화장품 등의 안전성평가에서 동물실험 금지에 대한 국제 규제가 강화됨에 따라 국제 조화된 동물 대체시험법의 국내 도입 및 개발을 위한 지원이 필요하다. 동물대체시험법의 확립과 개발·검증 연구를 위해서는 시험법의 예측력 확인이 필요하며 이를 위한 시험물질은 in vivo 및 in vitro 시험 결과를 토대로 선정할 필요가 있다. 이를 위해 식품의약품안전평가원에서는 피부감작성 물질 데이터베이스(ChermSens DB) 및 안자극 물질 데이터베이스(ChemEye DB)를 마련하여 검증연구에 활용('13~'17.)해 왔으나, 내부에서만 활용되었다. 본 과제에서는 내부에서만 활용되던 물질 데이터베이스를 확대·개선하여 동물대체시험법 개발·검증에 활용할 수 있는 시험물질 정보를 누구나 쉽게 이용할 수 있도록 '피부감작성 및 안자극 참고시험물질(reference chemical) 데이터베이스 시스템'을 개발하였다. 이는 온라인으로 공개될 예정이며 동물대체시험법의 활용 활성화에 기여할 것으로 기대된다. 또한, 본 과제에서는 국제 조화된 피부감작성·안자극 관련 OECD 시험가이드라인을 국내 도입·보급하기 위해 민원인 안내서(제정) 발간, 표준작업지침서 마련 및 기술전수 워크숍을 개최하였다.
1차 연도의 과제 목표는 ChemSens DB를 확대·개선하고 국제 조화된 「IL-8 루시퍼라아제 시험법」 (OECD TG442E)을 도입하여 민원인 안내서를 제정하는 것이다. 연구 결과, 기존 ChemSens DB는 OECD 시험가이드라인(in vivo 및 in vitro 시험법) 7건의 8개 시험법에서 12개 시험법으로 시험물질 총 287종에서 396종의 데이터를 포함하도록 확대하였다. 그리고 물리·화학적 정보, Human date, in vivo data 및 in vitro data 등 동물대체시험법 개발·검증연구를 위해 필요한 정보들을 모두 포함할 수 있도록 정보 구성을 개선하였다. 최종적으로 마련된 데이터베이스를 '피부감작성 참고시험물질 데이터베이스'라고 정하였다. 또한, OECD 동물대체시험법인 「IL-8 루시퍼라아제 시험법」의 숙련도 평가를 통해 IL-8 루시퍼라아제 분석을 통한 피부감작성 안전성평가에 대한 기술력을 확보하였고, 이를 바탕으로 '화장품 등 동물대체시험법 가이드라인(민원인 안내서)을 제정('21.9.)하고 보도자료를 배포하였다. 추가적으로 「IL-8 루시퍼라아제 시험법」의 검증연구에서 사용되었던 시험물질 20종으로 실험을 실시하여 도입된 시험법이 잘 수행 되는지를 확인하였으며 실험 결과를 바탕으로 표준작업지침서를 마련하였다.
2차 연도의 과제 목표는 ChemEye DB를 확대·개선하고 최근 개정된 「단시간 노출법」(OECD TG491)을 확립하여 국내 비임상시험실시기관(GLP) 등에 보급하는 것이다. 연구 결과, ChemEye DB는 OECD 시험가이드라인(in vivo 및 in vitro 시험법) 7건의 10개 시험법에서 8건의 11개 시험법으로 확대되었다. 시험물질은 총 473종에서 514종의 데이터로 확대하였고 물리·화학적 정보, Human date, in vivo data 및 in vitro data 등 동물대체시험법 개발·검증연구를 위해 필요한 정보들을 모두 포함할 수 있도록 정보 구성을 개선하였다. 최종적으로 마련된 데이터베이스를 '안자극 참고시험물질 데이터베이스'라고 정하였다. 또한, 최근 개정된 OECD 동물대체시험법인 「단시간 노출법」 국내에 도입하기 위해 참고문헌에서 사용되었던 고 휘발성 시험물질 20종의 시험을 통해 「단시간 노출법」의 개정 시험법을 확립하였다. 추가적으로 「단시간 노출법」 결과가 없는 시험물질 9종을 선정하여 안자극을 평가였고 안자극 참고시험물질 데이터베이스에 추가하였다. 실험 결과를 바탕으로 표준작업지침서 개정 및 「단시간 노출법」시연 동영상을 제작하였고 GLP 기관 등에 시험법 활용을 지원하기 위해 기술전수 워크숍(온라인)을 개최하였다.
본 과제를 통해 개발된 '피부감작성(1차 연도) 및 안자극(2차 연도) 참고시험물질 데이터베이스 시스템'은 동물대체시험법 관련 산·학·관을 비롯하여 누구나 쉽게 활용할 수 있도록 온라인 전산시스템(모바일용 포함)으로 개발되었다. 전산시스템을 과학적·체계적인 동물대체시험법 관련 정보를 편리하게 이용할 수 있도록 개발하였는데 특히, 검색시스템에 자동완성 기능을 탑재하여 물질명 또는 CAS No.의 일부만 일치하여도 원하는 물질을 검색하고 인쇄할 수 있다는 장점이 있다. 또한, '피부감작성 및 안자극 참고 시험물질 데이터베이스 시스템의 사용설명 동영상을 제작하여 이용자들의 편의성을 도모하였다.
결과적으로 본 연구는 동물대체시험법 개발·검증을 위해 필요한 피부감작성 및 안자극 참고시험물질 데이터베이스를 마련하고 전산시스템을 개발하여 동물대체시험법 관련 연구자들이 쉽게 활용할 수 있도록 지원하였고, 국제 조화된 동물대체시험법이 국내에서 이용될 수 있도록 신규 피부감작성 동물대체시험법(TG442E) 및 최근 개정된 안자극 동물대체시험법(TG491)을 도입하여 안전성평가 기술력을 확보하였고 국내 동물대체시험법 활용 확산을 위한 기반을 구축하였다.

As the regulation banning the use of animal testing for the safety assessment of chemicals including cosmetics has been tightened, there should be support for introduction and development of internationally harmonized alternative test methods in Korea. To establish the alternative test methods that have been already developed, and to develop and validate novel alternative test methods, predictive capacity of the methods should be confirmed and the test chemicals used in such a confirmation should be selected based on in vivo and/or in vitro data. Although NIFDS (National Institute of Food and Drug Safety) have created databases on skin sensitizers (ChemSens DB) and eye irritants (ChemEye DB) and utilized them in a validation study conducted from 2013 to 2017, they have been used only within the NIFDS. In this study, the chemical DBs that had been only utilized internally were expanded and improved to develop Reference Chemical Database on Skin Sensitizers and Eye Irritants' so that anyone can access to the test chemical information. The DB will be available online and is expected to promote the use of alternative methods. In addition, we prepared test guidelines and SOPs (Standard Operating Procedures) and organized workshops in order to introduce and disseminate the OECD Test Guidelines for internationally harmonized skin sensitization and eye irritation test methods in Korea.
For the first year of the study, the goal was to update and improve ChemSens_DB, and to establish and publish the guideline on the internationally harmonized IL-8 Luc assay. Based on our research, ChemSens DB was expanded from 8 test methods (in 7 OECD Test Guidelines including in vivo and in vitro) to 12 test methods, and from 287 test chemicals to 396. Further, the information composition of ChemSens DB was improved to incorporate all the information needed for the development and validation of alternative methods including physiochemical information, human data, and in vivo, in vitro data, etc.. The final version of the DB was named as 'Reference Chemical Database on Skin Sensitizers. Further, we obtained technical proficiency with the safety assessment for skin sensitization using IL-8 Luciferase analysis by evaluating our proficiency with IL-8 Luciferase assay, an alternative to animal testing. Additionally, we confirmed the performance of the assay by conducting experiment using 20 test substances from the validation study of IL-8 Luciferase assay.
For the second year of the study, the goal was to expand and improve ChemEye DB, and to establish Short Time Exposure (STE) assay (OECD TG491) and disseminate the assay to the national GLP test facilities. As a result of our research, the ChemEye DB has been expanded to include 11 test methods (in 8 OECD TGS including in vivo and in vitro) from 10 test methods (in 7 OECD TGs) and the number of the test substances in the DB has increased from 473 to 514. Further, we improved the information composition of the ChemEye DB so that it can incorporate all information required for the development and validation of alternative test methods, including physiochemical information, Human data, in vivo and in vitro data, etc.. The final version of the DB was named as 'Reference Chemical Database on Eye Irritants'. In addition, we tested 20-highly volatile substances that were used in the references of Short Time Exposure (STE), the OECD alternative method that has been revised recently, to reproduce the assay and thus to introduce it in Korea. Also, we tested 9 additional test substances with no STE data to assess their eye irritation potential, and added the results to the DB. We revised the SOP of the assay, produced a video demonstrating it, and organized a training workshop to facilitate the use of the assay in the GLP test facilities.
The Reference Chemical Databases on Skin Sensitizers and Eye Irritants developed in this study (1st and 2nd year respectively) is build with an online computation system (including mobile access) to let anyone from the industry, academia and research institute can easily access to it. We enhanced the user experience of the computation system by allowing easy search of scientific and systematic information related to alternative methods. In particular, with the search engine equipped with predictive text input, users can search and print out the chemical information they want even if only part of the chemical name or Cas No. is agreed. In addition, we facilitate the use of the DBS by producing tutorial.
In this study, we established Reference Chemical Databases on skin sensitizers and eye irritants that are necessary for the development and validation of alternative methods, and developed a computation system so that researchers in the field can easily facilitate to the DBs. Also, we obtained technical proficiency with the safety assessment and established foundation for utilization of alternative methods by reproducing a novel alternative method for skin sensitization (TG442E) and a recently revised alternative methods for eye irritation (TG491) to promote the use of internationally harmonized alternative methods within the nation.
