This study was to improve the method for storing fresh raw ginseng seedling which used for year-round consumption of ginseng sprout. The quality of ginseng seedling surface-treated by chlorine dioxide (ClO2) gas was evaluated during storage at 4 °C, 10°C and 20°C. Currently, the raw ginseng seedling storing method was only low temperature storage. Gaseous ClO2 at 0, 0.3, 0.8, and 1.4 mg/L was used for the treatment in the humidification state of the chamber and the gas treatment time was set to 0, 5, and 10 minutes at each concentration. Weight loss of the treated ginseng seedlings by ClO2 was not significantly different among the treatment groups compared to the control. However, the weight reduction ratio (%) of ginseng seedling at 4 °C, 10 °C, and 20 °C storage was 5.93 ± 0.48, 6.18 ± 0.93, and 7.13 ± 0.30, respectively. It showed a significant difference in storage temperature. The hardness of ginseng seedling stored at 4 °C did not show significant differences between control groups and treatment groups at the range of 24.56∼32.65 kg/cm2 for 20 days. However the hardness of ginseng seedling stored at 10°C decrease from control groups (no treatment, 5 and 10 min moisturized only) at 16 days. The values were decreased to 4.26 ± 1.27, 6.15 ± 3.63, and 12.52 ± 5.37 kg/cm2, respectively, for control (CT), ClO2 0 mg/mL-5 min (ClO2 0-5), and ClO2 0 mg/mL-10 min(ClO2 0-10) at 20 days storage. At 20oC storage of the ginseng seedling, the reduction of hardness of the control groups was beginning at day 8, however, the treated groups was beginning at day 12 or later day. The hardness of each treatment group was sequentially decreased as the storage time increases. All ginseng seedling samples showed the less than 20 kg/cm2 of hardness for 20 days stored except for the treatment of ClO2 1.4 mg/mL-10 min (ClO2 1.4-10). The group treated with ClO2 1.4-10 showed the 4.3 log CFU/g reduction of the total microbial numbers and 4.7 log CFU/g for yeast and mold. Total microorganisms including yeast and fungi were observed tended to increase more during storage. Particularly the yeast and mold numbers in ginseng seedling rapidly increased at stored 20 oC. The total microorganism, yeast and mold were 1.4∼1.9 and 1.6∼1.8 log CFU/g, respectively, at 4 oC storage, while 2.1∼2.5 and 2.3∼3.5 log CFU/g at 20oC. The lower store storage temperature for the ginseng seedling, the higher ClO2 gas concentration, and the longer treatment time were proportional to the hardness, weight loss ratio, and antimicrobial effect. The isolated putrefactive fungus was identified as Botrytis cinerea in ginseng seedling. Therefore, mycelial growth inhibition of Botrytis cinerea on ginseng seedling was measured on PDA plates to test the inhibitory effect of gaseous ClO2. In the case of a low concentration of ClO2 0.3-10 treated group, it showed the inhibition rate of 4.55%. Each treatment group of ClO2 0.8-10, ClO2 1.1-7, ClO2 1.4-5, and ClO2 1.6-3 was 60.72, 49.33, 54.36, and 52.89%, respectively, of mycelial growth inhibition. Therefore, the results of this study showed that gaseous ClO2 treatment would be a sufficient potential fungicide as to ensure the safety and quality for long-term storage of ginseng seedling.
