Histamine, lysine, ornithine, tyrosine을 전구체로 하는 BAs screening broth 에서 Enterococcus 속의 BAs 생산성은 Bacillus 속의 BAs 생산성보다 높았다. 특히, tyramine의 경우, 대부분의 Enterococcus 속 분리주는 Bacillus 속 분리주 보다 약 20배 가량 높은 생산성을 보였다. 항균활성을 측정하기 위하여 한약재 18가지를 95% ethanol로 추출하여 사용하였으며, 높은 항균활성을 띈 한약재 추출물에 대하여 tyramine저감화 실험을 시행하였다. Tyramine 고생산성 Enterococcus 속 분리주와 purescine 고생산성 Bacillus 속 분리주에 대하여 높은 항균 활성을 가진 Coptis japonica, Platycladus, Gardenia seeds, Aphis chinensis, Terminalia chebula, Pinus densiflora 등이 1차적으로 선별되었다. 선별된 추출물을 첨가하여 tyramine의 저감화 가능성을 검토한 결과 Coptis japonica, Aphis chinensis, Terminalia chebula 에서 대조구와 비교하여 30%이상 저감화 효과가 있었다. To this, the contents of BAs in 15 of commercial Doenjangs purchased from retail markets were determined using RP-HPLC. Moreover, high BAs producing microorganisms were selected with amine screening medium and their BAs productivities were also analyzed.
Most of the samples were shown safe and the average content of each of the eight BAs such as typramine, phenylethylamine, putrescine, cadaverine, histamine, tyramine, spermidine, and spermine were less than 500 mg/kg, allowable limit suggested by the US Food and Drug Administration for scombroid fish and fishery product. Whereas, the contents of tyramine, histamine and putrescine were highly detected to 84.0-818.6 mg/kg, 41.9-1068.9 mg/kg, and 43.1-1188.8 mg/kg, respectively. Especially, high tyramine content in the foods could be considered as a hazard factor. The aerobic plate counts in all samples ranged from 8.3 logCFU/g to 10.2 logCFU/g, and then genus Bacillus and genus Enterococcus had the higher proportions compare with other microorganisms in the foods. The BAs productivities of the genus Enterococcus isolates were higher than those of genus Bacillus isolates in culture condition using amino acid enrichment medium. Especially, tyramine productivity of almost of genus Enterococcus isolates showed more than 20 times higher than that of genus Bacillus isolates. To investigate the antimicrobial activity, total 18 samples of medicinal herbs were extracted with 95% ethanol. In addition, the reduction effect of tyramine formation were tested by improve medium with medicinal herbs extracts which primarily chosen by antibiotic activities. The EtOH extracts of Coptis japonica, Platycladus, Gardenia seeds, Aphis chinensis, Terminalia chebula, and Pinus densiflora were primarily chosen due to high antimicrobial activities against an isolate of the genus Enterococcus and a putrescine high producing Bacillus isolate. Moreover, the change in tyramine content in liquid culture of tyramine high producing isolates with the EtOH extracts of Coptis japonica, Aphis chinensis, and Terminalia chebula were determined. The tyramine reduction rate of these extracts showed over 30% compared with the control group. Coptis japonica, Aphis chinensis, Terminalia chebula, and garlic which already presented about biogenic amine reduction activity, were applied to the Doenjang. Tyramine and histamine contents were decreased in Doenjang which added garlic and Ahpis chinensis during dipping step. One of the reasons likely that the viable cell counts of genus Enterococcus from the Doenjang was decreased about 3-6 log/CFU/g during the dipping period. Therefore, control of genus Enterococcus was effectively to biogenic amines reduction in Doenjang and medicinal herbs could use to decrease biogenic amines.
