Ergothionein 함량은 0.41 mg/ml DW 였다. 5 mg/ml과 10 mg/ml에서 천마열수추출물의 DPPH 그리고 ABTS radical 소거능의 범위는 31∼44% 였다. SOD 유사활성은 10 mg/ml과 25 mg/ml에서 각각 57%와 76% 였다. UVA를 조사한 Human Dermal Fibroblast (HDF) 세포에서 천마열수추출물은 UVA 조사에 의해 감소된 procollagen type I의 생성을 증가시키며, UVA 조사에 의해 증가된 matrix metalloproteinase-1 (MMP-1) 생성과 elastase-1의 활성은 감소시킨다. HDF 세포에서 UVA 조사는 procollagen type I의 mRNA 수준을 유의적으로 감소시키지만 고농도의 천마열수추출물은 감소된 mRNA 수준을 회복시킨다. 반대로, HDF 세포에서 UVA 조사는 MMP-1과 elastase-1의 mRNA 수준을 증가시키지만 천마 열수추출물이 UVA 조사에 의해 증가된 MMP-1과 elastase-1의 mRNA 수준을 유의적으로 감소시킨다. 따라서 이러한 연구 결과들은 천마열수추출물이 항산화 효과와 UVA가 조사된 인체 피부세포에서 피부 노화를 방지효과를 가지는 것을 뒷받침하고, 나아가 천마열수추출물이 피부노화를 방지하는 효과를 가진 피부기능성 소재로 활용이 가능함을 제시하였다.

Gastrodia elata Blume (GEB), a traditional herbal medicine, has been used to treat a wide range of neurological disorders such as paralysis and stroke and skin problems such as atopic dermatitis and eczema in oriental medicine. Therefore, this study was aimed to investigate antioxidant levels and capacity of aqueous GEB extract and its anti-aging effects in human dermal fibroblast cells (HDF). The total phenolic and flavonoid contents of GEB extract were 21.8 and 0.43 mg/g dry weight (DW), respectively. Ergothioneine contents of GEB extract was 0.41 mg/ml DW. The DPPH and ABTS radical scavenging activities of GEB extract at 5 and 10 mg/ml approximately ranged from 31% to 44%. The superoxide dismutase activity of GEB extract at 10 and 25 mg/ml was 57% and 76%, respectively. GEB extracts increased procollagen type I production and inhibited matrix metalloproteinase-1 (MMP-1) production, elastase-1 activity in UVA-irradiated HDF cells. UVA irradiation significantly decreased mRNA level of procollagen type I but higher doses of GEB extract recovered mRNA level in HDF cells. In comparison, GEB extract significantly decreased mRNA level of MMP-1 and elastase-1, which were markedly induced in UVA-irradiated HDF cells. Collectively, this result suggests that aqueous GEB extract has sufficient amounts and capacity of antioxidants to prevent skin aging signs in UVA-irradiated human skin cells, suggesting its potential as natural anti-aging (anti-wrinkle) products.
