Persimmon is one of the most frequently consumed fruits in north-east Asian countries including Korea, Japan and China. Fruits are classified as sweet and astringent types, and are consumed as fresh fruit or dried persimmon according to their types. In production area of dried persimmon products, large amount of persimmon peels are wasted despite their nutritional composition. In this study, therefore, to enhance utilization of persimmon peels as bio-resources, physicochemical properties and antioxidant activities of the powders obtained by drying, pulverization, and screening were investigated. Daebong persimmon peel powder classified as DP-0 (whole), DP-1 (60 mesh) through screening showed clearly different appearances and color values. In contrast to coarse and reddish color of DP-1, DP-3 showed a fine and bright-yellowish color. Approximate compositions of the powders showed no significant difference depending on particle sizes. The reducing sugar content of DP-3 was approximately 10% higher than that of DP-1. In contrast to the vitamin C contents, which showed no significant difference, total carotenoid content of DP-1 was about 35% higher than that of DP-3. Contents of antioxidants, including total phenolics and total flavonoids, increased inversely to the particle sizes of the powders, and DP-3 had the strongest antioxidant activities.
Although the usefulness of the peel has been demonstrated, study on the functional compounds from Daebong persimmon peel is much less studied. Thus, antioxidant activity-guided fractionation and purification were used to isolate antioxidant compounds from Daebong persimmon peels. Ethyl acetate fraction showed the strongest DPPH radical scavenging activity compared to those of other fractions. The ethyl acetate fractions were isolated using column chromatography systems. Three antioxidant compounds were finally isolated. Among them, compound 2 was identified as a myricitrin by LC/MS, 1H and 13C NMR analyses. In antioxidant activities, compound 2, identified as a myricitrin, showed the strongest antioxidant activity in both of DPPH and FRAP assay. Thus, myricitrin was found as a major antioxidant compound and principally responsible for the antioxidant activities of Daebong persimmon peels.
The utilization of persimmon peels as bioactive materials has been limited due to their heterogeneous composition. In this study, therefore, carbohydrate-hydrolyzing enzyme was applied to enhance the reducing sugar contents for vinegar fermentation. Response surface methodology (RSM) was used to determine the treatment conditions of optimal enzymatic hydrolysis for vinegar preparation from Daebong persimmon peel. It was hydrolyzed with different concentrations of Viscozyme L (0–4%) at different temperatures (40–60°C) for different lengths of time (2–10 h) followed by vinegar fermentation. Reducing sugar and soluble solids of the hydrolyzed juice were significantly affected by the hydrolysis conditions. Optimal condition was obtained with the enzyme concentration of 2.38 g/100 mL, reaction temperature of 49.19°C, and reaction time of 6.32 h. The titratable acidity of vinegar prepared from the juice hydrolyzed under the optimized condition increased by 2.76-fold (4.25 g/100 g). Acetic acid, citric acid, oxalic acid, and succinic acid were identified in the vinegar. Results suggest that the enzymatic hydrolysis pretreatment could be used for increasing the yield of the vinegar fermentation.
Finally, to evaluate utilization of the peels as novel bio-materials, the general composition and antioxidant activities of DPPV were investigated. The pH was slightly higher than that of other vinegars. The titratable acidity was 2.77%. The reducing sugar contents showed that DPV had slightly higher than that of other vinegars. The browning degree of DPV was lower than CPV in contrast to the turbidity. The results of Hunter’s color value have showed that overall values of CPV had significantly higher than DPPV and DPV. The total phenolic contents of DPV and CPV were 19.49 and 17.13 mg/100g GAE, respectively. The total flavonoid contents of DPPV (8.04 mg/100g CE) were two fold higher than that of DPV (3.85 mg/100g CE). The antioxidant activities, by DPPH and FRAP assays, of DPV showed stronger than those of other vinegars. Free sugars were mainly composed of fructose and glucose. Organic acids were presented in the order acetic acid, succinic acid, malic acid, tartaric acid, citric acid and oxalic acid. These results suggest that the Daebong persimmon peels could be utilized for vinegar production. Taken together our results, strong antioxidant activities and useful physicochemical properties of the Daebong persimmon peels and their vinegar seem to guarantee their utilization to a promising natural health beneficial product.
