본 연구과제를 통해 총 17편의 SCI(E) 논문과 2편의 비SCI 논문을 게재하였고, 3건의 국내특허 등록과 7건의 국내특허가 출원되었다. 이와 관련하여 등록된 2건의 특허를 기술이전 하였고, 연구를 통해 신규로 발견된 균주에 대해서 1건에 대해서는 물질이전을 수행하였다. 향후 저감화 소재 및 균주의 추가 기능성을 입증하고 김치 및 식품 적용을 통해 개발된 기술을 지속적으로 기술이전을 추진할 계획임. 또한 위해요소 신속검출 기술인 식품첨가물 분석법에 대해서는 김치 및 타식품에 적용하여 관련 기관과 협의를 통해 표준 분석법으로 활용 가능성에 대해 서도 검토할 계획임. 본 연구에서 확립 및 개발된 분석법을 활용한 김치 모니터링 결과는 김치의 위생안전기준설정 및 저감화 관련 분야의 정책자료로 활용할 계획임.

▪ Establishment of a physicochemical hazardous factor detection method for confirming the safety of kimchi
- We analyzed potential hazardous factors to evaluate the safety of domestically distributed kimchi. Based on individual analysis using conventional HPLC, a novel detection method for analyzing 20 typed of kimchi additives in 5.5 minutes was developed using a sensitive LC/MS/MS method. The results showed a higher sodium saccharin level in Chinese kimchi products compared to domestic kimchi products. The versatility of the method was confirmed by applying the developed detection method to other products. The method may be used for food additive analysis not only in kimchi products but also in other foods. In addition, analysis of residual pesticides, nitrosamines, nitrogen compounds (biogenic amines), and phthalates, which are possible hazardous factors present in kimchi ingredients, was optimized, and domestically distributed kimchi products were analyzed to obtain basic data. In most kimchi products, physicochemical hazardous factors were found to be at safe, low, or undetectable levels. Furthermore, the contamination level and safety were confirmed through the arsenic speciation method used for salted fish. Based on the establishment of a hazardous factor detection method and monitoring, the safety results for distributed kimchi product may be used as basic data for determining safety standards for kimchi products in the future.
▪ Research on rapid detection of hazardous factors in kimchi
- Among the possible hazardous factors, biological hazardous factors during the manufacturing process of kimchi require sustained control. We have developed a technology based on PCR/CE without the DNA extraction step, for simultaneous detection of food poisoning bacteria, as indicators of kimchi sanitation. We applied the method for detection of six species of food poisoning bacteria in kimchi products to confirm its potential. Through PMA treatment, dead cell reactions that do not actually cause food poisoning were blocked and an optimal condition was established with CE and PCR for amplifying selective reactions in live cells. The method developed in this study could be used for screening six food poisoning bacterial species in kimchi, thus contributing to ensuring the safety of kimchi products. Additionally, we validated and applied a separation pretreatment technology using magnetic nanoparticles and a color-based diagnostic kit on four food poisoning bacteria species in kimchi. We believe that continuous research is required for manufacturers to develop a kit for direct use on-site.
▪ Establishment and validation of quarantine standard inspection methods for kimchi-importing countries
- Each country importing kimchi from us has very different quarantine standards and there a solution is needed to address these differences. In this study, the standards and detection methods adopted by four countries (China, Japan, USA, and Taiwan) to which domestic kimchi products are exported were investigated to establish the detection method required for exporting. According to the Chinese pickling product standards, nitrate and nitrite standards have already been established using an ion chromatography method, which is different from the domestic color-based diagnostic method. In this study, a detection method was established to monitor kimchi products in the market.
Currently, kimchi products do not indicate allergen-containing ingredients and the presence of allergens and GMO may need to be addressed in the future. Therefore, a kimchi optimization experiment was conducted using commercially available detection kits, and the presence of allergen substances in salted shrimp was confirmed. However, in the case of GMO, low GMO DNA could not be amplified, thus confirming that GMO crops were not in use in kimchi products. Thus, it was shown that a more sensitive kit needs to be developed in the future.
2. Development of technology to reduce levels of kimchi hazardous factors
▪ Search for natural antimicrobial ingredients
- We have identified natural ingredients that can reduce levels of biological hazardous factors (food poisoning bacteria, norovirus) and quality hazardous factors (cabbage soft-rot disease) that may contaminate kimchi during its manufacture and confirmed their antimicrobial activity against these factors. Hence, totally, 19 ingredients, comprising 5 species of salt plants (Glehnia littoralis, Salicornia herbacea, Spergularia marina Griseb, ice plant, and Suaeda japonica), 10 species of kimchi-derived lactobacillus, and 4 species of phenolic compounds were identified. On testing for antimicrobial activity, 10 ingredients (5 salt plant extracts and 5 lactobacillus) were found to be excellent against hazardous factors. The application of the selected natural ingredients for reducing hazardous factor levels demonstrated 100% inhibition of food poisoning bacteria and Pectobacterium carotovorum subsp. carotovorum (PCC), which causes cabbage soft-rot disease at a 3-4 log scale.
▪ Development of norovirus detection technology and control materials
- Human norovirus, a water- and foodborne virus, is difficult to detect in food and environmental samples. In this study, we developed a technology to induce RNA to bind propidium monoazide (PMA) via a photoactive reaction to inhibit the amplification of inactive viruses and allow for selective detection of active norovirus. In addition, we confirmed that norovirus can be controlled during the cabbage pickling stage, as shown by a 3 log reduction, on treatment for 12 hours with salt plant extracts (S. herbacea and S. japonica), which are known for their hazardous factor-reducing effects.
▪ Identification of major compounds in microbial level-lowering ingredients
- The active components of the selected hazardous factor-reducing ingredients (G. littoralis and S. herbacea) were identified by LC/MS/MS, HPLC, and GC/MS analyses, and the food poisoning bacteria-lowering effects of the main compounds were confirmed. Thus, 91 G. littoralis-derived compounds and four novel flavonoid derivatives (kaempferol 3-O-β -rutinoside, cynarin, spiraeoside, purpurin), which have antimicrobial activities, were identified. In S. herbacea, 5 phenolic acids (vanillic acid, caffeic acid, p-coumaric acid, chlorogenic acid, sinapic acid) with antimicrobial activity and 6 species of fatty acids (linoleic acid, palmitic acid, oleic acid, linolenic acid, stearic acid, butyric acid) were identified.
▪ Evaluation of the applicability of natural antimicrobial ingredients to kimchi
- To commercialize the selected hazardous factor-lowering ingredients, we evaluated their action at each step of the kimchi manufacturing process. After treatment of the cabbage pickling process with the antimicrobial ingredient (e.g., G. littoralis extract) at 1% concentration for 24 hours, 100% reduction of B. cereus was achieved at 3-4 log CFU/g.
When the antimicrobial ingredient was added at 1% concentration during kimchi seasoning, no B. cereus was detected after 30 days of storage, whereas 1.5 log CFU/g was detected in seasoning not treated with the ingredient. Furthermore, the antimicrobial ingredient did not affect the sensory characteristics of the pickled cabbage, seasoning, and kimchi, which confirms its applicability. On evaluating the applicability of the antimicrobial ingredient in other foods, 99.9% reduction in B. cereus was achieved in a commercial salad treated with the ingredient compared to an untreated salad.
3. Development of quality control technology for intelligent safe distribution system of kimchi products
- Kimchi is a fermented food that is affected by conditions such as temperature during its distribution. It is difficult for consumers to understand the product condition. Therefore, development of quality control technology is required to identify the quality characteristics, depending on various distribution condition changes, and to ensure security of consumer choices. In this study, lactobacillus pH and total acidity were set as a standard index and a regression equation was derived for prediction of a quality table at 4 and 10 °C by analyzing the correlation patterns between indicator color changes and external environmental changes. Development of an intelligent quality index prediction system for kimchi led to the verification of quality improvement in products before and after (within 70% confidence level and within ±5% error). The establishment of a distribution system for kimchi is difficult at this point and the applicability may be likely if Kimchi is added to the distribution of fresh produce in the future.
