본 연구는 L-ascorbic acid를 함유하는 마이크로 캡슐 제조 공정의 최적화에 대한 연구이다. 이때 캡슐의 shell material로는 polycaprolactone을 도입하였으며 W/O/W 다중 유화법과 용매 증발법을 이용하여 캡슐을 제조하였다. 우선 안정한 W/O 에멀젼을 형성하도록 한 후, 이를 다시 외부수상에 재분산시켜 유화함으로써 W/O/W 다중유화액이 형성되면 oil상의 유기용매를 감압 건조하여 증발시킨다. 이후 동결건조 과정을 거쳐 최종적으로 얻어지는 마이크로 캡슐들을 pH 4.3의 완충액에 재분산시켜 방출 실험을 진행하여 순수한 L-ascorbic acid와 캡슐화된 L-ascorbic acid의 시간에 따른 활성도 감소를 비교하였다. 이 때 실험 결과에 영향을 주는 변인으로는 각 단계에 도입하는 유화제와 안정화제의 종류와 양, oil 연속상의 종류와 각각에 대한 수상의 비, 유화 속도와 시간, core material의 양과 이에 대한 shell material의 비 등을 들 수 있다. 실험 결과 W/O 1차 에멀젼의 경우, 안정도는 75%(w/w)의 Span 80과 25%(w/w)의 Tween 80을 조합한 HLB 6.975의 co-surfactant를 연속상 대비 20%(w/w) 도입한 경우 얻을 수 있었으며, (W/O)/W 2차 에멀젼의 경우 HLB와는 관계없이 2차 안정화제로써PVA 0.5%(w/w)나 gelatin 1% (w/w)를 도입한 경우 가장 안정한 유화액을 얻을 수 있었다. 이 때 최종 유화액의 분산 안정도는 침전율과 입자크기에 반비례하는 경향을 나타내었고 대부분의 경우 유화제나 안정화제의 농도가 증가할수록 분산 안정도는 더욱 증가하는 경향을 보였다. 생성되는 캡슐 입자의 평균 직경은 2-10um 범위이다. 또한 캡슐화 효율은 2차 안정화제로서 PVA 0.5%를 도입, 기포식으로 1차 유화한 후 임펠러형으로 2차 유화한 경우 가장 우수하였으며 core의 L-ascorbic acid 농도가 증가할수록 효율 및 분산안정도는 감소하였고 방출 실험 결과 캡슐화 된 L-ascorbic acid 는 캡슐화 되지 않은 경우에 비하여 시간에 따른 활성도의 감소율이 상대적으로 매우 작은 값을 나타내었다. 이상의 연구를 통하여 불안정한 L-ascorbic acid의 안정한 마이크로 캡슐의 제조를 성공적으로 수행할 수 있었다.

Microencapsulation of L-ascorbic acid (vitamin C) with polycaprolactone (PCL) was carried out by a W/O/W multiple emulsification and solvent evaporation technique. Various factors affecting microencapsulation were studied : type of emulsifiers or stabilizers, their concentration in each step, type of oil continuous phase in W/O primary emulsion, PCL concentration, agitation speed and agitator type. Degradation rate, particle size, encapsulation efficiency, stability were investigated. It was found that the maximum stability of the primary W/O emulsion indicated HLB 6.975 using 75% (w/w) of Span 80 and 25% (w/w) of Tween 80 without any additives. The 20% (w/w) emulsifier was most effective to stabilize the primary W/O emulsion. In the secondary (W/O)/W emulsion, dispersion was more stable with decreasing precipitation ratio and particle size, which was not much related with HLB of secondary emulsifier, contrary to the primary W/O emulsion. Then, final emulsion could be more stabilized by adding PVA, Pluronic, gelatin or its blending with Tween series. It was more stable with increasing emulsifier or stabilizer content. While, in that case of using PVA, emulsion was less stable with increasing its concentration. The effect of L-ascorbic acid content in the inner phase of microcapsules also indicated that higher core concentration brought down stability of final emulsion. Mean efficiency of encapsulation with low content of L-ascorbic acid (1% w/w) was higher than high content (5% w/w). At the similar dispersion stability, mean encapsulation efficiency as using PVA (0.5% w/w) was much higher than gelatin (1% w/w). Further analysis of the results indicated that the homogenizing type of each step might be playing a major role in the encapsulation efficiency. In this experiment, primary emulsion was formed by using air-bubbling homogenizer and secondary emulsification was performed by impeller type homogenizer for preparation gratifying products. Its release test showed that prepared PCL microcapsules could slow down degradation rate of pure L-ascorbic acid. Then, PCL microcapsules were successfully produced in W/O/W emulsion, and spherical mean particle size of the microcapsules containing L-ascorbic acid was in the range of 2-10 ㎛
