Mammalian neural development is continued to form neural circuit and synapse from the birth. Not only generation of the tissue but also differentiation of neuronal cell is activated by programmed cell death. Especially, autophagy which has been known as type II cell death is not elucidated. Melatonin is a pineal hormone to regulate daily circadian rhythm, and has a role in anti-oxidative molecule. During 2~4 months from birth, there is a transient melatonin deficiency because of a lack of neural circuits to produce it. The purpose of this study was to verify the beneficial effects of exogenous and/or endogenous melatonin on the brain development. 7-week-old Sprague-Dawley rats were randomly divided into the two groups with/without melatonin. Treatment of melatonin was performed via subcutaneously injection at 19:00. It was indirectly injected to pups through maternal rat on embryonic days, and it was directly injected to them on postnatal days. As a result, exogenous melatonin induced a significant weight loss in maternal rats compared to the control (p<.01). Treatment of melatonin was significantly increased a body weight in male pups on P21 (p<.05). Histological analysis using Golgi cox staining showed that exogenous melatonin promotes gliogenesis, and assists to neuronal cell arrangement in cerebral cortex. As a result of double staining, the formation of skeleton and cartilage was confirmed both two groups. Using RT-PCR, neuronal cell differentiation and cell death mechanism was confirmed. The expression of NG2 was significantly increased on E20 (p<.01) melatonin groups than control. Compared to control, melatonin group was shown GFAP mRNA level also significantly increased on E20 (p<.01), P3, and P7 (p<.05). The expression of HIF-1a, as a marker of response to hypoxia, was increased at P7 on control, but was not statistically changed by melatonin treatment. As a results of cell death, not only autophagy was significantly increased on melatonin groups at P28 both mRNA and protein level, but also the expression of Bax, proapoptotic molecule, was increased on melatonin groups at P3 (p<.05). Related to aging, administration of melatonin for 6 month was induced significant decrease of Bcl2 mRNA level (p<.05). These results show that exogenous melatonin promotes the maturation of neuron as well as the formation of skeleton and plays an important role as a protective mediator from oxidative stress. In conclusion, we suggest that melatonin is an effective assistance as a nutritional supplement for neural development.
