본 연구는 마늘과 양파의 첨가형태와 첨가량에 따른 저장 중 식육과 식육제품의 이화학적 성상, 항산화 및 항균효과를 측정함으로써 마늘과 양파를 이용한 기능성 식육가공품을 제조하고 평가하기 위하여 실시하였다. 마늘과 양파의 이화학적 성상, 항산화 및 항균 활성에 대한 최적의 조건을 선별하기 위하여 가열 전 열처리와 추출 용매에의한 차이 (물, 메탄올)를 평가하였다 (Chapter 3-1). 마늘과 양파의 항산화 효과는 추출용매에 따라 뚜렷한 차이가 발견되었다. 즉, 메탄올로 추출한 경우 antiradical 활성과 환원력 그리고 총 페놀성 화합물의 추출에 있어서 수용성추출물보다 더 효과적인 반면, 수용성 추출물은 iron chelating 활성과 추출수율이 메탄올 추출물보다 더 효과적이었다. 또한 마늘의 DPPH-radical 소거활성은 수용성 추출물보다 메탄올 추출물에서 더 높게 나타났다. 마늘에 존재하는 수용성 항균물질들은 열처리에 의해 그 활성을 잃은 반면, 열처리에 의해 새로운 메탄올성 항균물질들이 생성되었다. 그 결과, 가열 처리하지 않은 마늘로부터 추출된 수용성 물질들 (WEFG)과 가열 처리된 마늘로부터 추출된 메탄올성 물질들 (MEHG) 이 E. coli O157:H7 과 L. mon℃ytogenes에 대하여 농도 의존적인 항균 활성을 보였고, WEFG가 MEHG보다 더 뛰어난 항균활성을 보였다. 그러나 양파 추출물들은 E. coli O157:H7 과 L. mon℃ytogenes에 대한 항균 활성을 보이지 않았다. 육제품내에서 마늘과 양파 추출물의 효과를 살펴보기위해 마늘과 양파 추출물이 첨가된 돈육 페티의 이화학적 성상, 항균 및 항산화 효과가 평가되었다 (Chapter 3-2). WEFG와 MEHG 의 단독 혹은 복합첨가 (WEFMEHG)는 돈육 페티의 pH 값과 적색도 (a)를 감소시킨 반면 황색도 (b)를 증가시켰다. 마늘 추출물 (단독첨가 WEFG 0.5%, MEHG 0.5%, 복합첨가 WEFG + MEHG 1.0%)과 양파 추출물 (단독첨가, WEHO 0.5%, MEHO 0.5%, 복합첨가 MEHO + MEHO 1.0%) 의 이용은 신선한 돈육 페티의 지방산화를 (Thiobarbituric acid reactive substances, TBARS) 제어하는 효과를 가지고 있었으며, 특히, 복합첨가구인 WEFMEHG (1.0%) 와 WEFMEHO (1.0%)는 butylated hydroxyl toluene (BHT 0.01%)과 유사한 항산화 활성을 보임으로써 단독첨가구에 비하여 활성이 더 우수하였다. 특히 마늘 추출물들은 돈육 패티의저장 중 Enterobacteriaceae의 성장을 억제시켰고, 대조구와 비교하여 유도기간을 최소 3-4일 연장시켰다. 마늘과 양파의 추출물들이 육제품에 내에서 항산화 및 항균효과를 보임에 따라 신선한 양파와 마늘균질물의 일정한 함량을 직접 돈육 패티의 제조 시 첨가하여 냉장저장 중 이화학적 특성과 항산화 및 항균효과를 측정하였다 (Chapter 4-1,2). 마늘의 경우 건조물의 0.5와 1.0%에 해당하는 마늘 균질물 1.4와 2.8%를 첨가한 후 이화학적 특성과 항산화 및 항균효과를 측정한 결과, 마늘 균질물의 첨가 수준에 상관없이 돈육 패티의 pH값과 적색도 (a)를 감소시켰으나 황색도 (b)는 증가시켰다. 마늘 균질물을 첨가한 돈육패티들은 오히려 산화 생성물 (hexanal과 총 휘발성 산화화합물)의 생성을 가속화 시킴으로써 오히려 대조구보다 빠르게 산화되었다. 하지만 마늘 균질물을 첨가한 패티는 냉장저장 중 Enterobacteriaceae의 성장을 억제시켜 저장성을 증진시켰다. 한편 양파 균질물의 첨가에 (0.5, 1.0, 1.5, 2.0%) 따른 냉장저장 중 돈육패티의 이화학적 성상을 평가한 결과 (Chapter 4-2), 돈육패티의 적색도 (a)를 감소시킨 반면, 1.5% 이상의 양파 균질물의 첨가는 황색도 (b)를 증가시켰다. 그러나 TBARS, 총균수 그리고 Enterobacteriaceae에 있어서 양파 균질물의 첨가효과는 뚜렷하게 나타나지 않았다. 전반적으로 양파 균질물의 2.0% 수준까지는 신선한 돈육 패티의 지방산화와 미생물성장에는 영향을 미치지 않았다. 냉장 (8℃) 및 냉동 (-10℃) 저장하는 동안 돈육의 부위와 (삼겹, 등심) 포장방법 (함기, 진공)에 따른 이화학적 특성 및 미생물적 부패의 정도를 평가하고, 저장 중 지방산화와 미생물의 성장을 억제하기 위하여 마늘과 양파의 분말을 이용하였다 (Chapter 5). 동결 및 냉장 저장 중에 돈육 삼겹은 등심에 비해 더 높은 TBARS 값, pH 값과 저분자 휘발성 화합물이 많이 검출된 반면에 돈육 등심은 삼겹에 비해 더 높은 유리지방산 (FFA)가와 고분자 휘발성 화합물이 생성되었다. 냉장 저장 중에 있어서 진공 포장은 함기 포장과 비교 시 돈육 삼겹과 등심의 화학적 변질과 미생물의 성장을 억제하였다 (Chapter 5-1). 한편 돈육 삼겹과 등심을 동결 저장하는 동안에는 포장 방법에 따른 차이는 나타나지 않았다 (Chapter 5-2). 0.5%의 마늘 또는 양파 분말이 주입된 돈육 삼겹과 등심은 마늘 또는 양파 분말이 주입되지 않은 대조구에 비해 더 낮은 TBARS가, 유리지방산가, 과산화물가, 총균수 그리고 대장균군수를 보였다 (Chapter 5-3). 또한 0.5%의 마늘 또는 양파 분말은 돈육 삼겹과 등심에서 발생되는 휘발성 산화물의 생성을 억제하였으며, 특히 aldehyde 류(hexanal) 화합물들의 생성이 억제되었다. 따라서 본 연구의 결과를 종합하면, 마늘과 양파의 추출물은 추출용매에 따라 항균 및 항산화 효과가 차이를 보였으나, 육제품 내에서 모두 항산화 효과를 뚜렷히 나타내었고, 마늘추출물의 경우 항균효과도 높았다. 하지만 마늘과 양파는 균질물 자체로는 항산화 효과는 미미하였고, 마늘균질물(1.4~2.8%)의 경우 약간의 항균효과를 보여주었다. 한편 삼겹과 등심에 주입된 마늘과 양파의 분말은 냉장 저장 중 지방산화와 미생물 성장을 억제시켰다. 따라서 이러한 마늘과 양파 조제물들은 합성 항산화제와 항균제를 대체할 수 있는 천연의 항산화 및 항균제로 사용될 수 있을 것으로 기대된다.

This study was conducted to investigate physic℃hemical properties, and antioxidative and antimicrobial activities of meat and meat products as affected by the addition types and levels of garlic and onion, and propose optimum preparative conditions in order to manufacture functional meat and meat products. To screen the optimum conditions for physic℃hemical properties and antioxidant and antimicrobial activities of garlic and onion, the effects of pre-heating and different solvents (water and methanol) on these characteristics were evaluated (Chapter 3-1). Antioxidant activities of garlic and onion were affected by extraction solvent. In both garlic and onion, methanol was more effective than water in total phenolic content and reducing power, whereas, the latter was more effective than the former in extraction yield and iron chelating ability. DPPH-radical scavenging activity of garlic was higher in methanol extracts than in water extracts. Water-soluble components extracted from garlic were altered by thermal treatment, resulting in reduced antimicrobial activity, whereas, heating the garlic produced new methanol-soluble compounds containing antimicrobial activity. Thus, water extract from fresh garlic (WEFG) and methanol extract from heated garlic (MEHG) showed distinctive antimicrobial activities against E. coli O157:H7 and Listeria mon℃ytogenes, concentration-dependently, and WEFG showed better antimicrobial activity than MEHG. However, onion extracts did not show any antimicrobial activity. Physic℃hemical properties and antioxidant and antimicrobial activities of pork patties with garlic and onion extracts were evaluated (Chapter 3-2). The addition of garlic and onion extracts decreased pH and redness (a) values of fresh pork patties, whereas, yellowness (b) values were increased with the addition of garlic extracts. Pork patties with garlic homogenate (1.4 and 2.8%) were even oxidized faster than control because fresh garlic homogenate accelerated development of oxidative product (hexanal and total oxidative products) in pork patties. However, 1.4 and 2.8% of garlic homogenate inhibited the growth of Enterobacteriaceae. Based on the physic℃hemical properties of pork patties with onion homogenates (0.5, 1.0, 1.5 and 2.0%) during refrigerated storage (Chapter 4-2), redness (a) values decreased with the addition of onion homogenate, whereas, yellowness (b) values increased when the addition level of onion was higher than 1.5%. However, no differences in TBARS and microbial counts were observed with the addition of onion homogenate. Overall, up to 2.0% of onion homogenate did not affect lipid oxidation and microbial growth in fresh pork patties. Lipid oxidation, oxidative products and microbial growth as affected by pork meat cut and packaging method during refrigerated (8℃) and frozen (-10℃) storage were evaluated, and garlic and onion powder were used for the inhibition of chemical deterioration and microbial spoilage in fresh pork belly and loin during refrigerated storage (8℃) (Chapter 5). In both frozen and refrigerated storage, pork belly had higher TBARS, pH values and oxidative volatiles with low carbon numbers (C 10) than belly. In refrigerated storage, vacuum packaging inhibited the chemical deterioration and microbial growth in both belly and loin, as compared to the aerobic packaging (Chapter 5-1). However, most parameters tested did not show significant differences by packaging method during frozen storage (Chapter 5-2). Pork belly and loin injected with 0.5% of garlic or onion powder had lower TBARS, FFA, peroxide values and total plate counts and Enterobacteriaceae than those without injection, except for few cases (Chapter 5-3). In addition, garlic and onion powders suppressed the formation of oxidative volatile compounds in both belly and loin, particularly aldehydes (hexanal). In conclusion, although antioxidant and antimicrobial activities of garlic and onion extracts showed differences by extraction solvents, their antioxidant effects were applicable to the meat products. In addition, significant antimicrobial activities of garlic extracts were observed. However, antioxidant activities of garlic and onion homogenate were not observed but garlic homogenates (1.4~2.8%) showed antimicrobial activities. Garlic and onion powder injected into pork belly and loin inhibited lipid oxidation and microbial growth during refrigerated storage. Therefore, these garlic and onion preparations should be used as natural antioxidants and antimicrobial agents in meat and meat products to substitute synthetic antimicrobial agents and antioxidants.

In both frozen and refrigerated storage, pork belly had higher TBARS, pH values and oxidative volatiles with low carbon numbers (C 10) than belly. In refrigerated storage, vacuum packaging inhibited the chemical deterioration and microbial growth in both belly and loin, as compared to the aerobic packaging (Chapter 5-1). However, most parameters tested did not show significant differences by packaging method during frozen storage (Chapter 5-2). Therefore, these garlic and onion preparations should be used as natural antioxidants and antimicrobial agents in meat and meat products to substitute synthetic antimicrobial agents and antioxidants.
