Sephadex LH-20과 MCI gel CHP-20을 resin으로 사용하였으며 open column chromatography system을 통해 단일 compound를 분리하였다. 1H-NMR과 13C-NMR spectrum 및 FAB/MS를 이용하여 구조 분석한 결과 active compound는 ferulic acid로 구조가 결정되었다. 번행초로부터 분리한 단일 compound인 ferulic acid의 항염증 효과를 확인하기 위하여 hyaluronidase 저해 효과를 확인한 결과, 50, 100, 150, 200 μg/mL의 농도에서 55~97%의 높은 저해 효과를 나타내었다. Ferulic acid의 세포 독성을 확인하기 위하여 Raw 264.7 세포에 ferulic acid를 농도별 (5, 10, 25, 50, 100 μg/mL)로 처리한 결과 50 μg/mL 처리군에서 80%, 100 μg/mL 처리군에서 59%의 cell viability를 나타내어 NO 측정 및 western blot, cytokine은 90% 이상의 cell viability를 나타내는 25 μg/mL 이하의 처리군에서 실험을 진행하였다. 염증반응의 지표 물질인 nitric oxide (NO) 생성 저해 효과를 확인하기 위하여 ferulic acid를 5, 10, 25 μg/mL 농도별로 처리한 결과, 농도 의존적으로 유의서 있게 NO 생성이 감소하였으며 25 μg/mL의 ferulic acid 처리군에서는 30% 이상의 NO 생성 저해 활성을 나타내었다. NO 생성에 관여하는 iNOS 단백질 발현 억제 효과를 확인한 결과, 25 μg/mL 처리군에서 70% 이상의 생성 억제 효과 나타내었으므로 ferulic acid는 iNOS 단백질 발현을 효과적으로 억제하여 NO 생성을 감소시키는 것으로 판단되었다. COX-2 단백질 발현 억제효과 확인한 결과, 25 μg/mL 처리군에서 70% 이상의 생성 억제 효과를 나타내었으며, COX-2 단백질에 의해 생합성되는 PGE2 생성 역시 60% 이상 억제하였다. 따라서 ferulic acid는 COX-2 단백질을 효과적으로 억제하여 PGE2 생성을 감소시키는 것으로 판단되었다. Ferulic acid의 pro-inflammatory cytokine 생성 억제효과를 확인하기 위하여 TNF-α, IL-1β, IL-6의 생성을 측정한 결과, ferulic acid 25 μg/mL 처리군에서 각각 40%, 75%, 77%의 생성 저해 효과를 나타내어 ferulic acid가 PGE2, TNF-α, IL-1β, IL-6 생성을 효과적으로 억제하는 것을 확인하였다. 이상의 결과로 LPS로 염증 반응이 유도되어진 Raw 264.7 세포에 대한 번행초로부터 분리한 ferulic acid는 염증성 질환의 분자생물학적 기전과 관련이 되어 있으며, NO, PGE2 생성 억제, iNOS, COX-2 단백질 발현 억제, pro-inflammatory cytokine 생성 억제를 통해 ferulic acid가 염증 반응을 효과적으로 저해하는 것으로 판단되었다. 따라서 번행초로부터 분리한 ferulic acid는 염증 질환을 완화시킬 수 있는 새로운 항염증 소재연구의 기초 자료로써 활용 될 것으로 예상되며, 추후 산업적 응용을 위한 지속적인 연구가 진행되어야 할 것으로 판단되었다.

Tetragonia tetragonoides is widely cultivated throughout the world for use as a vegetable and medicinal plant effect of gastritis. In this study, we determined the anti-inflammatory effect of purified ferulic acid from Tetragonia tetragonoides in lipopolysaccharide (LPS) simulated Raw 264.7cells. An anti-inflammatory substance from Tetragonia tetragonoides was extracted with water and fracted with H2O/ethyl acetate/n-butanol. Phenolic content and hyaluronidase inhibition effect of H2O fraction appeared to the highest among the fraction. Thus, H2O fraction was set for target purification. The single compound was purified through the open column chromatography system with sephadex LH-20 and MCI gel CHP-20. The results of 1H-NMR, 13C-NMR spectrum and FAB/MS, active compound structure were confirmed as ferulic acid. Hyaluronidase inhibition activity related to the anti-inflammatory effect was 97% with the treated of ferulic acid 200 μg/mL. Anti-inflammatory effects in terms of nitric oxide (NO), inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), prostaglandin E2 (PGE2) and Tumor necrosis factor-α (TNF-α), Interleukin-1β (IL-1β), Interleukin-6 (IL-6) production were measured using LPS-treated Raw 264.7 cells. Cell cytoxicity was identified by treating purified ferulic acid in Raw 264.7 cell at different concentrations (5, 10, 25, 50, 100 µg/mL). As a result, ferulic acid at concentration ranging from 5 ~ 25 µg/mL did have above 95% cell viability. Therefore, ferulic acid were used concentration of 5, 10, 25 µg/mL in all subsequent experiment. To determine the inhibition of NO production in Raw 264.7 cells, ferulic acid was treated 5, 10 and 25 µg/mL concentrations. The inhibitory activity of ferulic acid was determined 30% at 25 µg/mL, decreased concentration dependantly. iNOS protein expression inhibitory effect related to NO production was determined 70% at 25 µg/mL. Therefore, we confirmed ferulic acid reduced NO production by inhibiting iNOS protein expression. The COX-2 protein expression and PGE2 production were decreased 70% and 66% at 25 µg/mL. Thus, we confirmed ferulic acid reduced PGE2 production by inhibiting COX-2 protein expression. The pro-inflammatory cytokines inhibitory effects, TNF-α, IL-6, IL-1β, were determined each 40%, 75%, 77% at 25 µg/mL. According to the results, ferulic acid was inhibited inflammatory mediators by reducing the inflammatory response. These results show that anti-inflammation activity of ferulic acid was expressed by decreasing the expression and secretion of inflammation inducing substances in Raw 264.7 cell induced by LPS. Therefore, it was found that ferulic acid can be developed as a functional ingredient with anti-inflammatory activity.
