본 연구는 Ginsenoside Rg2, Rg3, Rg5, Rg6, Rh1 그리고 F4 등의 인삼의 활성 prosapogenin 성분을 고농도로 함유하는 조성물을 개발하여 건강기능식품 및 화장품의 핵심 원료 소재 획득을 목표로 하였다. 농업 부산물로 폐기물인 인삼열매 및 인삼화뢰에 물리화학적 가공 방법의 하나로서 증류수 추출(100℃)시간, 용매별 추출, 초음파 처리(100℃)를 함으로써 인삼사포닌배당체를 인삼 prosapogenin으로 전환하였고, 이의 성분변화를 HPLC법으로 검토하였다. 인삼화뢰의 물추출 시간에 따른 기능성분 고농도 함유 최적조건을 탐색한 결과, ginsenoside Rg3의 경우는 20시간 추출 시 가장 높은 함유량을 나타내었고, ginsenoside Rd, Rg2, Rh1의 경우는 16시간 추출시 가장 높은 함유량을 나타내었으며, 인삼화뢰의 주 사포닌 배당체 성분인 ginsenoside Re의 함량은 2시간 추출시 가장 높은 함량을 나타내었다. 인삼화뢰의 추출용매에 따른 기능성분 고농도 함유 최적조건을 탐색한 결과, 홍삼 특유사포닌으로 활성사포닌인 ginsenoside Rh1, Rg3 고농도 함유 조성물은 증류수 24시간 추출물이 좋고, 인삼사포닌 배당체인 ginsenoside Rb2, Rc, Rd 고농도 함유 조성물은 70% ethylalcohol 24시간 추출물이 가장 높은 함량을 나타내었다. 인삼화뢰의 초음파처리를 통한 기능성분 고농도 함유 최적조건을 탐색한 결과, 100℃에서 16시간 초음파 처리시 ginsenoside F4가 8.833%, Rg2 1.002%, Rg3 2.230%, 그리고 Rg5가 2.339%로 고농도 함유량를 보여주었다. 또한, 인삼열매의 초음파처리(100℃)를 통한 기능성분 고농도 함유 최적조건을 탐색한 결과, 10시간 초음파 처리시 ginsenoside Rh1이 15.872%, 6시간 초음파 처리시 ginsenoside Rg3가 13.632%로 고농도 함유를 보여주었다. 한편, 초음파처리 인삼화뢰 조성물의 피부 주름에 미치는 영향을 검토한 결과, 초음파처리 인삼화뢰 조성물 이소프로필알코올 분획(GFB-IF)은 Hs68세포에 대하여 0.05%이하의 농도에서 세포독성이 관찰되지 않았으며, 자외선을 처리하지 않은 상태에서 GFB-IF는 낮은 농도(0.000008 ~ 0.0002 %)에서 사람의 섬유아세포 유래의 Hs68세포에 있어서 콜라겐 합성을 촉진하는 효과를 나타내었고, 콜라겐을 분해하는 human MMP-1의 양을 0.025, 0.05%의 농도에서 유의하게 감소시켰다. 이와 같은 결과는 GFB-IF의 주름개선 효과에 일부분 기여하는 결과라고 사료된다. 이와 같은 결과를 검토해 보면, 인삼화뢰 및 열매를 이용한 초음파 처리 prosapogenin 고농도 함유 제제는 기능 식품 및 화장품의 기능성 강화 소재로서 활용이 가능할 것으로 사료되며, 향후, 본 제제를 이용한 새로운 기능성 제품의 개발이 기대되고 있다.

This study aims to develop extracts containing high-concentrations of ginseng's activating components of prosapogenin such as Ginsenoside Rg2, Rg3, Rg5, Rg6, Rh1 and F4, and obtain raw materials essential to health functional foods and functional cosmetics. To this end, the study utilized an HPLC method in which ginseng saponin glycosides were converted into ginseng prosapogenin by means of ultrasonication by distilled water(100℃) time, Solvent extraction, Ultrasonic treatment (100℃) to ginseng berries and flower buds, which are agricultural by-products and wastes, by means of a physicochemical processing method. An examination of the optimum condition in which to contain high concentrations of functional components through water extraction of ginseng flower buds indicates that ginsenoside Rg3 reached a high of concentrations when extracted at 24-hours, ginsenoside Rd, Rg2, Rh1 did at 16-hours and the content of ginsenoside Re, the main saponin glycoside ingredient, reached a high when extracted at 2-hours. An examination of the optimum condition in which to contain high concentrations of functional components through extraction solvents of ginseng flower buds shows that compositions having high concentrations of functional components of ginsenoside Rh1, Rg3, activating saponin unique to red ginseng, reached a high when extracted at 24-hours and extracted in 70% ethylalcohol. An examination of the optimum condition in which to contain high concentrations of functional components through ultrasonification shows that ginsenoside F4 pointed to 8.833% when ultrasonificated for 16 hours at 100℃, Rg2 to 1.002%, Rg3 to 2.230%, and Rg5 to 2.339%, respectively, all showing contents of high concentrations. At the same time, an examination of optimum conditions in which to contain functional components of high concentrations through ultrasonification(100℃) of ginseng berries indicates that ginsenoside Rh1 reached 15.872%, when ultrasonificated for 10 hours and ginsenoside Rg3 reached 13.632% when ultrasonificated for 6 hours, all showing contents of high concentrations. On the other hand, an examination of effects of ginseng flower compositions that have been ultrasonificated on skin wrinkles indicates that no cytotoxicity was detected in isopropyl alcohol extract (GFB-IF), an ultrasonificated ginseng flower composition for Hs68 cells, at concentrations 0.05% or lower, and GFB-IF is low concentrations (0.000008 ~ 0.0002%) without ultrasonification took effects in the form of facilitating collagen synthesis for Hs68 derived from a human fibroblast, and significantly lowered the amount of human MMP-1 that disintegrates collagen at concentrations 0.025% and 0.05%. Such results are understood as GFB-IF being contributive partially to improvement of facial wrinkles. A review of such results suggests that preparations of ultrasonificated prosapogenin of high concentrations that have used ginseng flower buds and berries could be utilized as a function strengthening material for functional foods and cosmetics. As such, new functional products using this preparation are expected to follow in the days to come.
