The substance was purified with HPLC and its properties were analyzed. The isolated substance was peptidal compound with lipid moieties and its molecular mass by ESI/MS/MS, m/z 3400. It was tentatively named BSAP-254. The BSAP-254 had an adequate effect on the B. cereus group: Bacillus anthracis, B. cereus, Bacillus mycoides, Bacillus pseudomycoides, Bacillus thuringiensis, and Bacillus weihenstephanesis. Its antagonistic spectrum was narrow on lawn cell plates (<10 μg). Several Gram-negative and positive pathogenic bacteria such as Salmonella enterica, Salmonella enteritidis, Staphylococcus aureus, and Listeria monocytogenes were not affected. The growth of major soybean-fermenting bacteria within the same genus such as B. subtilis, Bacillus licheniformis, Bacillus amyloquefaciens and lactic actid bacteria such as Enterococcus faecium, Lactobacillus plantarum, Pediococcus pentosaceus, and Strepococcus pyogeneus was not inhibited. The range of pH stability of the purified antagonistic substance was wide, from 4.0 to over 10.0, and the substance was thermally stable up to 60oC. In the various enzyme treatments, the antagonistic activity of the purified substance was diminished with proteinase K, protease, and lipase. Its activity was partially destroyed by esterase. The BSAP-254 was stable in organic solvents such as acetone, chloroform, ethanol, ether, hexane, and methanol. The production of BSAP-254 of B. subtilis SC-8 was influence on pHs, temperatures, carbon sources, nitrogen sources, and minernal sources. However, the highest production of biomass and BSAP-254 (5~6 g/L) were in the basal medium (LB medium). Minimum inhibitory concentration (MIC) of BSAP-254 against B. cereus in liquid culture medium was 8 μg/mL. B. cereus (105 cells) was not grown in the concentration of 30 μg/mL of BSAP-254. Spores (103 spores) of B. cereus did not grow at all in the presence of 5 μg/mL of the purified antagonistic substance. K+ was rapidly released within 10 min from B. cereus exposed to BSAP-254. BSAP-254 inhibited B. cereus in 10 min and reduced a 50% of the population of B. cereus within 30 min. We assumed that the narrow inhibitory mechanism of BSAP-254 on the B. cereus group is due to structural differences of peptidoglycan between the B. cereus group and other bacteria cell wall. Stress proteins induced in B. cereus exposed to BSAP-254 were also analyzed using 2-DE coupled with MS. The heat shock protein (Hsp60), which is generally expressed after a sudden increase in the ambient temperature, was increased in B. cereus exposed to BSAP-254.
