This study was carried out to evaluate the residual characteristics and safety of difenoconazole and pymetrozine in water celery during cultivation period and to investigate the changes in concentration of the pesticide residues in raw water celery during processing and storage. For elucidating the decline field experiment for the test pesticide residues, the test pesticides were sprayed once and twice by seven days interval on water celery growing in greenhouse, followed by sampling from two treatment plots at 0, 1, 2, 3, 5, 7, 9, 10, 13 and 15 days after last spraying. In order to determine the residue changes of the pesticides during processing and storage, processing of raw water celery was done by washing, steaming, blanching, and microwave heating, and the harvest crop was stored at 4℃ for 12, 24, 48, and 72 hours. Residues of difenoconazole and pymetrozine in raw and processing products of water celery were analyzed using a gas chromatograph equipped with electron capture detector and a high performance liquid chromatograph equipped with diode array detector, respectively. Possible risk of the pesticides in raw water celery was assessed by using their acceptable daily intake. The limits of detection of difenoconazole and pymetrozine were 0.001 and 0.01 mg/kg and the limits of quantitation were 0.004 and 0.04 mg/kg, respectively. Recoveries were ranged from 79.4 to 102.1%. Amounts of difenoconazole residues in once and twice spray plots ranged from 0.24 to 2.12 and from 0.35 to 2.53 mg/kg, respectively, while those of pymetrozine residues in two treatment plots ranged from 0.29 to 3.17 mg/kg and from 0.46 to 4.23 mg/kg, respectively. Residue amounts of the test pesticides in decline trials dissipated time-coursely to make their biological half-lives to 4.9 and 5.7 days for difenoconazole and 4.5 and 5.3 days for pymetrozine. Estimated daily intakes of difenoconazole and pymetrozine residues in raw water celery at harvest were 0.07-1.47 and 0.07-1.79% of their acceptable daily intakes, respectively, representing that the residue levels of the pesticides were evaluated as safe to consumers. Amounts of the pesticide residues in the test crop removed by washing with tap water were 92.2% for difenoconazole and 82.9% for pymetrozine, indicating that washing was necessary to remove large amount of these pesticide residues in water celery. Processing factors of the pesticides by washing, blanching, steaming and microwave heating were less than 0.17. No significant residual changes were observed from both test pesticides during three days of storage at 4°C. These results indicated that washing with water as well as cooking process, such as blanching, steaming, and microwave heating was important to eliminate the pesticide residues from water celery.
