In the present study, anti-inflammatory activity and inhibitory effect on neurotoxicity of ginsenoside Re (G-Re) was investigated. Microglial cells were activated by LPS, damaged HT-22 hippocampus cells, whereas pre-treatment with G-Re inhibited neuronal cell death. G-Re inhibited LPS-induced pro-inflammatory mediators such as iNOS, COX-2, nitric oxide (NO), reactive oxygen species (ROS), interleukin-1β (IL-1β), interleukin-6 (IL-6), and tumor necrosis factor alpha (TNF-α) in BV-2 microglial cells. Moreover, G-Re also inhibited nuclear translocation of NF-κB and c-Jun, degradation and phosphorylation of I-κB, and phosphorylation of extracellular signal-regulated kinases (ERK) and c-Jun N-terminal kinases (JNK). These results indicate that G-Re could suppress neurotoxicity by inhibiting pro-inflammatory mediators in LPS-stimulated microglial cells.
