To investigate the physiological changes of lactic acid bacteria induced by the adaptation to acidic environment, pH dependence of glycolysis activity, cytoplasmic membrane-bounded ATPase activity and new protein expression patterns during the adaptive acid tolerance response were studied. Furthermore, related to the cytoplasmic membrane's physiological changes, proton permeability of intact cell, the stability of liposomes composed of total lipid, and fatty acid composition were investigated. First, to obtain the maximal physiological changes between normal and ATR-induced cells, the maximal ATR inducing condition was studied. Maximal acid resistance was observed when this strain was exposed to pH 3.5 for 1hr prior to challenge at pH 2.5, although intermediate levels of protection were induced by exposure to pH values ranging from 3.0 to 6.0. Because chlorarnphenicol, a protein synthesis inhibitor, and dicyclohexylcarbodiimide, a ATPase F_(o) site inhibitor prevented full induction of acid tolerance response, full expression of the ATR required new protein synthesis and inclosed activity of ATPase. The result of pH dependence of glycolysis showed that the ATR-induced cells exhibited higher activities than control cells at acidic pH. And ATR-induced cells marked maximal glycolysis activities at pH 5.5, this pH was lower than control cells at the degree of 0.5 pH unit. The pH and maximal specific activities of ATPase originated from the control and ATR-induced cells were 0.21 units at pH 6.5 and 0.18 units at pH 5.5, repectively. Although the maximal specific activity of ATPase originated from ATR-induced cells was not higher than control cells, the overall ATPase acitivities of ATR-induced cells were higher than control cells at pH values ranging from 3.0 to 6.0. The proton permeability represents the membrane's integrity composed of membrane's physical charcteristics and membrane-bounded enzyme's functions. Assessment of the movement of protons into control and ATR-induced cells after an acid pulse at various pH values indicated permeability to protons were minimal at pH values about 6.0 and 5.0, at which the average half time for pH equilibration across the cell membrane were about 5.2 and 7.5 min, respectively. The ATPase inhibitor dicyclohexylarbodiimide acted to increase the permeability of control cells to protons at higher values than the ATR-induced cells, and this action indicated that the ATR-induced cells were more dependent on the no energy-consumping mechanisms for pumping out protons than control cells. The corresponding value for DCCD-treated control cells was 1.0min, and the value for the DCCD-treated, ATR-induced cells was 2.6min at the same pH 4.0. Liposomes composed of total cellular lipids extracted from the control and ATR-induced cells were analyzed for their stability at pH 2.5 shock and at 45℃ thermal shock conditions. At pH 2.5, ATR-induced cells' liposomes were considerably more stable about 2.25 times than liposomes of control cells. And the liposomes of ATR-induced cells acquired additional stability at 45℃ against thermal stress. Recently the questions were asked as to what kind of the changes of fatty acids composition during acid tolerance response caused the increased growth in the acidic environment. As acid tolerance response was acquired, nonadecanoic acid(C19:Ocyclo) were markedly increased and hepatadecenoic acid(Cl7:1), stearic acid(Cl8:0), and arachidic acid(C20:0) were slightly changed. Analysis of protein expression during the acid tolerance response by two-dimensional gel electrophoresis revealed a change in the expression of at least 31 proteins compared to the control clells. 23 proteins showed induced expression while 9 proteins were repressed. This result implied that the ATR is a complex response involving a modulation in the expression of many genes. These data reported here suggest that Leu. mesenteroides develops acid tolerance response upon exposure to prior, sublethal acid conditions.
