Continuous NSAIDs use results in a 40% to 50% reduction in relative risk for colorectal cancer. The precise mechanism by which NSAIDs prevent and/or cause the regression of colorectal cancers is not known. Some investigators have reported that some NSAIDs induced apoptosis and altered the expression of cell cycle regulatory genes in some carcinoma cells when administered at relatively high concentration. However, NSAIDs application as a chemoprevention agent in lung cancers still remains to be elusive. Therefore, human lung cancer cell line NCI-H1299 was used to investigate whether NSAIDs might induce apoptotic death of NCl-H1299 cells. Method: Viability test was carried out by MTT assay. Apoptosis was measured by flow cytometric analysis and nuclear staining(DAPI). The catalytic activity of caspase family was measured by fluorogenic cleavage of biosubstrates. To define the mechanical basis of apoptosis, western blot was performed for the expression of cleavage of death substrates(PARP and ICAD). Results: Sodium salicylate(NaSaL) significantly decreased the viability of NCI-H1299 cells, which was revealed as apoptosis characterized by an increase of SubG_(0)/G_(1) population and nuclear fragmentation. The catalytic activity of caspase-3 protease began to increase at 24 hours and reached a peak at 30 hours after treatment of 10 mM NaSaL, but the enzymatic activities of caspase-6, 8, and 9 proteases were not changed significantly. Consistent with activation of caspase-3 protease, NaSaL induced the cleavage of biosubstrate of the protease. Furthermore, Nasal-induced death of NCI-H1299 cells was augmented by the inhibitors of p38 MAP kinase. Conclusion: Nasal induces the apoptotic death of NCI-H1299 human lung cancer cells by activation of caspase-3 protease. Moreover, the survival signals of cells treated with NaSaL are partly dependent on p38 MAP kinase-mediated pathway.
