia have been screened for microbial toxins.
- Three microorganisms producing toxins were selected and cultured to isolate microbial toxins.
- Two known toxins and two new toxins were isolated from the selected strains on the basis of combined results of various chromatographic techniques and spectroscopic methods.
- Optimum culture conditions of the strain 168ST-16.1 producing new toxins were determined.
4. Development of useful industrial materials from marine biotoxins
A. Evaluation of usability of marine biotoxin as medicinal and functional materials
○ Evaluating the cytotoxicity of the compounds isolated from toxic shellfishes
- The cytotoxicity evaluation of compounds isolated from the Conus pulicarius which indicated that the novel three steroidal glycoside exhibit potent growth inhibitory activity against human leukemia cell line K562.
○ Screening of biological activities of new toxins
- The known and new toxins isolated in this study were screened for biological activities to evaluate the potential as high-value bio-based chemicals and materials.
- The known toxin, asteltoxin, showed a inhibitory effect on the production of nitric oxide (NO) in BV-2 microglia cells. The new toxins, ophiobolin derivatives, displayed potent cytotoxicity against six cancer cell lines(HCT-15, NUGC-3, NCI-H23, ACHN, PC-3 and MDA-MB-231).
- Our results suggested that these toxins could be potential bioprobes for the development of anticancer drugs or neurodegenerative drugs.
