B. cereus의 보다 정확한 검출과 B. cereus group의 선택적 검출을 위해 PCR을 수행하였으며, B. cereus group의 enterotoxin 생성을 확인하기 위해 BCET-RPLA assay를 사용하여 특성을 분석하였다. B. cereus group 중 B. mycoides는 선택배지상의 형태적 특징인 rhizoid form으로 구분이 가능하였으며, B. cereus는 BC1, BC2r primer, B. thuringiensis는 BT1, BT2r primer를 사용하여 구분이 가능하였으나, B. thuringiensis subsp. kurstaki의 경우 BC1, BC2r primer에서 PCR amplicon이 확인되었고, BT1, BT2r primer에서 확인되지 않아 선택적 확인을 위해 RB-19, U8-15 primer를 이용하여 PCR 수행으로 확인할 수 있었다. 그리고 hblA gene을 가진 B. cereus, B. thuringiensis와 B. mycoides의enterotoxin 생성 여부를 확인하기 위해 BCET-RPLA assay를 한 결과 모두 enterotoxin이 생성됨을 확인할 수 있었다. B. cereus와 같이 자연에 널리 존재하는 장구균(Enterococci)을 즉석섭취·편의식품인 생식과 선식에서 분리하고, 분리균의 항생제 감수성을 확인하여 그 위험성을 조사한 결과 Enterococcus는 선식과 생식 등의 즉석섭취식품에서 약 60%의 분포도를 보이고 있으며 분리 Enterococcus중 E. facium은 약 20%로 검출되었으나 E. faecalis는 전혀 검출되지 않았으며, 분리균주들의 항생제 내성은 비교적 높지 않은 것으로 나타나 위험성은 높지 않은 것으로 판단되었다. 비가공 농수산 식품소재와 생식제품에 오염되어 있는 세균성 식중독균인 B. cereus, Enterococcus 및 가공쌀 부패효모의 NaCl, 유기산과 에탄올의 생육저해 효과를 확인한 결과, B. cereus를 30% 에탄올에서 5분간 처리하였을 경우 생육저해효과를 보였다. 또한 처리시간이 경과함에 따라 뚜렷한 증식저해 효과를 나타냈으며 NaCl(10%)-ethanol (20%) 혼합액에서 5분, 10분간 처리한 경우 모든 균에서 생육이 저해되었다. In three hundred and twenty seven samples, the total aerobic bacteria count showed approximately 102 to 106 CFU/g and the highest coliform count was 104 CFU/g, Escherichia coli was detected in seven samples and Clostridium perfringens was detected in six samples. However, prevalence of Bacillus cereus was more than 30% samples. In Saengsik, the total aerobic bacteria count showed more than 107 CFU/g for some samples, but B. cereus was contaminated in more than 40% samples. Therefore, these results suggest that pre-treatment with sanitizer for removal of B. cereus in such the produces be necessary. Selective detection and enterotoxin gene detection of the B. cereus, B. thuringiensis, and B. mycoides from B.cereus group by PCR was performed. This study was performed PCR by gyrB gene target of B. cereus group. gyrB gene PCR was able to distinguish between B. thuringiensis and B. cereus, whileas B. mycoides was identified by morphological characteristics on nutrient agar. The typical pattern shown in cry gene PCR was able to distinguish between B. thuringiensis, B. thuringiensis subsp. kurstaki, and B. cereus as well as B. mycoides. Specific amplifications and good differentiations were obtained by using those strains, suggesting the possibility of the effective method described to identify the B. cereus group in food sample. The nheA gene was detected in some strains by PCR. B. cereus group strains, food-poisoning strains, were tested for enterotoxin production, using the B. cereus enterotoxin reverse passive latex agglutination technique and the Bacillus diarrheal enterotoxin visual immunoassay technique, according to the manufacturers' instructions. Amounts of the produced enterotoxin were evaluated with index values derived from the Oxoid and Tecra reading scale. Not only the B. cereus but also the B. thuringiensis and B. mycoides are potentially toxigenic strains. To evaluate the vancomycin resistance of Enterococcus spp.(VRE) from Saengsik and Sunsik, 39 Enterococcus, 16 strains from Saengsik and 23 strains from Sunsik, were ultimately isolated. However, E. faecalis was not detected in those foods. Minimum inhibitory concentrations of vancomycin against the isolates were below 4 μg/mL and no strains evidenced profound levels of resistance. The antibiotic resistances of isolates were relatively low, and this low vancomycin resistance was similar to that evidenced by Enterococcus isolates obtained from the other foods. The growth of B. cereus was not affected to exposure for five minutes in 20% ethanol while inhibition was shown in 30% ethanol solution. As being exposed longer, B. cereus decreased more effectively. We found synergistic growth inhibition on B. cereus when the combination treatment of 10% sodium chloride and 20% ethanol solution for five minutes was tested. Its viable counts was reduced after five minute treatment and almost did not show in ten minutes treatment. Enterococcus spp. was inhibited when the combination treatment of 20% ethanol and 1% lactic acid solution for ten minutes. However, Enterococcus spp. was not affected to exposure in 10% sodium chloride and 20% ethanol solution. For yeast, although growth inhibition effect by the sanitizer of 20% ethanol, 1% acetic acid, or 1% lactic acid respectively were very low, the combined sanitizer of 20% ethanol and 1% acetic acid, or 1% lactic acid showed very high sterilizing effect toward the yeasts at 50℃. This result may be applied to the effective pre-treatment of many ready-to-eat foods and agricultural food stuffs, especially uncooked stuffs, without any hazards from special sanitizers and nutritional loss from harsh sterilization.
