Objectives: This study is aimed to predict the cariogenic effects of fresh and dried fruits by analyzing salivary sugar clearance using high-performance liquid chromatography, and to compare the stabilities of sugars in saliva samples processed via four methods. Methods: To test sugar stability, saliva samples were obtained from five subjects after mouth-rinsing with a 10% (w/w) glucose or sucrose solution and processed via four methods (freezing, pyridine addition, butylparaben addition, and filtration). To test sugar clearance, saliva samples were obtained from five subjects before and up to 60 min after eating fresh or dried fruits. Glucose, sucrose, and fructose were measured by anionexchange chromatography with a pulsed amperometric detector using 50 mM sodium hydroxide solution at room temperature. Results: The long-term stabilities of sugars were highest using the frozen method, and filtration prevented sugar decomposition prior to freezing. Total sugar concentration in the salivary was higher after eating dried fruits than after eating fresh fruits. It is unclear whether sugar clearance was more rapidly after eating fresh or dried fruits. Conclusion: This analysis showed that dried fruits had a higher risk of causing dental caries than fresh fruits. This method could be used to quantitatively and objectively evaluate the cariogenic effects of foods.
