The fungal pathogen Fusarium graminearum causes Fusarium head blight that reduces yield and grain quality in wheat, barley, rice and maize. This fungus produces mycotoxin like thrichothecene and zearalenone. These toxins remain in grain and are harmful to livestock and human through contaminated food. Conidia play a role as primary inoculum for disease epidemics and study on conidial germination can provide development of disease control method in this fungus. Microcycle conidiation is that conidia directly produce conidia again instead of normal germination. This phenomenon has not been observed in F. graminearum. As screening the transcription factor mutant library previously generated in F. graminearum, we identified Mic1 deletion mutant that showed typical microcycle conidiation. We also performed RNA-sequence analysis to identify genes related to microcycle conidiation or regulated by Mic1 gene as comparing gene expression profiles in Mic1 deletion mutant and wild type strain during conidial germination. To characterize each gene function identified from RNA seq analysis, we performed targeted gene deletions and identified FGSG_11027, FGSG_11192 deletion mutants that showed microcycle conidiation. Also we found that treatment of diphenyleneiodonium, which suppresses the NADPH oxidase, resulted in microcycle conidiation in the wild type strain. This study will help to understand the evolutionary history in microcycle conidiation and make good use of disease control system development.
