RA (1, 2, 4, 또는 8 mg/kg)의 급성 투여는 probe trial의 platform crossing 횟수에 대해 종형 용량의존성을 보였으나 그 효과는 유의적이지 않았다. 그러나 아만성 투여는 probe trial의 platform crossing 횟수를 대조군에 비해 유의적으로 증가시켰으며 종형 용량의존성을 보였다. 특히 아만성 투여군 중 2 mg/kg 투여군의 platform crossing이 가장 높았으며 이는 대조군에 비해 통계적으로 유의한 차이로 나타났다 (P < 0.05). 이러한 결과로 보아 RA는 학습 및 기억의 강화 효과가 있으며 그 효과는 POP 억제에 의해 나타나는 것으로 보인다. 다음으로, 용안육의 학습 및 기억의 작용을 조사하였다. 용안육은 용안나무 (Euphoria longan (Lour) Steud.)의 가종피로 전통 동양의약에서 건망증, 불면증, 초조 등의 질환에 오래 전부터 활용되어 온 한약이다. 본 연구에서는 용안육 물추출물 (ELE)의 학습과 기억에 대한 효과를 행동시험인 수동회피시험을 통해 증명하고, 그 기전을 면역조직화학 염색법을 사용하여 조사하였다. It is well known that inhibition of prolyl oligopeptidase (POP, E.C. 3.4.21.26) elevates the levels of brain arginine-vasopressin, substance P and other biological substrates of POP that are involved in memory-related function. In this study, we observed that rosmarinic acid (RA) inhibits POP activity with an IC50 of 63.7 μM. Subsequently, we investigated the cognitive-enhancing effects of RA employing the Morris water maze paradigm. When the number of platform crossings was measured, we found an inverted U-shaped dose-response curve with acute (4 training days) and sub-chronic (2 or 3 weeks) treatments of RA (1, 2, 4, or 8 mg/kg). Moreover, there was a significant increase in the platform crossings at a dose of 2 mg/kg, although there were no significant changes in the escape latency during training trials or the time spent in the target quadrant during the probe trial. Taken together, these results suggest that RA has a cognitive-enhancing effect, and that this effect may be mediated by its ability to inhibit POP. On the other hand, the memory-enhancing effects of The fruit of Euphoria longan (Lour.) Steud. (Sapindaceae) was also investigated. Dried Euphoria longan fruit is prescribed as a tonic and for the treatment of forgetfulness, insomnia, and palpitations caused by fright in traditional Chinese medicine. In the present study, the authors investigated the effects of the aqueous extract of the fruits of Euphoria longan (ELE) on learning and memory using behavioral and immunohistochemical methods in mice. By oral treatment with ELE for 14 days, the latency time was significantly increased versus vehicle-treated controls (P < 0.05) in the passive avoidance task. Immunohistochemical studies revealed that the number of cells immunopositive for brain-derived neurotrophic factor (BDNF), phosphorylated cAMP response element binding protein (pCREB), or phosphorylated extracellular signal-regulated kinase (pERK) 1/2 was significantly increased in the hippocampal dentate gyrus and CA1 regions after ELE treatment for 14 days (P < 0.05). Doublecortin (DCX) and 5-bromo-2-deoxyuridine (BrdU) immunostaining also revealed that ELE significantly enhanced immature neuronal survival, but not neuronal cell proliferation in the subgranular zone of the dentate gyrus. These results suggest that subchronic administration of ELE enhances learning and memory, and that its beneficial effects are mediated, in part, by BDNF expression and immature neuronal survival.
