Gisela 5 is a well-known dwarfing rootstock for sweet cherry, bred through cross-breeding between P. cerasus × P. canescens. It is examined as very effective, convenient and economically significant dwarfing rootstock for intensive sweet cherry cultivation in temperate zone. The present study was carried out to evaluate the optimum cultural conditions for in vitro propagation of aforementioned Cherry rootstock. The study was focused on primary, secondary and rooting culture with regard to different standard and modified mediums as well as acclimatization. It was observed that MS medium containing half strength macro salt (MS1/2) supplemented with 0.5~1.0 mg․L-1 BA, 0.5 mg․L-1 IBA, 30 g․L-1 sucrose, and 7 g․L-1 agar was suitable for primary culture Shoot proliferation and growth were decreased significantly when MS culture medium concentration were reduced, while, number of shoot proliferation were increased with increase in BA concentration. However, combine effect of BA and IBA mixture were proved more effective, therefore, the best medium of secondary culture was MS+BA 0.5mg․L-1 +IBA 0.5 mg․L-1 +30 g․L-1 sucrose+7 g․L-1 agar. Further, in vitro the maximum rooting was observed on 1/2 MS medium supplemented with IBA 2.0mg․L-1, 30 g․L-1 sucrose, and 8 g․L-1 agar. On the other hand, to investigate the In vitro rooting and domestication assay was laborious, time consuming and extravagant. So, ex-vitro rooting experiment was designed. Plants were tested in different soil matrix ratio to know the most appropriate one. Vermiculite 1: Perlite 1: Peat-moss 1; was listed as the best soil matrix ratio for rooting and plant domestication, when shoots were grown in the ventilation culture flask. Study revealed that it is possible to produce numerous seedlings successfully by using this method.
