행주와 수세미는 식품의 교차오염에 일으키는 병원성 세균의 활동 원천으로 이용 될 수 있다. 본 연구는 다양한 행주 및 수세미에 Staphylococcus aureus, Escherchia coli O157:H7, Listeria monocytogenes, Salmonella Typhimurium와 같은 병원성 미생물과 유기물질(TSB, 쌀뜨물, 육수)을 함께 오염시켰을 때의 병원성 미생물의 생장 경향을 조사하였다. 또한 병원성 미생물이 오염된 행주와 수세미에 다양한 유기산과 상업적 살균 소독제를 사용하여 항균 효과를 평가하였다. 멸균된 5가지 행주와 수세미(cellulose sponge, color sponge, cotton dish cloth, filter sponge, nylon dish cloth)에는 총 4종의 균주를 0.1 ml씩 취하고 TSB, 쌀뜨물, 육수를 2 ml씩 각각 접종한 다음 25℃ 배양기에 저장하였다. 병원성 미생물의 생장 경향을 측정하기 위해서 정해진 시간 (0, 3, 6, 9, 12 그리고 24시간)에 따라 시료 (TSA: Difico)를 37℃ 배양기에서 꺼내 계수하였다. 총 4종의 병원성 미생물을 Tryptic Soy Broth (TSB: Difico)와 함께 접종시켰을 때 병원성 미생물은 cellulose sponge에서 12시간 경과 후 한계 수준 이하로 자랐으나 이를 제외한 나머지 행주 및 수세미에서 병원성 미생물의 수준이 두드러지게 증가하여 결과적으로 24시간 경과 후 높은 농도의 오염을 나타내었다. E. coli O157:H7 와 S. aureus를 각각 쌀뜨물, 육수와 함께 접종하였을 때 유사한 형태의 결과가 관찰되었다. 그러나 nylon dish cloth에서 12시간 경과 후 2종의 병원성 미생물이 color sponge, cotton dish cloth, filter sponge 보다 낮은 농도로 생육 되었다. 또한 color sponge, cotton dish cloth, filter sponge를 육수와 함께 접종하였을 때 S. aureus가 12시간 경과 후부터 높은 농도로 증가하여 차이를 나타내었다. 상업적 살균소독제 및 유기산으로 처리한 E. coli O157:H7 와 S. aureus 저해 효과 실험에서는 동일한 처리 시간에 처리에 따른 항균 결과 처리 시간 30분에서 과산화수소계, 5.0% 농도의 젖산과 프로피온산에서 균의 수가 detection limit 이하로 나타나 처리 농도에 따른 E. coli O157:H7 와 S. aureus에 대해 높은 살균 효과를 나타내었다. 그러나 염소계와 2.5% 농도의 유기산 처리에서 E. coli 가 5.4 log-4.0 log, S. aureus가 6.3 log-3.1 log 수준으로 감소하여 5.0% 농도의 유기산 보다 낮은 항균효과를 나타냈다. 처리 시간 60분에서는 처리시간 30분과 동일한 결과를 나타내었다. 동일한 처리 시에 처리시간 경과 후 E. coli, S. aureus 에 대한 항균 효과에서는 처리 30분경과 후 5.0% 농도의 4종에 유기산과 과산화수소에서 3.1 log 수준 이하로 감소하여 60분경과 후 균수가 detection limit 이하로 나타나 E. coli, S. aureus 에 대해 높은 항균 효과를 나타내었다. 본 연구 결과를 통해 상업적 행주와 수세미에서 병원성 미생물은 유기물질에 의존하여 높은 농도로 생존할 수 있으며, 유기산과 과산화수소계와 같은 화학적 소독제로 처리하였을 때 병원성 세균을 제어하는데 효과적이었다. 그러므로 가정과 상업공간에서 교차오염을 제어할 수 있는 적절한 살균, 소독 방법 및 위생적인 관리가 요구된다.

Dish sponges and cloths could act as sources for pathogenic bacteria through cross-contamination to foods. This study was conducted to investigate the survival and growth of pathogenic bacteria such as Escherichia coli O157:H7, Salmonella Typhimurium, Listeria monocytogenes, and Staphylococcus aureus on various commercial dish sponges and cloths when contaminated with organic materials (laboratory broth, rice water, and beef gravy) and effects of chemical sanitizers and organic acids on inhibiting pathogens contaminated on dish sponges and cloths. Five commercial sponges or cloths (cellulose sponge, color dish cloth, cotton dish cloth, filter sponge, and nylon dish cloth) were sterilized, inoculated with pathogens using cell suspension with tryptic soy broth, rice water or beef gravy and stored at room temperature (25°C). Survived levels of pathogens were enumerated on tryptic soy agar at selected time intervals (0, 3, 6, 9, 12, and 24 h) during storage time. When pathogens were inoculated with tryptic soy broth, all four pathogens did not grow in cellulose sponge until 12 h of storage however levels of pathogens on other samples were significantly increased and resulted in high populations (ca. 107-8 log CFU/samples) after 24 h of storage. When E. coli O157:H7 and S. aureus were inoculated with rice water or beef gravy, similar patterns of results were observed however levels of both pathogens in nylon dishcloth after 12 h of storage were significantly lower than in other three samples (color dish cloth, cotton dish cloth and filter sponge). Also, there were significantly differences in levels of S. aureus between cotton sponge and color dish cloth or filter sponge after 12 h of storage when beef gravy was added. When chemical sanitizers and organic acids were treated on dish sponge and cloths, treatments with hydrogen peroxide based sanitizer, lactic acid and propionic acid were the most effective to reduce levels of E. coli O157:H7 and S. aureus whereas treatment with chlorine sanitizer did not significantly reduce levels of pathogens. Levels of both pathogens were under detection limit (2.3 Log CFU/sample) when treated with hydrogen peroxide based sanitizer and 5% propionic acid. There is no significant difference on the results depending on treatment time for 30 and 60 min. As the results, pathogens contaminated on commercial sponges or cloths could grow rapidly depending on materials. Therefore, proper sanitizing methods should be applied to prevent the cross-contamination of pathogens by dish sponges and cloths in foods and environments. Also, from results of this study, some types of organic acids and chemical sanitizer such as hydrogen peroxide could be used as effective ways to inhibiting pathogens contaminated on dish sponges and cloths.
