Hydrogen ion 농도는 pH의 결과와 반대로 증가하는 경향을 나타내었으며 사료의 ADF수준(P<0.01)과 buffer(P<0.001)의 수준에 크게 영향을 받았으며 특히 배양 6시간 이후 크게 증가하는 경향을 보였다. 배양액의 buffering capacity는 배양시간이 경과함에 따라 감소하는 경향을 보였으며 특히 sodium bicarbonate의 처리수준에 영향을 받는 것으로 나타났다(P<0.001). 그러나 ADF수준에 대한 영향은 배양초기에 나타나지 않았으나 배양 24시간에 ADF수준이 증가함에 따라 증가하는 결과를 보였다. Ammonia-N 농도는 배양시간, ADF 수준, sodium bicarbonate 처리수준에 따라 다양한 경향을 나타내었으며 배양 6시간에 증가하여 12시간에 감소하며 다시 24시간에 증가하는 경향을 나타내었다. 건물 소화율과 ADF 소화율은 배양시간이 경과함에 따라 일정하게 증가하는 경향을 보였으며 건물소화율은 ADF수준이 증가함에 따라 감소하는 경향을 나타내었다. Sodium bicarbonate 처리 수준이 증가할수록 높은 경향을 보였다. ADF 소화율은 건물 소화율과 비슷한 경향을 보였으나 ADF 수준에 따른 결과에서 sodium bicarbonate를 처리한 것과 처리하지 않은 것과는 유의한 차이를 보였다(P<0.001). 그러나 처리구간에서는 차이를 나타내지 않았다. 배양액의 총 VFA 생성량은 ADF 수준에 있어서 현저한 차이를 보였으나 (P<0.01) sodium bicarbonate 처리수준에 따라서는 차이가 없었다. 배양 6시간 이후 크게 증가 하였으며 배양 24시간의 총 VFA양은 ADF 수준이 10, 20 및 30% 처리구에서 각각 70.57, 61.31 및 48.62 Mm/100ml이었다. Acetate/Propionate 비율은 배양초기 3.89에서 배양시간이 경과함에 따라 급격하게 감소하는 경향을 보였으며 sodium bicarbonate를 처리함으로써 모든 처리구에서 높은 경향을 보였다(P<0.001). ADF 30% 처리구에서 배양 3시간에 다소 증가하는 경향은 보였으며 ADF 수준이 증가할수록 높은 경향을 나타내었다. 실험 2 사료의 ADF 및 buffer의 종류가 반추위내 발효성상 및 buffering capacity에 미치는 영향을 in vitro 방법으로 연구하였으며 ADF 공급원으로는 alfalfa, NH_(3)-treated rice straw 및 beet pulp를 이용하였으며 buffer 공급원으로서 sodium bicarbonate, magnesium oxide, 그리고 limestone을 사용하였다. 시료중의 단백질 함량과 ADF 수준은 17%와 20%로 하여 배양하였으며 이의 결과는 아래와 같다. 배양액의 pH는 배양시간이 증가함에 따라 감소하는 경향을 보였으며 사료의 ADF 종류(P<0.01) 및 buffer 종류에 따라 영향을 받았으며 (P<0.05) beet pulp 처리구에서 다른 처리구에 비해 낮은 경향을 보였다. These experiment were conducted to study the effect of levels of ADF and sodium bicarbonate and the sources of ADF and buffer on fermentation, bacterial mass, and buffering capacity in vitro. Diets were prepared from purified form and extracted with selected feedstuffs, and formulated with ADF levels of 10, 20, and 30% of DM and sodium bicarbonate levels of 0, 1, and 2% of DM in experiment 1. In experiment 2, feeds were formulated to contain 20% ADF with alfalfa hay cube, NH_(3)-treated rice straw, and beet pulp, as a major ADF sources and contained 17% CP on a dry matter basis in these experiments. Buffers were used sodium bicarbonate, magnesium oxide, and limestone. Diets were alotted by 3?3 Factorial design with levels of ADF and sodium bicarbonate in Exp. 1, and sources of ADF and buffers in Exp. 2, respectively. The results obtained from these experiments are as fellows. Experiment 1. pH of medium were significantly affected by levels of ADF and sodium bicarbonate(P?0.001). 10% ADF without buffer were lower than other treatments during incubation time. Hydrogen ion concentrations were largely increased by decreased levels of ADF at 6hr incubation and highly affected by sodium bicarbonate(P<0.001) and levels of ADF(P<0.001) Buffering capacity of medium fluid were highly affected by levels of sodium bicarbonate(P?0.001). and were affected by levels of ADF(P?0.001). In this study, buffering capacity tended to be decreased in all treatments during incubation time. In ADF 10 and 20% treatment, the influence of sodium bicarbonate level on medium acid-base status were the highest(P?0.001). Ammonia-N concentrations were variable among treatments and primary affected by ADF level, and tended to be in increased 12hr after incubation. At 24hr incubation, ammonia-N concentration were the highest for 20%(15.23 mg/100ml), intermediate for 10%(12.89 mg/100m1), and the lowest for 30% level of ADF(9.57 mg/100m1). Dry matter(IVDMD) and ADF digestibilities(IVADFD) were influenced significantly by levels of ADF(P?0.001). and sodium bicarbonate (P?0.01). By the increasing of ADF level tended to be declined for digestibilities of DM and ADF, and increased by treatlments with sodium bicarbonate. Concentration of total VFA were not affected by levels of sodium bicarbonate, whereas it were significantly affected by levels of ADF(P?0.001). Total VFA were largely increased after 6hr incubation in medium fluid. A/P ratio were affected by the increased levels of ADF(P?0.01). and sodium bicarbonate(P?0.001). Experiment 2. Ruminal fluid pH were largely decreased during incubaion time. and affected(P?0.01) by dietary ADF and buffer source, respectively. However, group of beet pulp was not affected by the levels of buffer. Buffering capacity of medium fluid were highly affected by both ADF (P?0.01) and buffer sources(P?0.001), and tended to be decreased during incubation time. Based on major ADF sources, treated group with beet pulp were the lowest(P?0.05) as compared with that of alfalfa and NH_(3)-rice straw. The highest buffering capacity was observed in magnesium oxide treatmet except group of alfalfa. Hydrogen ion concentrations were affected by dietary ADF(P?0.01) and buffer source(P?0.05), and there were drastically increased 6hr after incubation among treatments. There were lower in treated group of limestone and beet pulp but higher in NH_(3)-rice straw treated group throughout the incubation time. No differences showed in group of alfalfa and NH_(3)-rice straw. Ammonia-N concentrations were variable among treatments, affected by ADF source(P?0.01), and tended to be increased at 24hr incubation time. As compare with the other treaments, group of NH_(3)-rice straw was decreased at 12hr, and thereafter increased. In the case of beet pulp treated group, ammonia-N concentration teded to be increased during incubation. Dry matter and organic matter disappearances were significantly affected by dietary ADF(P?0.001) and buffer sources(P?0.05) except group of limestone. There were largely increased by the increasing time after incubation. DM and OM disappearance were the highest for group of beet pulp with sodium bicarbonate and beet pulp with mahnesium oxide, respectively. But DM and OM digestibilities for groups of limestone and NH_(3)-rice straw were the lowest irrespective of sources of ADF and buffer during incubation. Crude protein and ADF disappearances were highly affected by ADF(P?0.01) and buffer source(P?0.05). Protein disappearance for group of beet pulp were the highest. ADF disappearance were the lowest in group of NH_(3)-treated rice straw. Microbial cell dry matter yield were affected by dietary ADF sources(P?0.05) but not affected by sources of buffer. No differences showed in group of alfalfa hay cube and NH_(3)-rice straw. In beet pulp treatment, sodium bicarbonate were the lowest, intermediate for magnesium oxide and highest for limestone. In these results, levels and sources of ADF and buffer were influenced on ruminal parameters such as pH, buffering capacity and yields of VFA, and digestibilities of DM, ADF, and CP were more affected by levels and sources of ADF. More importantly, buffering capacity are largely affected by sources of ADF and levels of buffer, thus it is needed to study the influence of these factors on practical feeding systems.

In this study, buffering capacity tended to be decreased in all treatments during incubation time. In ADF 10 and 20% treatment, the influence of sodium bicarbonate level on medium acid-base status were the highest(P?0.001). Ammonia-N concentrations were variable among treatments and primary affected by ADF level, and tended to be in increased 12hr after incubation. No differences showed in group of alfalfa hay cube and NH_(3)-rice straw. In beet pulp treatment, sodium bicarbonate were the lowest, intermediate for magnesium oxide and highest for limestone. In these results, levels and sources of ADF and buffer were influenced on ruminal parameters such as pH, buffering capacity and yields of VFA, and digestibilities of DM, ADF, and CP were more affected by levels and sources of ADF. More importantly, buffering capacity are largely affected by sources of ADF and levels of buffer, thus it is needed to study the influence of these factors on practical feeding systems.
