본 연구에서는 김치 재료가 풍미를 증진시키는 목적 외에 유해세균에 대한 항균효과가 있는지를 알아보기 위해 김치 재료인 고춧가루, 무, 생강, 양파, 마늘, 갓, 배추에 대해 식중독균인 Salmonella typhimurium, Listeria monocytogenes, Staphylococcus aureus와 분변오염 지표균인 Escherichia coli, 진균류Aspergillus sp. Penicillium sp.에 대한 항균활성을 살펴보았다. 김치재료를 methanol로 추출하여 그 추출물에 대해 항균활성을 paper disc agar diffusion법으로 살펴보았으며, 각 추출물의 최소저해 농도(MIC : minimum inhibitory concentration)를 구하고 추출물들이 사용미생물균주에 어떤 영향을 주었나를 주사전자현미경(SEM : scanning electron micoscope)으로 확인하여 다음과 같은 결과를 얻었다. 1. 각 김치 재료의 methanol추출물의 수율은 고춧가루 추출물이 20.47%로서 가장 높은 수율을 나타내었고, 생강이 9.12%, 마늘과 갓이 각각 7.29%, 7.44%였으며 배추가 6.86%, 양파가 4.64%, 무가 3.15%의 수율을 나타내었다. 2. Paper disc agar diffusion법을 사용하여 조사한 methanol 추출물의 항균효과는 김치 재료의 methanol추출물 모두에서 항균효과를 나타내었다. 특히 가장 우수한 항균성을 나타낸 것을 살펴보면 고춧가루, 무, 생강, 마늘, 갓 추출물이 E. coli에 대해 각각 22.20, 18.94, 10.13, 22.20, 21.11mm의 생육저지대를 보였고, 양파와 배추는 L. monocytogenes에 대하여 각각 13.81mm, 24.40mm의 생육저지대를 나타내어 우수한 항균성을 가진 것으로 검색되었다. 고춧가루와 무는 L. monocytogenesis에 대하여 11.38, 12.34mm의 저지대를 나타내었으며, 양파, 마늘, 갓, 배추는 S. typhimurium에 대하여 10.68, 17.63, 19.72, 18.73mm로 저지대를 나타내었고 생강은 S. aureus에 대해 9.80mm의 저지대를 나타내었다. 3. 김치 재료 methanol 추출물의 Aspergillus sp.과 Penicililum sp.의 곰팡이류에 대한 항균성을 검토한 결과는 마늘 추출물이 Aspergillus sp.에 대해 63.33%와 Penicillium sp.에 대해 49.27%의 저해를 나타내어 가장 우수하였다. Aspergillus sp.에 대해서는 생강 추출물이 57.69%, 무 추출물이 50.94%로 그 다음을 이었고 Penicillium sp.에 대해서도 역시 생강과 무 추출물이 각각 33.27%, 26.73%로 우수하였다. 4. 김치 재료 methanol 추출물을 0, 250, 500, 1000ppm으로 농도를 달리하여 유해미생물에 대한 생육저해곡선을 측정한 결과, S. typhimurium에 대해서는 고추, 무, 양파, 배추에서 농도별로 생육저해효과를 보였다. L. monocytogenes에 대해서는 고추, 양파 추출물이, S. aureus에 대해서는 농도와 관계없이 전체적으로 완만한 생육곡선을 보였으며, E. coli에 대해서는 모든 추출물이 농도별로 생육억제효과가 있음을 보여 주었다. 5. 김치재료 methanol 추출물을 1,000ppm으로 하여 생육저해율을 측정한 결과는 마늘 추출물이 S. aureus에 대하여 60%이상의 강력한 생육저해를 나타내었으며, 그 다음으로 양파 추출물이 E. coli에 대해 60%이상, S. typhimurium에 대하여 50%이상, 배추 추출물이 E. coli에 대하여 50%의 생육저해율을 나타내었다. 6. 유해균의 MIC를 측정한 결과, 고춧가루 추출물이 500 μg/mL로 L. monocytogenes에 대해 MIC를 나타내었고, S. typhimurium에 대해 무, 양파, 마늘, 갓, 배추 추출물이 1,000 μg/mL, L. monocytogenes에 대해 무, 양파, 배추 메탄올 추출물이 1,000 μg/mL, E. coli에 대해 고추, 무, 마늘, 갓 추출물이 1,000 μg/mL로 MIC를 나타내었다. 7. 김치 재료 methanol 추출물 중 가장 항균성이 뛰어난 갓과 마늘 추출물의 농도를 1mg/disc로 하여 40, 80, 100, 120℃에서 각각 1시간 동안 열처리 한 후 S. typhimurium과 E. coli의 생육정도를 측정한 결과, 갓 추출물은 모든 처리구에서 대조구와 차이가 크지 않은 항균활성을 보였으며 특히 120℃의 처리에 의해서도 항균활성이 있는 것으로 나타나 열에 매우 안정한 물질임을 알 수 있었으나, 마늘 추출물은 40℃를 제외한 모든 열처리구에서 항균활성이 낮아져 열처리에 약함을 알 수 있었다. 8. 갓과 마늘 추출물의 pH 안정성을 조사하고자 methanol 추출물의 농도를 1mg/disc로 하여 pH를 2, 5, 7, 9, 11로 조절한 후 S. typhimurium과 E. coli의 생육정도를 조사한 결과 갓 methanol 추출물은 모든 pH 조건에 대하여 대조구와 비교 시 항균활성에 있어 차이가 나타나지 않아 pH 안정성이 매우 우수함을 알 수 있었고, 마늘 추출물은 산성에 대해서는 안정한 반면, 알칼리쪽으로 갈수록 활성이 낮아지는 경향을 나타내었지만 활성을 완전히 잃지 않았다. 9. 갓과 마늘 추출물을 처리한 S. typhimurium과 E. coli의 주사전자현미경(SEM)사진을 본 결과 비처리군은 표면이 매끄럽고, 오염이 없는 깨끗한 모습인 반면, S. typhimurium과 E. coli에서 갓과 마늘 추출물 처리군은 세포질막의 변형으로 인한 형태변화는 없었으나 끈끈한 물질이 균체내에서 유출되어 응집되는 현상이 관찰되었다. 이상의 결과로서 김치 재료 중 고춧가루, 무, 생강, 양파, 마늘, 갓, 배추methanol 추출물은 항균활성물질을 갖고 있으며, 풍미를 증진시키는 목적 외에도 김치의 저장성 향상과 안전성 증대에 기여할 수 있음을 확인하였으며, 특히 갓 추출물은 열과 pH에 안정하여 다양한 가공적성에 이용될 수 있고, 더 나아가 항균 소재로서도 이용 가능성이 높음을 시사하였다.

Aspergillus sp.에 대해서는 생강 추출물이 57.69%, 무 추출물이 50.94%로 그 다음을 이었고 Penicillium sp.에 대해서도 역시 생강과 무 추출물이 각각 33.27%, 26.73%로 우수하였다. 4. 김치 재료 methanol 추출물을 0, 250, 500, 1000ppm으로 농도를 달리하여 유해미생물에 대한 생육저해곡선을 측정한 결과, S. typhimurium에 대해서는 고추, 무, 양파, 배추에서 농도별로 생육저해효과를 보였다. This study was carried out to determine the inhibitory effect of methanol extracts from Kimchi ingredients against Salmonella typhimurium, Listeria monocytogenes, Staphylococcus aureus and Esherichia coli which are pathogenic microorganisms and Aspergillus sp., Penicillium sp. Antimicrobial activity of methanol extracts was tested against bacteria by paper disc agar diffusion method and the MICs(Minimum Inhibitory Concentrations) of each methanol extract were obtained. It was also confirmed that the methanol extracts help improve the savor of Kimchi and assist in its storage preservative qualities for long term safely. Also, the methanol extract from leaf mustard is stable against heat and pH, so it can be processed widely over a broad range and is invaluable for development as natural antimicrobial material.
