In this study, the ability of Mulberry leaves cultivated with Hericium erinaceum (MLHE-LM) and Robusta cultivated with Monascus ruber (RMR-LM) after fermentation from Leuconostoc mesenteroides extracts on treating obesity were examined. The control group was given distilled water for 6 weeks. The treatment groups were given with 50% MLHE-LM, 100% MLHE-LM, 10% RMR-LM and 20% RMR-LM. To induce the obese mouse for investigating the hypolipidemic effects, all groups were fed with the same high fat diets (60% Kcal fat diet) throughout the study. Body weight and food intake were recorded once a week at the appointed time. Adipocyte size of visceral fat was determined by histological analysis as the number of adipocytes compared to the area. For the glucose tolerance test, 6h fasted mice were injected intraperitoneally with glucose (1g/kg body weight) and blood glucose levels were determined from the tail vein at 0, 15, 30, 60 and 120 min. Serum biochemical analysis was examined from fasted blood serum. The mRNA expression level was quantified by real-time PCR. Body weight changes in 20% RMR-LM were lower than the control group, but the average food intake between treatment groups and control group was similar. The visceral adipose tissue was significantly decreased in 10% RMR-LM and 20% RMR-LM compared to the control group, and this lead to the significant decrease in adipocyte size in 20% RMR-LM. The total area under the curve (AUC) of blood glucose levels between 0 to 120 min were significantly lower in 10% RMR-LM and 20% RMR-LM compared to the control group. The serum AST level and ALT level were significantly decreased in 50% MLHE-LM and 20% RMR-LM compared to the control group. The hepatic mRNA levels of sterol regulatory element-binding protein 1 (SREBP-1), fatty acid synthase (FAS) and peroxisome proliferator-activated receptor gamma (PPARγ) in 20% RMR-LM were significantly lower than the control group. In conclusion, the 20% RMR-LM has anti-obesity effects which decreased the visceral fat and adipocyte size. The AUC of glucose determined by GTT also showed effects of 20% RMR-LM and 10% RMR-LM on glucose tolerance. These findings suggest that 20% RMR-LM can be used for inhibition of obesity and amelioration of diabetes and lipid metabolism.
