Mast cells have been implicated in the expression of a wide variety of biological responses, including immediate hypersensitivity reactions, host responses to parasites and neoplasms, angiogenesis, tissue remodeling, and immunologically non-specific inflammatory, and fibrotic conditions. Recent findings suggest that an important mechanism by which mast cells influences such biological responses is through the production of multifunctional cytokines. In the present study, I investigated the signal transduction pathways of secretion and gene expression of various cytokines on the mast cells. First of all, I examined the signaling pathway involved in calcium-related cytokine secretion in lgE-stimulated RBL-2H3 cells because Ca^2+ acts as an important second messenger in mast cell activation. I report that treatment with BAPTA-AM, a chelator of intracellular calcium, can inhibit IgE-stimulated TNF-αand IL-6 secretion and expression. In activated RBL-2H3 cells, the expression level of NF-κB/Re/ A protein increased in the nucleus. However, the level of NF-κB/Re/ A in nucleus was decreased by treatment of BAPTA-AM. In addition, BAPTA-AM completely inhibited the IgE-induced IκB kinase β (IKKβ) activation and IκBα phosphorylation. These observations demonstrate that the intracellular Ca^2+ may play an important role in IgE-induced TNF-α and IL-6 secretion from mast cells via IKKβ activation. I next examined the signal transduction pathways of cytokines expression in the ethanolstimulated HMC-1 because mast cells promote alcoholic fibrosis by producing cytokines contributing to fibrosis. Ethanol significantly increased the intracellular calcium level in HMC-1. Ethanol also significantly enhanced TNF-α, IL-6, and TGF-β1 production compared with media control in a dose-dependent manner at 24 h, but did not significantly affect the IL-1β production. The increased cytokine level was significantly inhibited by intracellular calcium chelator, BAPTA-AM, ERK (p44/p42) pathway inhibitor, PD98059 and p38 MAPK pathway inhibitor, SB203580. These inhibitors also inhibited ethanol-induced p44/p42 and p38 MAPK phosphorylation. In conclusion, my observations show that calcium mobilization and MAPK activation (p44/p42 and p38) are necessary for ethanol-induced cytokines expression. Finally, I investigated the effects of dexamethasone and p38 MAPK inhibitor, SB203580 (to support the effect of dexamethasone through p38 inhibition) on SCF-induced migration of RPMCs because previous work has demonstrated that glucocorticoids decreased tissue mast cell number and SCF-induced migration of mast cells required p38 MAPK activation. Here i report that dexamethasone and SIR203580 inhibits chemotactic movement of RPMCs induced by SCF. The ability of SCF to enhance morphological alteration and F-actin formation was also abolished by treatment of dexamethasone. Dexamethasone and SB203580 inhibited SCFinduced p38 MAPK activation to near basal level. In addition, i also found that SCF-induced inflammatory cytokine production was significantly inhibited by treatment of dexamethasone or SB203580. My results show that dexamethasone potently regulates SCF-induced migration and inflammatory cytokines production through blockade of p38 MAPK activation in RPMCs. Therefore, overall my results suggest that studies of signal transduction pathways of the cytokines in mast cells may form the basis of a new strategy for mast cell mediated disorders.
